Effects of restriction of amino acid supply to the isolated perfused guinea-pig mammary gland.

When individual essential amino acids were omitted for periods of 40--100 min from the infusate substrate solution in isolated perfused guinea-pig mammary gland experiments, uptake of methionine, tyrosine, phenylalanine, histidine and tryptophan (group 1) was significantly depressed by a mean of 49.8%, whereas the remaining essential amino acids (group 2) showed no significant decrease in uptake. During depletion periods oxidation of [14C]amino acids was increased. The possible significance of the differences in absorption between the 2 groups of amino acids is discussed.

Effects of exogenous cortisol on the uptake of corticosteroid by the isolated perfused guinea-pig mammary gland.

Assay of corticosteroids in the plasma of guinea-pigs showed that concentrations were higher in lactating than in non-lactating and male animals. In five isolated perfused mammary gland experiments in which corticosteroids were at concentrations of 1,3--3,0 microgram/ml in the perfusate, equivalent to concentrations during lactation, the mean (+/- S.E.M.) uptake by the mammary gland was 900 +/- 210 ng g-1 h-1. In five experiments in which the concentrations of corticosteroid in the perfusate were 190--580 ng/ml, the uptake by the mammary gland was significantly lower (316 +/- 73 ng g-1 h-1; P less than 0.05). For the ten experiments there was a significant correlation between the concentration of corticosteroid in the perfusate and uptake by the mammary gland (P less than 0.05). No convincing evidence for a galactopoietic role of corticosteroids in the guinea-pig was obtained, although administration of cortisol signficantly stimulated the uptake of glutamine by the perfused gland.

Dystocia in a snake.

Herpetology is a rapidly growing interest in this country, reptiles being kept both for research purposes and as pets. However, there is a lack of literature on reptilian medicine and surgery; the authors know of only one available text (Frye 1973). Furthermore, there are only two published case histories of dystocia in the snake (Huff 1976, Hime 1976) and thus this case was considered to be of particular interest.

Evaluation of five protocols for quantification of two-dimensional MR spectroscopic images.

Five protocols were tested for quantification on in vivo two-dimensional spectroscopic imaging datasets. The datasets are duplo measurements from three individuals. The peaks for N-acetyl aspartate, creatines, and cholines were quantified. The first of the protocols is conventional integration of a selected interval around the peak. The others are various implementations of a Gauss-Newton-based least-squares time-domain fitting algorithm. Fitting a half-echo (free-induction decay curve, FID) to a full echo was compared; this was combined with or without filtering out the water signal using the Lanczos-Hankel singular-value decomposition (LHSVD). It appears that conventional integration yields consistent and accurate results in comparison to the fitting methods. The combination of LHSVD and echo fitting performs equally well; advantages and disadvantages are discussed. The protocols using FID fitting perform poorly with these experimental datasets.

NMR studies of interactions between inhibitors and porcine pancreatic phospholipase A2.

Two-dimensional NMR studies were performed on the complexes of porcine pancreatic phospholipase A2, bound to a micellar lipid-water interface of fully deuterated dodecylphosphocholine, with competitive inhibitors derived from the following general structure: [formula: see text] X and Y are alkyl chains with various 'reporter groups'. The interactions between the inhibitor and the enzyme were localized by comparison of 2-D nuclear Overhauser effect spectra using protonated and selectively deuterated inhibitors, and inhibitors with groups having easily identifiable chemical shifts. These experiments led us to the following conclusions for the phospholipase A2/inhibitor/micelle complex: i) the His48 C2 ring proton is in close proximity to both the amide proton and the methylene protons at the sn-1 position of the glycerol skeleton of the inhibitor, ii) the acyl chain of the inhibitor at the sn-2 position makes hydrophobic contacts near Phe5, Ile9, Phe22 and Phe106; iii) no interactions between the acyl chain at the sn-1 position and the protein could be identified. Comparison of our results on the enzyme/inhibitor/micelle ternary complex with the crystal structure of the enzyme-inhibitor complex shows that the mode of inhibitor binding is similar. However, in several cases we found indications that the hydrophobic chains of the inhibitors can have multiple conformations.

Induction of parturition in dairy cows with dexamethasone.

Sixty Holstein cows were paired by parity and sire, and one of each pair was allocated at random to treatment or control; 17 cows were injected with 7.5 mg/100 kg dexamethasone trioxa undecanoate 14 days before the predicted date of calving, 13 cows received the same dose five days before term and 30 cows were left untreated. The treatment significantly advanced parturition and 29 of the 30 induced cows calved within 72 hours of the injection. Induction at day 14 before term was safe for calf and dam, the calves were 3.2 kg lighter than control calves and there was a high incidence of retained placenta. Treatment for this condition resulted in increased veterinary costs of 14.50 pounds per cow exclusive of dexamethasone treatment. Treatment at this stage was also associated with low pregnancy rates in the next breeding season. Calves born after induction at five days before term were not significantly lighter than calves from control cows, the problem of retained placenta was less marked and there were no subsequent effects on fertility. There were no significant effects of induction on milk yield or milk quality up to 200 days of lactation.
