An attempt to obtain a specific antiserum for in vitro immunological conditioning of bone marrow in acute non-lymphoblastic leukemia.

An antiserum was produced in rabbits to acute undifferentiated leukemia (AUL) antigens. Partial absorption of the antiserum allowed the removal of the cytotoxic activity against normal mononuclear cells of peripheral blood and bone marrow, in contrast to blast cells from acute and chronic leukemias, which were killed by the antiserum in a standard NIH cytotoxicity test. These observations suggest that antigens present on blast cells of AUL are also expressed on some acute and chronic leukemic cells and that these antigens could be interpreted as B or leukemia-associated antigens because of the disappearance of all cytotoxic activity of the immune serum after extensive absorption with B cells from chronic lymphoid leukemia.

Cryopreservation of human lymphoid cells from various tissues.

Normal and pathological lymphoid cells, collected from different sources, were cryopreserved using a programmed freezing procedure. With this cryopreservation technique, the percentage of T and B cell surface markers and the proliferative response to mitogens were not influenced by 21 and 35 days of storage in liquid nitrogen. The recovery percentage of the lymphoid cells was satisfactory when fetal calf serum was added, as a protein source, to the medium during freezing and thawing phases, while a very low percentage of cells was recovered if the fetal calf serum was omitted.

A case of Langerhans histiocytosis with HIV-like immunodeficiency.

A case of histiocytosis X (Langerhans type) associated with bullous pulmonary emphysema and acquired immune deficiency, regarding CD4 positive cells, is described. Previous history was remarkable for skin lesions which first appeared in 1981 and progressively worsened, diabetes insipidus diagnosed in 1986, and bullous pulmonary emphysema detected in 1988. Biopsy results of skin lesions were consistent with histiocytosis X. Thyroid gland involvement was found by means of cytological examination. The search for HIV infection (also performed by means of immunoblotting and PCR) was negative. To our knowledge the immunodeficiency detected in histiocytosis X affects the T suppressor lymphocyte subset, so we thought this peculiar case was worth describing.

Friedreich's disease. A linkage study in southern and central Italy.

We studied linkage and linkage disequilibrium between the genetic locus of Friedreich's disease (FRDA) and two maker loci (D9S15 and D9S5) of chromosome 9q13-q21.1 in 49 subjects from 12 families in southern and central Italy. No recombination event occurred between D9S15 and D9S5, or between these polymorphisms and FRDA. Linkage disequilibrium was not observed between D9S15 or D9S5 or the extended haplotypes and FRDA, but was present between the two polymorphisms.

Mitoxantrone alone or in combination chemotherapy (VeMP) as second-line treatment in relapsed or refractory poor-prognosis non-Hodgkin's lymphoma. A report of the Non-Hodgkin's Lymphoma Co-operative Study Group (NHLCSG).

BACKGROUND AND METHODS: From October, 1986 to July 1989, 35 consecutive patients with high- and intermediate-grade non-Hodgkin's lymphoma, relapsed or refractory to first-line-anthracycline-containing regimens, were treated with mitoxantrone alone or in combination chemotherapy (VeMP: Ve = VP-16, M = Mitoxantrone, P = Prednisolone). RESULTS: In the first 15 patients, treated with Mitoxantrone alone, complete response (CR) and partial response (PR) each occurred in 4 patients, for a total response rate of 54%. In the following 20 patients, treated with the VeMP regimen, CR occurred in 10 patients (50%), PR in 1. The overall three-year survival was 27% in the first group and 40% in the second. Acute toxicity was generally mild. No patient developed cardiac symptoms or other toxicities requiring discontinuation of therapy. Myelosuppression was the most important side effect, being more remarkable for patients treated with VeMP regimen. CONCLUSION: Mitoxantrone, alone or in combination chemotherapy, appears to be a drug with significant activity in aggressive non-Hodgkin's lymphomas.

[Carcinoid of the appendix. Consideration on 5 incidentally diagnosed cases].

5 cases of carcinoid of the appendix discovered during 1094 appendectomies are described. Neither carcinoid syndrome or local metastasis was observed in any of the patients. The importance of the histological examination of all appendices removed is emphasised. Where there is no metastasis, radical surgery is appropriate. The prognosis is excellent but patients should be monitored by the measurement of 5HIIA in the urine.
