Efficacies of liposome-encapsulated enrofloxacin against Staphylococcus aureus infection in Anatolian shepherd dog monocytes in vitro.

The aim of this study was to evaluate the intracellular activity of two types of liposome-encapsulated enrofloxacin (LE) compared with free enrofloxacin and non-treated control against Staphylococcus aureus, phagocytosed by monocytes in healthy Anatolian Shepherd dogs. Enrofloxacin was encapsulated with two different types of liposome in multilamellar large vesicles (MLV). Type A MLV were composed of 15 mg phosphatidylcholine and 35 mg cholesterol, Type B MLV were composed of phosphatidylcholine, cholesterol and enrofloxacin in a molar ratio of 1:1:1. Intracellular activity was estimated by comparing the numbers of bacteria surviving intracellularly in monocytes exposed to free enrofloxacin and LE for 4 h at the doses of 0.25, 0.5 and 1 microg/ml, with those surviving intracellularly in untreated control monocytes. All three forms of enrofloxacin (free, Type A and B liposomes) increased the intracellular killing of S. aureus in a concentration dependent manner. Comparison of 1 microg/ml Type B LE revealed that killing activity was significantly higher than those of other concentrations. The results showed that LE was superior in reducing the number of intracellularly located bacteria compared to the free drug and control. The beneficial effect of liposomal encapsulation is presumably due to the fact that both liposomes and bacteria are localized at the same spot in phagocytic cells.

Determination of intracellular concentrations of free and two types of liposome-encapsulated enrofloxacin in anatolian shepherd dog monocytes.

In this study, it was evaluated the accumulation of free and two types of liposome-encapsulated enrofloxacin (LEE) at the doses of 0.25, 0.5 and 1 microg/ml, which were clinically relevant concentrations into monocytes of healthy Anatolian shepherd dogs. Enrofloxacin was encapsulated with two different types of liposome in multilamellar large vesicles (MLV). Type A MLV composed of 15 mg egg phosphatidylcholine and 35 mg cholesterol, Type B MLV composed of phosphatidylcholine (PC), cholesterol and enrofloxacin, in a molar ratio of 1 : 1 : 1. The mean sizes of Type A and Type B liposome were found to be 7.65 and 4.27 microm, respectively. However, the mean encapsulation rate determined of Type A (13 +/- 2%) was found lower than Type B liposome (44 +/- 3%). The amounts of intracellular enrofloxacin concentrations were determined by high performance liquid chromatography. Type B LEE accumulated significantly higher level into monocytes when compared to free drug or Type A liposome. This study showed that Type B LEE markedly concentrated within monocytes and may improve the antibacterial efficacy of the antibiotic.

Endovascular treatment of massive retroperitoneal haemorrhage due to inferior adrenal artery injury. A case report.

Massive retroperitoneal haemorrhage because of adrenal bleeding following blunt trauma is a rare entity. We describe a 23-year-old woman with massive retroperitoneal haemorrhage as a result of inferior adrenal artery (IAA) disruption following a car accident. Superselective transcatheter embolization of the IAA using polyvinyl alcohol particles was performed successfully to control haemorrhage. To our knowledge, this is the second report of massive retroperitoneal haemorrhage caused by IAA disruption successfully treated by transcatheter embolization.

Transient frontal hypoperfusion in Tc-99m hexamethylpropyleneamineoxime single photon emission computed tomography imaging during alcohol withdrawal.

BACKGROUND: Regional brain perfusion in patients during alcohol-withdrawal has been relatively less studied with brain SPECT technique. In this study, the hypothesis that possible regional cerebral blood flow (rCBF) alterations due to alcohol withdrawal might be transitory in a homogenous group of alcoholic patients in terms of their physical-nutritional and cognitive functional conditions was investigated. METHODS: Fifteen right-handed male inpatients with alcohol-withdrawal, diagnosed according to DSM-IIIR criteria, and 6 male physically-mentally healthy control subjects were included in the study. The first Technetium 99m-hexamethylpropyleneamineoxime (Tc-99m-HMPAO) brain SPECT investigation was performed on the day of admission in nonmedicated conditions and the second one was performed after all the withdrawal symptoms had subsided in the patients. As an indicator of the change in the brain perfusion, a relative perfusion index was used and the relative tracer activity was expressed as the ratio of mean cortical region of interest activity to mean the whole cortical brain activity. RESULTS: We found significantly reduced left frontal and right frontal, parietal and temporal rCBF values in the patients during the alcohol-withdrawal compared to those of their remitted state while they were not different from in the control group (p < 0.05). CONCLUSIONS: Our data indicate that the alterations in rCBF during the alcohol-withdrawal are more pronounced both in the frontal cortex and in overall right hemisphere regions. Furthermore, the frontal hypoperfusion may be transitory with recovery from alcohol-withdrawal whereas temporal hypoperfusion may continue after recovery probably depending on the previously administered high-dose benzodiazepines.

