Biological activity of rat proinsulin synthesized by Escherichia coli.

Fused proteins possessing insulin antigenic determinants are synthesized and secreted by several strains of Escherichia coli K-12 bearing plasmids containing cDNA copies of rat proinsulin mRNA. Small amounts of the secreted, fused protein from E. coli strains chi 1776 and HB101 bearing plasmid p147 were partially purified and assayed for biological activity by determining stimulation of [1-14C]glucose oxidation to 14CO2 in the rat epididymal fat pad assay. Guinea pig anti-insulin serum (GPAIS) was used to suppress glucose oxidation stimulated by insulin. After mild treatment with trypsin, fused protein stimulated 14CO2 production, and over 90% of this activity was suppressible by GPAIS. Untrypsinized fused protein demonstrated no GPAIS-suppressible 14CO2 production. Larger amounts of fused protein were obtained by transformation of strain PR13 of E. coli in which the yield of eukaryotic proteins is increased approx. 50-fold. A single intravenous injection of trypsinized fused protein from E. coli PR13 containing plasmid p287.47 resulted in a rapid decline in the plasma glucose levels of fasted mice, and the hypoglycemic effect persisted for at least 120 min.

[Laboratory diagnosis: correlated health and environmental risks].

Method description and initial results of a study to assess risks to health sector workers and environment due to chemical agents used and waste products generated in diagnostic clinical chemical laboratories, and image diagnostic testing. A survey was conducted of the methods and agents used and their toxicological classification, the number of workers exposed and an analytical profile of the waste produced. The assessment of risk to workers was based on cytogenetic tests (chromosome aberrations and micronuclei); the assessment of environmental risk from waste disposal was based on tests on plant systems.

[Hospital hygiene and disinfectants: experience in a teaching hospital].

A survey was conducted during March-July 1999 to assess and identify the disinfectants and chemical sterilants employed in a teaching hospital of Roma. The purposes were studying the problems of safety in the use also, and the treatment like waste of disinfectants and chemical sterilants. Were collected the data from 61% of 39 Department/Institutes of the whole hospital. A total of 635 records were examined, and 40 different active ingredients of 110 commercial forms were identified. This study was conducted by a survey chart. Data collected outline that to minimize the risk for nursing staff, workers and the patients a modern hospital staff should prepare a handbook that can explain the concentrations and correct use for various disinfectants and chemical sterilants that was choose and employed.

[Activities at the Rome Anti-Rabies Center in 2000: analysis and proposals].

The Authors analyzed data from the 1,817 patients (mean age 35.3 +/- 20.6, median 32) treated at the Antirabies Centre of Rome in the year 2000; of these 8.9% were immigrants. Dogs (84.1%) and cats (9.8%) were responsible for most bites. Stray animals were 39.9%, with an owner 58.3% and those without data 1.8%. Over 1/3 of patients could not refer the cause of bite. Most patients (93.0%) went first to hospital, where tetanus prophylaxis was administered preferably by immunoglobulins (25.0%) instead of vaccine (6.6%). Antirabies vaccine (HDCV and/or DEV) was prescribed to 777 patients (42.8%) and administered to 642 (35.3%), but only 296 (16.3%) of them received at least 5 doses. Veterinary observation always confirmed the absence of rabies in central Italy. Immunoglobulins were prescribed only to 14 patients. The data shown in this study emphasize the opportunity to differentiate the Italian antirabies protocol according to the geographic area.

Antibody response after vaccination with antigen-pulsed dendritic cells.

Dendritic cells (DCs) are the most potent antigen-presenting cells of the immune system capable of initiating immune responses to antigens. It is also well documented that cancer patients often experience anergy against tumor antigens. In this study we selected the best protocol for inducing the production of antibodies against the HER2 oncoprotein using DCs to overcome anergy. Murine DCs were pulsed in vitro, using different protocols, with recombinant HER2 fused to a human Fc (in order to improve DC antigen uptake) and were used to vaccinate mice. The obtained results indicate that antigen-pulsed DCs can induce an antibody response and that adding CpG after antigen pulsing greatly increases anti-HER2 antibody production.

Continuous measurement of vascular access recirculation by conductivity.

Vascular access recirculation is one of the main factors influencing dialysis efficiency. The underestimation of or mistakes in the measurement of access recirculation causes a discrepancy between dialysis prescription and the actual dose of dialysis administered. Moreover, access recirculation is known to increase during a dialysis session. Therefore, the evaluation of recirculation is necessary to correctly prescribe dialysis and the ideal measurement method should be very accurate and feasible at any moment during a dialysis session.
