Usher syndrome 1D and nonsyndromic autosomal recessive deafness DFNB12 are caused by allelic mutations of the novel cadherin-like gene CDH23.

Genes causing nonsyndromic autosomal recessive deafness (DFNB12) and deafness associated with retinitis pigmentosa and vestibular dysfunction (USH1D) were previously mapped to overlapping regions of chromosome 10q21-q22. Seven highly consanguineous families segregating nonsyndromic autosomal recessive deafness were analyzed to refine the DFNB12 locus. In a single family, a critical region was defined between D10S1694 and D10S1737, approximately 0.55 cM apart. Eighteen candidate genes in the region were sequenced. Mutations in a novel cadherin-like gene, CDH23, were found both in families with DFNB12 and in families with USH1D. Six missense mutations were found in five families with DFNB12, and two nonsense and two frameshift mutations were found in four families with USH1D. A northern blot analysis of CDH23 showed a 9.5-kb transcript expressed primarily in the retina. CDH23 is also expressed in the cochlea, as is demonstrated by polymerase chain reaction amplification from cochlear cDNA.

Characteristic trend of persistent organochlorine contamination in wildlife from a tropical agricultural watershed, south India.

The residue levels of persistent organochlorines, such as HCH (BHC: 1,2,3,4,5,6-hexachlorocyclohexane) isomers, DDT [1,1,1-trichloro-2,2-bis (p-chlorophenyl) ethane] compounds, PCBs (polychlorobiphenyls) and HCB (hexachlorobenzene), were measured in wildlife. The wildlife were sampled from terrestrial and aquatic habitats in and around the agricultural watershed of Parangipettai, South India. On the basis of overall concentrations sigma HCH ranked first followed by sigma DDT, sigma PCB and HCB, reflecting the increasing usage of HCH in recent years in India. The residue levels of organochlorines in birds varied according to their feeding habits and showed the following pattern: inland piscivores and scavengers greater than coastal piscivores greater than insectivores greater than omnivores greater than granivores. High levels of HCH and DDT residues were recorded in Pond heron and Cattle egret which feed in the agricultural fields. Comparison of HCH concentrations in fish and birds in the study area to other locations was made to further understand the dynamics of contaminant accumulation in tropical wildlife. Regarding birds, a wide variation in residue levels could be seen among various countries, with tropical regions registering high levels. In contrast, the variation is not prominent in the case of fish. The residue levels in fish measured in the present study were generally comparable to values reported from other locations. This suggests that the bioavailability of contaminants to the aquatic fauna is less due to the smaller flux and shorter residence time of these chemicals in the tropics.

Genetic studies on the Kolams of Andhra Pradesh, India.

A total of 220 persons belonging to the Kolam tribe in the Adilabad District of northwest Andhra Pradesh have been tested for 18 red cell eyzyme systems and for haemoglobin. Generally, the gene frequencies for the systems which showed electrophoretic variation were within the range for Indian populations; the gene frequency for AK1 is high by Indian standards and Hb AS is present. LDH Calcutta 1 was not detected in this population and a single example of PHI 2-1 was observed.

Variation and inheritance of relative length of index finger in man.

The length of index finger relative to fourth finger has been measured on parents and children in 190 families belonging to an endogamous Reddy community of Nalgonda District, India. The distribution of relative index finger length among various family groups was unimodal and symmetric. Heritability estimates based on parent-offspring regressions indicate a moderate level (40--70 percent) of additive genetic variance. There was no evidence for the influence of sex-linked additive genes.

Adolescent

The genetic composition of an endogamous Adi-Dravidar population of Tamil Nadu.

A total of 204 blood samples were collected from healthy, unrelated Adi-Dravidars of Chengalput District, Tamil, India. These samples were screened for ABO and RH blood groups using the respective antisera. CP and ALB serum protein electrophoretic phenotypes were assessed. The chi 2 analysis for Hardy-Weinberg equilibrium revealed that the population fits in panmictic equilibrium. The results were compared with the findings reported for other populations of Southern India.
