186Re-HEDP: a potential therapeutic bone agent.

A lyophilized kit preparation of HEDP for labelling with 186Re has been developed as a potential bone agent useful for both diagnosis and therapy. High pressure liquid chromatographic analysis showed that the 186Re-HEDP is a mixture of several components. Preliminary in vivo studies in several animal species showed that both the 186Re-HEDP mixture and the HPLC-purified main component are bone-seeking agents with slow blood and soft-tissue clearance rates. HPLC purification slightly improved the quality of the image. In mice and rats with trauma-induced osteogenetic activity in one leg, the bone uptake of the traumatized leg was 1.7-1.9 times higher than of the normal leg. The lesion-to-normal bone ratio was 4-5.4.

Preparation and characterization of [99mTc(DMPE)2X2]+, X = Cl, Br (DMPE = 1,2-bis(dimethylphosphino)ethane).

The preparation of "no-carrier-added" [99mTc(DMPE)2X2]+ (X = Cl, Br) in greater than 95% radiochemical purity, suitable for in vivo evaluation as a myocardial imaging agent, is described. Reaction parameters that lead to the formation of the desired products, as well as those that lead to the formation of the major impurities, are delineated. These 99mTc species are characterized by HPLC using the known 99Tc analogs as standards. Myocardial images obtained in mongrel dogs with [99mTc(DMPE)2X2]+ (X = Cl, Br) are briefly discussed.

Complexing of Tc-99m with cyclam: concise communication.

The macrocyclic amine, cyclam, has been found to be an efficient complexing agent for Tc-99m. Complexes can be formed in yields of higher than 95% at pH 11 in 10(-3) M cyclam using 5 x 10(-6) M SnC4H4O6 as a reducing agent. The complex is positively charged and is stable in air. It is not decomposed by dilution, by 0.05 M NaOH, or 0.05 M H2O2, but does show slight decomposition in 0.05 M HCl. In unanesthetized mice the complex is cleared rapidly from the blood by the kidneys and liver. Preliminary studies show that derivatives of cyclam also complex Tc-99m.
