[Cell-cooperation integrated test for the detection of immune reactivity to Shiga-toxin in patients with acute intestine infection].

Described are the results of approbation of the cell-migration screening test reflecting the outcome of cooperation between T- and B-cells and macrophages. It was shown, in adults and children with intestine infection, to be a highly-effective tool for the detection of suppressed immune response during early disease stages at stimulation in vitro by Shiga toxin at nano- and picogram concentration; it can also be used for the evaluation of the shaping specific anti-Shiga-toxic immune response. The parameters of the migration activity of peripheral-blood leucocytes at exacerbation and convalescence were demonstrated to correlate with the age of sick children and with the severity of intestine infection as well as with the level of a Shiga-like toxin detected in coprofilters and circulating immune complexes.

[Immunologic mechanisms of the development of nonspecific aortoarteritis].

It has been shown that patients with nonspecific aortoarteritis undergoing acute and subacute stages of the process demonstrate sensitization to collagen, types I and III. The activity of inflammation was discovered to affect the disease progression and involvement of new arterial areas. Marked suppression of leukocyte migration in response to collagen, types I and III, in patients with nonspecific aortoarteritis points to a high probability of further disease progression.

[The viroimmunotest for mass determinations of nanogram amounts of antigens].

Simple highly sensitive screening variant of viral immunoassay for detection of nanogram quantity of antigen is proposed. The antigen linked with antibodies adsorbed on polystyrene 96-well plates was revealed by conjugate of Fab' fragments of antibodies with phage phi X174. The concentration of the antigen could be detected qualitatively and quantitatively (from 0.3 ng/ml to 1000 ng/ml).

[Determination of lymphokine-induced cytolytic activity of macrophages by measuring the 3H-thymidine residue in prelabeled tumor cells].

A technique of macrophage-activating factors (MAF) detection by the in vitro determination of macrophage (Mph) antitumour cytolytic activity by 3H-thymidine residue in the pre-labelled neoplastic target cells (TC) is suggested. Peptone-induced Mph were cultivated for 20-22 hours in the presence of crude supernatants from concanavalin-A-stimulated spleen cells or from the secondary mixed lymphocyte culture. 3H-thymidine-labelled cells of mastocytoma P815 were then added to the washed Mph. The lysis was measured in 48 hours according to the isotope remainder in the non-acid-soluble fraction. The optimal conditions for MAF detection have been selected. Parallel MAF testing with this particular method and with a standard technique using labelled 51Cr TC made it possible to conclude that the method suggested is more sensitive because it permits a combined cultivation of Mph and TC for a relatively long time period.

[Ultradian fluctuations in the natural antitumor resistance of mice undergoing transplantation and chemical carcinogenesis].

A study was made of the mouse life span in relation to the time of tumor cell injection on an hourly basis, while the carcinogenic effect was studied in relation to the time of urethan injection. Changes in both the values were shown to vary with ultradian rhythms. It was found that natural killer cell activity against tumor cells, migration index of normal spleen cells in the presence of tumor cells and the activity of carcinogen-metabolising enzyme system of intact animals also exhibited ultradian oscillations. It is assumed that oscillations of natural resistance against transplantable or chemically-induced tumor may be due to the changes of these parameters.

[Production of monoclonal antibodies to antigens of specific mouse T-suppressors].

F1(MSU X WAG) rats were immunized with anti B6 BALB/c specific suppressor T cells (SSTC), purified by absorption/elution technique, with the following fusion of splenocytes to NS-I myeloma cell line. Hybrids were screened for their ability to affect SSTC, cytotoxic T lymphocytes (CTL) and producers of macrophage migration inhibition factor (MIF-producers) all triggered by in vivo priming with allogeneic cells. Two hybridoma cell lines--C1 and C4 inactivated SSTC by approximately 50%, leaving CTL and MIF-producers intact. C4 were also active in vivo, if injected as ascitic fluid from nu/nu mice, though to a lesser extent than in vitro.

[Production in vivo of macrophage migration inhibition and stimulation factors in the inductive phase of the alloimmune response].

Macrophage migration inhibition and stimulation factors were revealed in the serum of alloimmune mice by polyacrylamide gel electrophoresis. CBA mice were primed intravenously with 90 X 10(6) and challenged subsequently intravenously with 20 X 10(6) spleen cells of BALB/c mice. Macrophage migration inhibition and stimulation factors were revealed in pre- and post-albumin fractions, respectively, already on days 1 and 6 after alloimmunization. It is suggested that at an early phase of immune response the immunomediators with alternative functions whose local activity gradients predetermined the macrophage behaviour, are released into the serum of alloimmune animals.

[Nonidentity of Lyt phenotypes and the radiosensitivity of early and late producers of macrophage migration inhibitory factor in reaction to H-2 complex antigens].

MIF production induced at different times after intravenous immunization of mice with irradiated allogeneic splenic cells showed different sensitivity to the treatment with anti-Lyt-antibodies and to gamma-irradiation. The "early" MIF producers induced several hours after alloimmunization were sensitive to irradiation at a dose of 500 rad and to the treatment with anti-Lyt-1- and anti-Lyt-2-antibodies and complement, while the "late" MIF producers which appeared 21 days after alloimmunization were resistant to irradiation at doses of 500 and 1500 rad and to the treatment with anti-Lyt-2-antibodies but sensitive to anti-Lyt-1-antibodies. It is supposed that the "early" MIF producers of the Lyt-1+2+ phenotype are immature precursors of T cells which, in contradistinction to the "late" MIF producers of the Lyt-1+2+ phenotype, are activated and produce MIF without proliferation after a twofold contact with antigen.

