Entropic forces in dilute colloidal systems.

Depletion forces can be accounted for by a contraction of the description in the framework of the integral equations theory of simple liquids. This approach includes, in a natural way, the effects of the concentration on the depletion forces, as well as energetic contributions. In this paper we systematically study this approach in a large variety of dilute colloidal systems composed of spherical and nonspherical hard particles, in two and in three dimensions, in the bulk and in front of a hard wall with a relief pattern. We show by this way the form in which concentration and geometry determine the entropic interaction between colloidal particles. The accuracy of our results is corroborated by comparison with computer simulations.

Effects of captopril, losartan, and nifedipine on cell hypertrophy of cultured vascular smooth muscle from hypertensive Ren-2 transgenic rats.

1. We hypothesized that tissular renin-angotensin system (RAS) induces vascular hypertrophy in hypertensive Ren-2 transgenic rats (TGR; strain name TGR(mRen2)L27). This assumption was tested in cell cultures of vascular smooth muscle (VSMC) from both hypertensive TGR and control normotensive Sprague-Dawley (SD) rats. Planar cell surface area, protein synthesis, and protein content per cell were studied, the role for locally produced angiotensin II (AII) was evaluated and the possible pharmacological interference by different drugs was analysed. 2. By use of radioimmunoassay techniques, AII could be determined in TGR cultures (10.25 +/- 0.12 pg per 10(7) cells) while it could not be detected in SD ones. 3. Under serum-free conditions, VSMC from hypertensive TGR were hypertrophic when compared to SD VSMC, as they presented a higher protein content per cell (335 +/-18 and 288 +/- 7 pg per cell respectively; P<0.05) and increased mean planar cell surface area, as determined by image analysis (4,074 +/- 238 and 4,764 +/- 204 microm2, respectively; P < 0.05). 4. When exogenously added to cultured SD and TGR VSMC, AII (100 pM to 1 microM) promoted protein synthesis and protein content in a concentration-dependent manner without affecting DNA synthesis. Maximal effects were observed at 100 nM. At this concentration, AII effectively increased planar cell surface area in both SD and TGR cultures by approximately 20%. 5. Treatment of TGR cultures, in the absence of exogenous AII, with the angiotensin-converting enzyme inhibitor captopril or the angiotensin AT1 receptors antagonist losartan (100 nM to 10 microM) reduced planar cell surface area in a concentration-dependent manner. In addition, both captopril and losartan (10 microM), decreased protein synthesis by approximately 15%. 6. Treatment of SD VSMC, in the absence of exogenous AII, with both captopril and losartan had no effect either on planar cell surface area or protein synthesis. 7. Treatment with the Ca2+ antagonist nifedipine (100 nM to 10 microM) reduced cell size in both SD and TGR cultures. Maximal cell reduction reached by nifedipine averaged 906 +/- 58 and 1,292 +/- 57 microm2, in SD and TGR, respectively (P<0.05). In addition, nifedipine, nitrendipine and nisoldipine (all at 10 microM) decreased protein synthesis in both cell types by 15-25%. 8. We concluded that cultured VSMC from TGR are hypertrophic in comparison with those from SD. This cell hypertrophy can be the consequence of the expression of the transgene Ren-2 that activates a tissular RAS and locally produces AII, which acts in a paracrine, autocrine, or intracrine manner. Cell hypertrophy in TGR cultures could be selectively reduced by RAS blockade, while nifedipine decreased cell size and protein synthesis in both hypertrophic and non hypertrophic cells.

The effect of LU-49938 (nexopamil) on the activation by serotonin of mesangial cells.

Intraglomerular platelet activation may release vasoactive agents such as serotonin (5-hydroxytryptamine, 5-HT) that may affect local hemodynamics and promote mesangial proliferation, eventually leading to glomerular sclerosis. The main purpose of this study is to analyze whether nexopamil, a verapamil derivative, with the property of blocking simultaneously calcium channels and 5-HT2 receptors, could modify the contractile and mitogenic effects of serotonin on cultured rat mesangial cells. Serotonin caused a concentration-dependent increase in [3H]thymidine incorporation into DNA, and mesangial cell proliferation. The effects of 5-HT on thymidine uptake and cell proliferation were blocked by the selective 5-HT2 receptor blocker ketanserin 10(-5) M. Nexopamil abolished in a concentration-dependent way the serotonin-induced [3H]thymidine incorporation into DNA, and the serotonin-induced increase in number of cells. Using 5-HT 10(-4) or 10(-5) M, nexopamil had significant effects at concentration above 10(-7) M. Serotonin induced a concentration- and time-dependent reduction of planar cell surface area. This effect was also completely blocked by ketanserin. Nexopamil partially blocked the serotonin-induced mesangial cell contraction, in a dose-dependent manner. All these data suggest that nexopamil inhibits both 5-HT-induced mesangial cell contraction and proliferation by blocking 5-HT2 receptors and the voltage-operated Ca2+ channels.

[Prevalence of dental caries and associated factors in children at day care centers].

UNLABELLED: Currently dental caries is regarded as a public health problem throughout world. Ninety percent of the Latin American population suffers from caries. In Mexico, a research informs of a prevalence of 78% in 5-year-old children from the Southern of the Federal District, while the nearby State of Mexico reports 90% in children the same age that suffers from this same problem. OBJECTIVE: Identifying the prevalence of dental caries, dmf-t and dmf-s indexes, and the relation ship with associated factors in daycare centers of the # 3 Southwest Delegation of the Instituto Mexicano del Seguro Social (IMSS). A total of 934 children from ages 1 to 6 years of age from October 1997 to May 1998, were studied; 52.3% were boys and 47.7%, girls. The presence of caries was examined and determined through WHO criteria. Socioeconomic level was identified the parents questionnaire. Prevalence of caries in the population studied was 22.1%; we also found a decayed, missing, filled-teeth index (dmf-t) for the whole population of 0.71. The average of this index was 3.71.

Expression of the beta-isoforms of Na,K-ATPase in the renal cortex of rats.

The aim of the present study was to assess the presence of beta1- and beta2-isoforms of the beta-subunit of Na,K-ATPase in the rat renal cortex. This has been accomplished by immunohistochemistry and Western blotting using isoform-specific antisera. Western blot of brain extract, used as positive control, revealed the bands corresponding to beta1- and beta2-glycosylated peptides, with a molecular weight (MW) of approximately 50-60 that, after exhaustive treatment with N-endoglycosidase-F, migrated to the MW corresponding to the core peptides (approximately 35). In the renal cortex, Western blot revealed the bands corresponding to beta1. After deglycosylation of the samples, the bands hybridizing with the anti-beta1-antibodies moved to the MW corresponding to a partially deglycosylated form and the core peptide. Bands with a MW of approximately 50-60 hybridized with anti-beta2, although digestion with endoglycosidase failed to move the band towards a lower MW. Immunohistochemistry revealed the presence of beta1- but not beta2-isoform. Northern blot for total mRNA showed strong signals for beta1 in renal cortex, the mRNA for the beta2-isoform being undetectable. In conclusion, only mRNA and glycopeptide of the beta1-isoform seem to be present in renal cortex of adult control rats.
