Effects of cyclic AMP on S-100 protein level in C-6 glioma cells.

Dibutyryl cyclic AMP (dbcAMP) markedly elevated S-100 protein level in C-6 glioma cells in vitro. Quantitative analysis by two-dimensional polyacrylamide gel electrophoresis revealed that the elevation was caused by a combination of increased synthesis and reduced degradation of S-100 protein in C-6 cells exposed to dbcAMP. These results suggest that dbcAMP affects both the synthesis and the degradation of S-100 protein in C-6 cells.

Serial observations of brain stem function by auditory brain stem responses in central transtentorial herniation.

Changes in auditory brain stem responses were serially investigated in a patient with downward transtentorial herniation due to acute obstructive hydrocephalus to correlate the neurological signs with the results of computerized tomography. Neurological deterioration correlated highly with disruption of auditory brain stem responses, especially with that of the components of waves V to VII. A noninvasive technique, the measurement of auditory brain stem responses is thought to be useful in detecting the severity of downward transtentorial herniation and in estimating the recovery of brain stem function after surgical treatment of intracranial hypertension.

Immunohistochemical demonstration of IgG in meningioma.

Twenty human meningiomas were examined for IgG and IgM by the direct immunofluorescence of immunoperoxidase methods, or both. IgG was conspicuously found in and around the blood vessels, whorls, and psammoma bodies. It was also clearly present on the cytoplasmic membranes of the tumour cells. On the other hand, IgM was seen only within the blood vessels. Significance of these findings is briefly discussed including possible humoral immune reactions in regard to whorl and psammoma body formation in meningioma.

Evaluation of the drug-induced morphological differentiation of rat glioma cells (C-6) from the aspects of S-100 protein level and con A binding pattern.

The intracellular content of the nervous system specific protein S-100 began to increase with 4 days latency following the morphological differentiation of cultured rat glioma cells (C-6) with 1 mM dibutyryl cyclic AMP (dbcAMP), rising to approximately 10-fold over the control level at 15 days after the treatment. The concanavalin A (Con A) binding pattern on the external cell surface of C-6 cells exposed to dbcAMP appeared as a smooth layer of 40-60 nm thickness whereas that of control cells was irregularly thick and patchy. The correlation between morphological and biochemical changes of C-6 cells after dbcAMP treatment is discussed in relation to the mechanism controlling the differentiation of glioma cells.

[An anatomical and technical note for microsurgery of the MCA aneurysm (author's transl)].

Several basic anatomical and technical principles helpful in selecting and planning the surgical treatment of the aneurysm of middle cerebral artery were discussed. 1. Superficial sylvian vein (SSV) usually runs on the temporal opercular bank, therefore, frontal opercular side may be preferred in opening the sylvian fissure(SF). In approximately 10% of specimens, there are two separate SSV lying close together in the SF which only rarely have communicating channels to each other. It should be kept in mind a vein of the one opercular bank may course to the surface, pass beneath the nearest sylvian vein on its side of the fissure and join the more distally located sylvian vein. 2. Deep sylvian vein (deep middle cerebral vein) which closely accompanies the trunk of MCA must be preserved. Surgical occlusion of this venous pathway may result in unanticipated serious postoperative complication, such as consciousness deterioration, hemiparesis or aphasia. 3. Perforators leaving the M1 over its posterosuperior surface form a loop which hang anteriorly. The loop occasionally comes forward and extends quite anteriorly where the sylvian fissure opens and therefore is in danger of injury. It is important to avoid tearing these fine vessels while exposing the middle cerebral artery. 4. The parent artery should be exposed proximally to the aneurysm to allow control of flow to the aneurysm if it ruptures during dissection in any way of approaching the aneurysm. 5. The aneurysm usually points in the direction of the long axis of the prebifurcation segment. 6. After the clip is applied, the area should be inspected to make certain the clip does not kink or obstruct the major vessels and that no perforating branches are included in it.

Chromosome analysis of a brain malignant lymphoma cell line.

