Purification and characterization of distinct type of mannose-sensitive fimbriae from Salmonella typhimurium.

The fimbriae (E4) of a virulent strain of Salmonella typhimurium were purified by ion exchange chromatography in an FPLC system. They had a channelled appearance under transmission electron microscope and showed a major structural subunit of 17-kDa on sodium dodecyl sulphate-polyacrylamide gel electrophoresis. The purified fimbriae were found to agglutinate guinea pig erythrocytes, but this effect was inhibited in presence of D-mannose. Immune sera raised against the Mono-Q purified fimbriae (E4) showed cross-reactivity with the type-1 fimbriae (F1) composed of 21-kDa fimbrin subunit, purified by a different method from the same strain.

Magnesium status and parenteral magnesium sulphate therapy in acute aluminum phosphide intoxication.

The results of an open randomized study on magnesium status and parenteral magnesium sulphate therapy in acute aluminium phosphide intoxication are presented. The study was conducted on 105 patients divided into two group (I & II). Patients of Group I did not receive parenteral magnesium and acted as blank. Magnesium levels were monitored every 6 h for 24 h. Patients of group II received magnesium sulphate therapy. It was administered as 1.0 g (8.1 mEq or 4.05 mmol) magnesium sulphate dissolved in 100 ml of 5 per cent dextrose intravenously as a bolus dose followed by 1.0 g every hour for three successive hours, then 1.0 g every 6 h as a maintenance dose for the next 24 h as intravenous infusion in 5 per cent dextrose. The total dose of magnesium sulphate infused was 30.0 mmol over a period of 24 h (initial dose), then 16.0 mmol (4.0 g) daily till final outcome or a maximum of five days. All the vital parameters were monitored. All the patients were followed till final outcome. The resuscitation methods used were the same in both groups. At the end of the study, mortality rates were calculated in both groups. Hypomagnesaemia was observed as the constant finding in patients of Group I. It was transient and reversed itself without MgSO4. The mortality rate was 52 per cent. On the other hand, magnesium levels rose immediately after parenteral MgSO4 administration in patients of group II and they remained persistently above normal during the observed period.(ABSTRACT TRUNCATED AT 250 WORDS)

Chlamydial cervicitis: role of culture, enzyme immunoassay and Giemsa cytology in diagnosis.

Three different laboratory tests were carried out to find the occurrence of chlamydial infections in a selected group of 150 female patients presenting clinically with cervicitis. The tests included isolation using a cell culture system, enzyme immunoassay (EIA) for antigen detection and Giemsa cytology of endocervical smears. Contrary to earlier reports on cytological diagnosis of chlamydial cervicitis, endocervical smears stained by Giemsa stain and EIA for antigen detection were found to be of comparable sensitivity and specificity (73.1% and 86.3% for smear, and 76.1% and 90.9% for EIA, respectively) when cell culture was used as the gold standard.

Pattern of interleukins in minimal-change nephrotic syndrome of childhood.

Assays of interleukin-1 (IL-1) and IL-2 were done in supernatants from phytohaemagglutinin-activated lymphocyte cultures from 10 children suffering from minimal-change nephrotic syndrome (MCNS) to assess their role in the aetiopathogenesis of this disorder. Increased levels of IL-1 and IL-2 had been found in supernatants from patients having MCNS compared with controls, suggesting a significant role of these cytokines in the immunopathogenesis of proteinuria in this syndrome.

Monoclonal antibody for the diagnosis of lymphogranuloma venereum: a preliminary report.

Diagnosis of lymphogranuloma venereum is impeded by difficulty in culturing the causative agent and by serological cross-reactivity between the causative serotypes L1, L2, L3 and other serotypes of Chlamydia trachomatis. Monoclonal antibodies were raised against L1 and L3 serotypes. Smears of 25 patients were tested with these type-specific antibodies by indirect immunofluorescence, of which 17 were found to be positive for L1 monoclonal antibody (Clone D12.1) and four were positive for L3 serotype (Clone B3.6.3). Smears prepared simultaneously and stained with Giemsa were negative in all these cases. This monoclonal antibody may prove to be useful in the diagnosis of lymphogranuloma venereum.

Screening for genital chlamydial infection in symptomatic women.

During a two year period, the prevalence of genital chlamydial infection was assessed by Papanicolaou and Giemsa cytology of endocervical specimens from 396 women attending a gynaecology outpatient department in a Delhi hospital, with symptoms of lower genital tract infection and primary or secondary infertility. The results of cytological examination were confirmed by isolation in a cell culture system. Prevalence was found to be 41 per cent (93 of 227) in patients presenting with vaginal discharge and 36 per cent (61 of 169) in infertility women. Genital chlamydial infection is prevalent in gynaecological patients in this region.

Peroral pneumo colon--a double contrast technique to evaluate distal ileum and proximal colon.

The technique of peroral pneumocolon (POPC) has been found to be simple and easy to obtain excellent double contrast visualization of the distal ileum and proximal colon. Thirty-four patients were subjected to the POPC examination of which 29 had undergone prior barium meal follow through studies. In 86.2% of these cases POPC was found to be superior as compared to conventional studies. In 5 patients who could not retain Ba-enema, POPC was performed as first line of investigation with 100% efficacy.

Uptake of purine and pyrimidine nucleosides by macrophage-resident Mycobacterium leprae: 3H-adenosine as an indicator of viability and antimicrobial activity.

Freshly extracted human- and armadillo-derived Mycobacterium leprae maintained within murine macrophages incorporated significant levels (p less than 0.05 to p less than 0.001) of 3H-adenosine and 3H-hypoxanthine by 6 and 9 days of the culture period. The incorporation of 3H-adenosine was twofold or more higher than 3H-thymidine in 10 out of 15 human-derived M. leprae isolates. Macrophage-adapted bacilli incorporated 10-14-fold higher levels of 3H-adenosine compared to the same bacilli maintained in axenic cultures. The incorporation of these two labels was inhibited by dapsone and rifampin, indicating the utility of in vitro radiometric assays for screening antileprosy drugs and drug sensitivity/resistance in patients.

Thymic lymphoma cells as a model for complex-formation between thymocytes and thymic medullary epithelial cells.

The formation of complexes between thymocytes and thymic stromal elements is known to be involved in T cell differentiation. We have previously described one type of lympho-stromal interaction involving CD4+ CD8+ thymocytes and a medullary epithelial cell line (E-5). In this study we report the potential for complex formation of two different thymic lymphoma cell lines (Ti-6 and RDM-4). Ti-6 cells were shown to adhere to the E-5 cells, while RDM-4 cells were totally incompetent. Phenotypic characterization of these cell lines suggests that immature thymocytes are not capable of forming complexes with medullary epithelium and that a certain level of differentiation is required to do so. Comparison of their phenotypes showed that the possibility of some classical T cell surface markers being the receptor for the E-5 ligand can be dismissed.
