[Interaction of sex steroids with proteins from the soluble fraction of swine and cattle liver (a preliminary analysis)].

The interactions of [3H]estradiol, [3H]testosterone and [3H]progesterone with soluble proteins from porcine and calf liver were studied. The specific binding of [3H]progesterone and [3H]testosterone in calf liver cytosol seems to be due to serum transcortin or its intracellular precursor (analog). Contrariwise, the specific binding of [3H]progesterone observed in porcine liver cytosol was absent in the serum. This binding was characterized by slow association and dissociation dynamics, moderate affinity for the [3H]-ligand and a high binding capacity. The structural determinants of the ligands were studied by competitive inhibition of the [3H]-ligand binding. The delta 4-3-keto group in the steroid A-ring was found to be the most important determinant. An intensive metabolism of [3H]progesterone was observed during its incubation with cytosol (data from thin-layer chromatography). A 3H-metabolite (presumably, 20 beta-dihydroprogesterone) was predominant in the bound ligand fraction. The data obtained suggest that proteins of a steromodulin type are widely distributed in the mammalian liver.

[The cellular localization of the unusual estrogen-binding protein in the liver of rats under different hormonal states].

The localization of an unusual estrogen-binding protein (UEBP) was studied in the rat liver using indirect immunoperoxidase reaction in intact rats and after different hormonal influences. The distribution of the UEBR in normal males displays a form of a gradient with the maximum near the central veins. The gradient is absent in normal females. Castration or hypophysectomy of males, or their injection with estradiol or triiodyronine leads to a decrease in the UEBP concentration, though the gradient character of its distribution is persisting. There is a stable increase in the UEBP concentration in the first two layers of cells adjacent to the central veins in the female rat liver after ovariectomy and especially in combination with androgen injections. The revealed changes in the intensity of immunoperoxidase reaction after different hormonal influences correlate with the UEBP concentration in liver studies performed by the radioligand assay.

[Cysts of the common bile duct in children].

From 1975 to 1989 operations were carried out in the clinic by various methods on 30 children with cysts of the common bile duct. Hepaticoor choledochojejunostomy after Roux is the operation of choice in cysts of the choledochus. However, recurrent cholangitis caused by jejunobiliary reflux was encountered in some patients in the postoperative period. The accumulated clinical experience has also shown that the Roux type operation does not guarantee a successful functional result. To prevent reflux of the intestinal contents into the biliary tract after Roux's anastomosis, the authors used an antireflux valve on the excluded loop of the jejunum. Experimental study was undertaken of the theoretically substantiated antireflux valve on an excluded loop of the type of a "nonspilling inkpot" with a shortened loop. The creation of an antireflux valve in the form of an invaginated segment prevents jejunobiliary reflux and therefore provides the possibility for reducing significantly the length of the excluded loop in Roux's biliodigestive anastomoses. The obtained positive results confirm the rationality of this construction which does not need additional time for the operation. Since 1987 four children have been operated on by this method in the clinic. The length of the excluded loop was reduced from 30 to 15 cm. Postoperative complications linked with reflux into the biliary system were not encountered.

[17 Beta-estradiol interaction with haptoglobin and its complex with hemoglobin].

The estradiol-binding capacity of sheep haptoglobin and its complex with hemoglobin was investigated. It was shown that in the presence of H2O2 the haptoglobin-hemoglobin complex tightly binds 17 beta-estradiol. No dissociation of the estradiol-protein complex occurs after its precipitation with trichloroacetic acid and steroid extraction with the ester. It is supposed that the 17 beta-estradiol is bound to the protein by covalent bonds; this binding is characteristic only of the haptoglobin-hemoglobin complex but not of haptoglobin alone.
