Identification and quantification of Bifidobacterium species isolated from food with genus-specific 16S rRNA-targeted probes by colony hybridization and PCR.

A Bifidobacterium genus-specific target sequence in the V9 variable region of the 16S rRNA has been elaborated and was used to develop a hybridization probe. The specificity of this probe, named lm3 (5'-CGGGTGCTI*CCCACTTTCATG-3'), was used to identify all known type strains and distinguish them from other bacteria. All of the 30 type strains of Bifidobacterium which are available at the German culture collection Deutsche Sammlung von Mikroorganismen und Zellkulturen, 6 commercially available production strains, and 34 closely related relevant strains (as negative controls) were tested. All tested bifidobacteria showed distinct positive signals by colony hybridization, whereas all negative controls showed no distinct dots except Gardnerella vaginalis DSM4944 and Propionibacterium freudenreichii subsp. shermanii DSM4902, which gave slight signals. Furthermore, we established a method for isolation and identification of bifidobacteria from food by using a PCR assay without prior isolation of DNA but breaking the cells with proteinase K. By this method, all Bifidobacterium strains lead to a DNA product of the expected size. We also established a quick assay to quantitatively measure Bifidobacterium counts in food and feces by dilution plating and colony hybridization. We were able to demonstrate that 2.1 x 10(6) to 2.3 x 10(7) colonies/g of sour milk containing bifidobacteria hybridized with the specific nucleotide probe. With these two methods, genus-specific colony hybridization and genus-specific PCR, it is now possible to readily and accurately detect any bifidobacteria in food and fecal samples and to discriminate between them and members of other genera.

[Effects of early biologic and psychosocial risk factors on mental performance and disease-promoting behavior of adult medical patients].

UNLABELLED: Early biological and social risks were related to measures of cognitive functioning and illness behaviour in adult patients. RESULTS: 1. Self-reported data on risks in early developmental stages are consistent with longitudinal data and theoretical assumptions. 2. Patients with biological risks show deficits in fluid abilities and signs of mild cerebral insufficiency. 3. Socially handicapped patients tend to coronary-prone behaviour and report more emotional strain while coping with complex problems.

[Results of health education in preputial hygiene of boys].

The education to clean the prepuce is a part of education in cleanliness of the boys and had international succeeded against the general circumcision of newborns. While 15 school years from 1969/70 to 1983/84 we could point out the driving back of the retention of smegma, especially when heavy. To the farther correction the chief educators of health, the parents, must be motivated to teach their boys this cleaning with perfect technic. Physicians and teachers should reinforce this. Near by we must take in consideration that only parents and physicians could inspect the prepuce of boys. Every visible retention of smegma should provoke the education in washing this region. It was proposed anew to admit the inspection of the prepuce of the penis from the third year of life into the standard programme of mass examination in GDR during infancy and youth. It was referred to the danger of the early traumatism of the foreskin by the newborns and the babies and also to the problems of the late education in the hygiene of the prepuce only while and after the puberty. Also in the further life of all men this region should be inspected at every medical examination.

[About the sexual hygiene of mental retarded boys, an exercise of the sexual pedagogic (author's transl)].

The sexual hygiene of mental retarded boys is examinated. In comparison with normal school boys is pointed out the necessity of sexual pedagogic education. The striking more numerous quote of balanitis in mental retarded boys is discussed with a critical review of Literature about the cleanness of the hands while miction of man, about the cleanness of the inferior abdomen, the cleanness of the pants and the hygiene of the toilet. The conclusion is drawn, that the cleaning of the foreskin has also to be observed by the mental retarded men, boys once a week and youths day by day.