Chromosome studies of a malignant lymphoma cell line derived from the brain were made by Q- and G-banding techniques. The modal number of chromosomes was 45. Complex structural rearrangements were present, but the 14q+ marker chromosome frequently seen in malignant lymphomas was not identified in the cell line. The main karyotype in cells analyzed was 45, X, -Y, del (2) (q21q23), t (3;?) (p25;?), t (p12;?), -8, 11q+, 18q+, +mar. Absence of the 14q+ may be explained by: firstly, clones which possessed 14q+ marker chromosome in brain tumor cells may have been selected out with increasing culture time and repeated passages; or secondly, the presence of the 14q+ marker chromosome depends on the type of lymphoma.

Primary intracranial choriocarcinoma.

The authors report four cases of primary intracranial choriocarcinoma, all in young males. There tumors occurred in the pineal region and one in the lateral ventricle. Besides the signs of increased intracranial pressure and ophthalmological disorders, skin pigmentation or eruption was observed in three cases and precocious puberty in two cases. Angiography revealed tumor stain with irregular vessels in all cases. Enhanced computerized tomography scans in the last two cases demonstrated a round, lobulated lesion of high density, with relatively low density in the central portion. Hormonal study was carried out in three cases. In addition to high levels of human chorionic gonadotropin (HCG) in the urine and cerebrospinal fluid and/or of plasma luteinizing hormone (LH), LH in tumor tissue or medium obtained via cell cultures was also high in all three cases, suggesting inherent activity of HCG secretion by the tumor cells. The tumors were relatively demarcated from the surrounding brain. They were very hemorrhagic, and fresh hematoma was identified in the tumor in case. Good results were obtained in two cases treated with surgical removal followed by simultaneous chemotherapy and 60Co irradiation.

Surgical removal of hemangioblastoma in the fourth ventricle.

Three cases of successful removal of hemangioblastoma in the fourth ventricle are reported. Two patients are doing well four years and six months, respectively, after operation. One patient succumbed 15 weeks after operation, most likely due to cardiac arrest. Microsurgical coagulation and dissection of individual arterial feeders permitted dissection of the tumor. When the tumor was elevated, hemorrhage followed. Control of hemorrhaging was associated with postoperative neurological dysfunctions in each case.

[Effect of various kinds of glucocorticoids on experimental brain edema (author's transl)].

Although the effectiveness of glucocorticoids for the treatment of cerebral edema has been widely accepted in neurosurgical practice, but its mechanism is still obscure. Also the dose equivalent of various kinds of glucocorticoid preparation is still uncertain. Only the dose equivalent determined by the test examining an antiinflammatory effect on a growth of glanulation, has been applied for the treatment of brain edema. The authors attempted to establish the dose equivalent of each kinds of glucocorticoid for the treatment of cerebral edema. Cold induced cerebral edema was produced by touching a metal probe at the temperature of -90 degrees C for 3 minutes on the right parietal cortex of rat brain. Hydrocortisone (16 mg/kg), prednisolone (3.2 mg/kg), methylprednisolone (3.2 mg/kg), dexamethasone (0.8 mg/kg) and betamethasone (0.8 mg/kg) were administered at the time of 3 hours prior or after the cold injury. The animals were sacrificed 24 hours after injury and the grade of brain edema was determined by the changes of water content in the brain tissue. For this purpose, the authors developed a new method of measuring water content in the brain tissue using tritiated water and a liquid scintillation counter. In the group of animals which were treated with prednisolone, dexamethasone or betamethasone, the water content was significantly decreased compared to the value of non-treated animals. But in the animals treated with hydrocortisone or methylprednisolone, no significant changes of the water content was observed. But in the animals treated with double doses of hydrocortisone (32 mg/kg) or methylprednisolone (6.4 mg/kg), cerebral edema was improved significantly. From these results the authors concluded that the dose equivalent of each kinds of glucocorticoid for the treatment of the brain edema was as follows: hydrocortisone: prednisolone: methylprednisolone: dexamethasone: betamethasone = 40 : 4 : 8 : 1 : 1.
