Comparison of two methods of labeling proteins with 111In.

DTPA N-hydroxysuccinimide pentaester (DHSE) can be used as an alternative to bicyclic anhydride for coupling proteins with DTPA. However, in-vitro analysis of labeled proteins coupled with DHSE compared to bicyclic anhydride showed more high molecular weight product and less serum stability. We studied the magnitude of these differences in vivo by coupling HSA and IgG with DTPA using both methods, labeling with 111In, and analyzing the biodistribution in mice. The results of this analysis showed comparable activity in the liver, kidneys and blood.

Electrophoretic analysis of different technetium-99m (SnCl2) methylene diphosphonate complexes.

Technetium-99m MDP was prepared from MDP kits from several different sources. The resulting [99mTc]MDP preparations were analyzed by electrophoresis. The results demonstrated the presence of at least four different 99mTc complexes in these preparations. Modified kit preparations were analyzed to show the effects of concentration and pH on formation of the impurities. The electrophoresis results were correlated with scintillation camera imaging studies in rabbits and suggest that hydrolysis of MDP to phosphate and methylphosphate results in formation of 99mTc complexes with poor biological behavior as bone scanning agents.

The preparation of DTPA-coupled antibodies radiolabeled with metallic radionuclides: an improved method.

Isotopes of iodine are often employed as radiolabels for antibodies used in radioimmunodetection studies in which tumor localization is determined by external imaging. Because of drawbacks associated with the use of these isotopes, alternative labeling methods have been considered; such as covalently attaching strong chelators so that the coupled protein may be radiolabeled with metallic radionuclides by chelation. We have developed a method of coupling the strong chelator diethylenetriaminepentaacetic acid (DTPA) which is simple, efficient, and superior to reported methods. Using the cyclic anhydride, coupling to IgG antibody is about 75% efficient and is completed in less than 1 min at neutral pH. Because the concentration of hydrolytic products is small, the coupled protein is rapidly purified for use or storage. Labeling of the protein is also accomplished rapidly and the labeled product has been shown to be stable both in vitro and in vivo.

Lymphoscintigraphy with Tc-99m-labeled dextran.

Current agents for lymphoscintigraphy have limitations because of slow migration of the colloidal tracers from the injection site and the unknown effect of phagocytosis on the removal of the labeled particles. The usefulness of Tc-99m dextran (TcDx) with a molecular weight of 110,000 has been tested for lymphoscintigraphy. Computer-assisted dynamic imaging and serial blood sampling in 13 dog experiments demonstrated that the tracer cleared only by lymph drainage from an interstitial injection site. Following interdigital injection of 1.0 ml (0.5-5.0 mCi), TcDx reached the knee or elbow lymph nodes in 12.4 +/- 6.5 (1 s.d.) sec, and the inguinal or axillary lymph nodes in 98.0 +/- 42.3 sec. It cleared from the injection site with a half-time of 31.5 min. In a dog with surgically induced lymphedema, tracer migration was markedly delayed in the edematous leg and the radionuclide lymphoscintigram resembled the contrast lymphangiogram. Initial studies in man yielded high-quality radionuclide lymphograms of the leg, and the pelvic and paraaortic lymph nodes. We conclude that TcDx is very promising for lymphoscintigraphy.

Remote, semiautomated production of F-18-labeled 2-deoxy-2-fluoro-D-glucose.

A routine production system for 2-deoxy-2-[18F]fluoro-D-glucose (2-18FDG) for medical use is presented. The method involves the preparation of F-18-labeled F2 by the 20Ne(d, alpha) 18F reaction, subsequent fluorination of 3,4,6-tri-o-acetyl-D-glucal, purification, and hydrolysis of the appropriate diastereoisomer to obtain 2-18FDG. These operations are performed using a completely remote, semiautomatic system. There have been no failures in more than 220 consecutive production runs. Currently, two preparations per day of 15-25 mCi of 2-18FDG are routinely performed, with a radiochemical purity, measured by TLC, in excess of 95% and with less than 1 mR radiation exposure to the chemist. Reaction conditions, operational variables, and systems are described in detail.

Cerebral blood volume in a rat model of ischemia by MR imaging at 4.7 T.

Perturbation of the cerebral circulation by occlusion of the vertebral arteries and a carotid artery can be visualized by using MR imaging and the intravascular contrast agent Gd-DTPA complexed to albumin. This tracer consistently reduced the T1 relaxation time in the brain and blood. The difference between hemispheres was revealed by less T1 reduction in the occluded hemisphere and by an adjustment in the display contrast of images that revealed the territory of decreased perfusion. These results were confirmed by comparing them with cerebral blood flow using radioactive microspheres and the intravascular blood volume tracer 51Cr-EDTA. This method, combined with high-resolution MR imaging, can be applied to serial noninvasive studies of cerebral blood volume in ischemia and other conditions.
