Differential recognition of Dirofilaria immitis antigens by human IgG and IgM positive sera. Preliminary data based on EITB analysis.

Preliminary data are presented on the differential recognition of Dirofilaria immitis antigens by sera positive for IgG and IgM (detected by enzyme linked immunosorbent assay, ELISA) using the enzyme-linked immunoelectrotransfer technique (EITB). The sera were followed prospectively over a year to detect seasonal variations. IgG mainly recognized antigens with a molecular weight (MW) lower than 74 kDa, while IgM recognized those with a MW higher than 43 kDa. The antigens with MW between 74 and 61 kDa were recognized by both isotypes. In IgG positive sera all bands were recognized throughout the year, albeit more mildly in summer. For IgM it is noteworthy that antigens with a MW between 43 and 20 kDa were not recognized in spring. These data are in accordance with those obtained with ELISA in the seasonal follow-up of positive sera.

Seasonal changes in the levels of anti-Dirofilaria immitis antibodies in an exposed human population.

A prospective follow-up study to define seasonal changes in the level of anti-Dirofilaria immitis antibodies in an exposed human population in Western Spain was conducted. Specific antibodies were detected using an enzymelinked immunosorbent assay (ELISA). Ig M antibodies predominated in Summer, during maximal vector activity, while Ig G rose to a maximum in Winter. The sole risk factor associated with higher optical densities in the ELISA proved to be age, indicating that repeated exposure to the parasite is the rule. It is concluded that, for serodiagnosis the endemic or non-endemic nature of the geographical area and the season of the year should be considered to define the basal level of positivity, and that as well as Ig G, Ig M should always be determined.

Inverse correlation between fibronectin mRNA levels and the metastatic potential of two murine mammary adenocarcinomas.

The ability of cells to metastasize is thought to be related with the loss of the capacity to synthesize and secrete fibronectin (FN). We found that primary cultures from 2 murine adenocarcinoma tumors, M3 of moderate metastasizing ability and the highly metastasizing MM3, had a dramatic difference in the immunohistochemical expression of FN. It was shown, as assayed by the S1 mapping technique that, while total FN mRNA was fairly abundant in M3 cells, it was almost undetectable in MM3 cells. This difference is not due to a significant deletion of the FN gene in MM3 cells.

A seroepidemiologic survey of human dirofilariosis in Western Spain.

The objective of this work is to define the prevalence of IgM and IgG antibodies against Dirofilaria immitis in an exposed human population. 395 individuals attending primary care facilities in an area endemic with canine dirofilariasis were studied, as well as 100 non exposed individuals. An ELISA method was used for the detection of IgG and IgM antidirofilarial antibodies. Sera were preadsorbed with antigens of Toxocara canis, Ascaris suum, Echinoccocus granulosus and Fasciola hepatica to prevent cross reactions. Global seroprevalence was 9.3%. IgG antibodies were observed in the oldest sector of the population, while IgM antibodies were found only in its youngest half. No sex differences were observed. This result show that dirofilariasis is common in humans from endemic areas. Repeated contacts with the parasite are common and begin at a young age.

Neurospora crassa cDNA clones coding for a new member of the ras protein family.

A new member of the ras gene family was characterized from Neurospora crassa cDNA libraries. The clone designated NC-ras codes for a polypeptide containing 213 amino acids (Mr 24,000). This polypeptide is 84% homologous to the H-ras-1 domain comprising the first 80 amino acids and 60% homologous to the next 84 residues. The NC-ras polypeptide contains all the well-known sequences involved in the interaction with GTP/GDP, the recognition of the Y13-259 neutralizing antibody, the 'effector site' for interaction with GAP proteins, and the CAAX acylation motif in the COOH-terminal.

Transient solitary pulmonary nodule caused by Dirofilaria immitis.

The second described case of solitary transient lung nodule caused by Dirofilaria immitis is reported. Diagnostic thoracotomy can be avoided with the use of enzyme-linked immunosorbent assay (ELISA), and hence a conservative serological follow-up is warranted in endemic areas.

[Serology of human dirofilariasis].

The development of an immunoenzymatic technique for the detection of specific IgG against Dirofilaria immitis in humans is reported. Two groups of rabbits were used as controls; one of them was hyperimmunized with D. immitis antigens and the other was not inoculated. Subsequently, the reactivity to D. immitis and T. canis of the sera of three patients with pulmonary nodules and 25 apparently healthy individuals was evaluated. In two of the patients with pulmonary nodules the reactivity against D. immitis was significantly higher than that of healthy individuals. Reactivity against T. canis was not found in any case. These initial results seem to indicate that, in the reported conditions, this technique could prevent crossed reactions between D. immitis and T. canis and could help in the differential diagnosis of other causes of pulmonary nodules.

Ultrasound diagnosis of ruptured hydatid cyst of the liver with biliary obstruction.

Communication between a hydatid cyst of the liver and biliary tract results in a clinical picture of obstructive jaundice because of occupation of the extrahepatic biliary tract by intracystic material. Five cases of this complication are presented. Ultrasound criteria for diagnosis are analyzed, based on previous publications as well as our own experience.

Bile cytology. A routine addition to percutaneous biliary drainage.

Exfoliative cytology of bile aspirated from percutaneously placed combined external/internal biliary drainage catheters yielded positive results in 34% of 100 cases. There were no false-positive results. Bile cytology is a simple, inexpensive, and useful method which can establish a diagnosis of malignant biliary obstruction after catheter decompression.
