[Mutants of ColE1 plasmid with altered expression of the colicin E1 immunity gene].

Mutagenesis with Tn1 transposon was used to isolate mutants of ColE1 plasmid with inactivated gene responsible for immunity to colicin E1. Cells containing such mutants synthesized active colicin E1 and were sensitive to its action. Spontaneous and UV-induced colicin synthesis was strongly changed in the mutants, as compared to the control. Mutations occurring outside the immunity gene, including those within the structural gene for colicin E1, could also affect the immunity gene expression.

[Detection of plasmids in actinomycetes that produce oxytetracycline and neomycin].

Crude preparations of DNA isolated from Act. rimosus 907, an oxytetracycline-producing organism and Act. fradiae 676, a neomycin-producing organism were analyzed with electrophoresis in agarose gel followed by electron microscopic analysis of DNA isolated from the gel. It was found that Act. rimosus and Act. fradiae contained ring molecules of DNA with the molecular masses of 37 . 10(6) and 59 . 10(6) dalton respectively.

[Isolation of messenger RNA for rabbit globin].

Methods are described of isolation of individual globin messenger RNA from rabbit reticulocytes using zonal centrifugation in sucrose density gradient and specific sorption of polyribosome RNAs on poly U-cellulose column. The addition of globin RNA into cell-free system from Krebs-2 ascites mouse cells resulted in the globin synthesis.