[Induction of T-cell producers of the macrophage migration inhibitory factor by the products of the K-, I- and D-regions of the H-2 complex in mice].

It has been demonstrated with the help of intravenous in-vivo immunization with irradiated splenocytes of A.AL anti-A.TL (difference at the K-region of H-2), A. TH anti-A. TL and A. TL anti-A. TH (difference at the I-region of H-2), C57BL/10Sn anti-B10.D2(R107) and B10.D2 (R107) anti-C57BL/10Sn (difference at the D-region of H-2) mice that T cells--MIF producers, induced at different times after induction of the alloimmune response, react to the products of the K- and I- but not to those of the D-region of the H-2 complex.

The effect of idiotype-specific and anti-ACA-1 sera on T cell subpopulations responding to H-2 antigens.

A xenogeneic anti-CBA-anti-C57BL/6 idiotype-specific serum reacted specifically only with CBA anti-C57BL/6 T cells. It inhibited T killers, T lymphocytes proliferating in one-way MLC and T cells which take part in the rejection of the cardiac allografts. However, it did not affect T suppressors and T MIF producers. Another antiserum, directed against ACA-1 (activated cell antigen), inhibited T killers of various specificities, but not T suppressors or T MIF producers. These results allow us to suggest that (1) the idiotype-like structures of T suppressors and T MIF producers differ from those of other T cell subpopulations responding to H-2 antigens; (2) ACA-1 is expressed on the surface of T killers, but not on T suppressors or T MIF producers; (3) T cells producing MIF form a separate subpopulation.

[Induction of T-cells--producers of macrophage migration inhibitory factors by mutant H-2 antigens in vivo].

C57BL/6 (H-2b) mice were immunized once intravenously by irradiated (1500 rad) spleen cells of mutant strains B6.C-H (z1)Y (H- 2bm1 ) and BB. M505Y (H- 2bm3 ), by the cells of allogeneic strain B10.D2 (H-2d) (in positive control) and syngeneic strain C57BL/6 (in negative control). It was demonstrated that MIF-producing T cells can react to mutant H-2 antigens and that this reaction is revealed on the 1st day after intravenous immunization in vivo. It is suggested that MIF-producing T cells can participate in the immunological surveillance of slight changes in the major histocompatibility complex antigens through mobilization of macrophage effector functions.

[Induction of T cells--producers of a macrophage migration-inhibitory factor by minor histocompatibility antigens].

The reaction of MIF-producing T cells was studied on the 1st, 6th and 13th days after intravenous immunization in vivo by irradiated spleen cells, immunized and immunizing strains of mice differing in: (a) H-2 antigens; (b) H-2 and minor histocompatibility antigens (non-H-2); (c) non-H-2 antigens, Mls identical; (d) non-H-2 antigens, Mls different. It was demonstrated that in combinations of mice differing in H-2 antigens as well as in H-2 and non-H-2 antigens, the reaction of MIF-producing cells was revealed already on the 1st day after immunization and maintained also on the 6th and 13th days. After intravenous immunization against non-H-2 antigens (Mls identical combination of mice) MIF-production was revealed on the 1st day, whereas in combination of mice differing in non-H-2 antigens and Mls locus on the 1st and 6th days after immunization. The data obtained show that in-vivo immunization by the minor transplantation antigens effectively induces MIF-producing T lymphocytes, and first of all, a special category of these cells--"early" MIF-producers.

[Idiotype-specific inactivation by xenogeneic antisera of T killers but not of T-MIF producers immune to H-2 complex antigens].

Following appropriate absorption rabbit serum to CBA anti-C57BL/6 lymph node cells (rabbit ISS) and rat serum to enriched CBA anti-C57BL/6 T killers (rat ISS) selectively inhibited the activity of K-anti-b T killers but did not affect T killers of other specificities (K-anti-d, d-anti-b). Rabbit ISS did not suppress the capacity of CBA anti-C57BL/6 (K-anti-b) T cells to produce MIF. Under the same conditions, anti-Thy-1.2 serum inactivated killers and K-anti-b MIF-producers. The findings indicate that the killers and MIF-producers immune to the antigens of H-2 complex constitute different subpopulations of T cells, which appear to carry nonidentical sets of idiotypes.

[Ultradian biorhythms in mouse bone marrow].

Ultradian oscillations in the number of karyocytes isolated from the femoral bone of intact ACR mice have been demonstrated. The periodicity of oscillations did not depend on the season or the site of mice breeding. The bone marrow also showed ultradian oscillations in relative and absolute amount of lymphoid, myeloid and mitotic cells. It is postulated that differentiation and migration of bone marrow cells might have ultradian biorhythms.

[Production of factors regulating macrophage motility in the early stages of mixed lymphocyte culture].

The macrophage migration stimulation factor (MSF) is produced during the first hours of mixed lymphocyte culture (MLC) in the H-2 system. The activity of MSF is characterized by sharp peaks of migration stimulation in the culture fluids of allogeneic MLC. The peaks appeared every 5 hours. After 12--16 hours in the culture fluids of allogeneic MLC, there has been demonstrated macrophage migration inhibition factor whose activity rose by 48 hours. Optimal concentrations were revealed for stimulation and inhibition of macrophage migrations on the dilution of 1--2-day MLC culture fluids. The existence of the biorhythmical mechanism that controls the macrophage mobility through the balance of soluble mediators with alternative activity is suggested.