Effects of dexfenfluramine on glucose drinking and glucose-conditioned flavour preferences in rats: taste versus post-ingestive conditioning.

A comparison was made of the effects of dexfenfluramine (DF, 3-10 mg kg-1) on intake of and conditioning with glucose solutions varying in orostimulant properties (taste) and post-ingestive actions (calories), in rats. First, sham-feeding, using gastric-fistulated rats, was performed to assess the orostimulant properties of the solutions. Then, two experiments were done. In the first experiment, we examined the effects of DF, given at doses of 13 and 10 mg kg-1, on the intake of two glucose solutions having different orostimulant properties and different caloric values. The solutions were a mix of 1% glucose plus 0.125% saccharin (low caloric, more orostimulant), and 4% glucose (high caloric, less orostimulant). At doses of 10 mg kg-1, DF administration markedly reduced intake of both solutions (P < 0.05 vs Control Group, respectively). In second experiment, we examined the effects of DF (10 mg kg-1) on flavour preference conditioning in two parts. In the first part of the experiment, rats consumed two distinctively flavoured solutions having equal orostimulant properties but different caloric value for a conditioning period of 16 days. The solutions were a mix of 1% glucose plus 0.125% saccharin (low caloric), and 6.1% glucose (high caloric). At the end of the conditioning period, the flavour paired with ingestion of more calories was subsequently preferred (P < 0.05 vs low caloric glucose-saccharin mix). DF, when given during and after the conditioning period, attenuated this flavour-calorie conditioning (P < 0.05 vs Control Group). In the second part of the experiment, rats were conditioned with flavours associated with a mix of 20% glucose plus 0.4% citric acid and 20% glucose solutions. These solutions were equally caloric but differed in orostimulant properties. The flavour paired with better orostimulant properties was subsequently preferred (P < 0.05 vs less orostimulant glucose-citric acid mix). DF, when given during and after the conditioning period, also attenuated this flavour-flavour conditioning (P < 0.05 vs Control Group). These results suggest that DF may impair flavour preference learning.

Clinical and magnetic resonance imaging features of L-2-hydroxyglutaric acidemia: report of three cases in comparison with Canavan disease.

We report three cases of L-2-hydroxyglutaric acidemia and three cases of Canavan disease. The L-2-hydroxyglutaric acidemia cases are the first biochemically proven Turkish cases. Magnetic resonance imaging findings in the cases and similarities between the two diseases are emphasized. Both diseases are characterized by predominant subcortical white-matter involvement and dentate nuclei lesions with variable basal ganglia involvement. Canavan disease differs from L-2-hydroxyglutaric acidemia by the presence of typical brainstem involvement.

Surgical treatment of rectal prolapse. A retrospective analysis of 94 cases.

METHODS: A retrospective analysis evaluating 94 patients who underwent posterior rectopexy in 48 patients (51%), resection with or without rectopexy in 19 patients (20%) and Delorme's procedure in 27 patients (29%) was carried out for rectal prolapse in the last 15 years. The surgical procedures are described in detail. Postoperative evaluation was possible in all patients and mean observation time was 3.2 years. RESULTS: Mortality rate was zero. Recurrence was seen in 4 cases (4.2%), only after Delorme's procedures. The proportion of continent patients increased from 69.2% preoperatively to 91.6% postoperatively. Defecation difficulties and incomplete evacuation did not change beneficially after the surgery. Important postoperative complications, retrograde ejaculation and impotence were seen in 5 male patients (17.2) after posterior rectopexy, and were a major cause of dissatisfaction. CONCLUSIONS: In conclusion, Delorme's procedure, posterior rectopexy and resection procedures are effective surgical operations for treatment of rectal prolapse but extensive pelvic dissection during the posterior rectopexy may create serious sexual problems in male patients.
