Chromatin structure in dementia.

Nuclei extracted from neocortex of patients with Alzheimer's disease and treated with micrococcal nuclease release a population of dinucleosomes that contain an increase in the linker histones H1o and H1oo . Five other degenerative brain diseases that clinically resemble Alzheimer's disease do not result in these changes, although Pick's disease is associated with an increase in H1 on dinucleosomes. Histones from nuclei of patients with Alzheimer's disease are also more resistant to salt-induced release from chromatin than are those from age-matched control subjects. These results support the hypothesis that an alteration in chromatin structure is a marker for Alzheimer's disease.

Amoxicillin and clavulanic acid combination: clinical and laboratory evaluation in pulmonary infections.

Twenty-four patients with pulmonary infections (7 pneumoniae, 17 exacerbations of chronic bronchitis) were treated with a combination of 250 mg of amoxicillin and 125 mg of clavulanic acid supplemented by 500 mg of amoxicillin every eight hours. All patients showed good clinical, roentgenological, and bacteriological response. Bacteria were cleared from sputum by the third day of treatment in half of the patients and by the end of treatment in all but one of the others. Pathogens eradicated from sputum, with concurrent clinical cure, included gram-negative bacteria producing cephalosporinases shown to be resistant to the combination drug by both disk and minimal inhibitory concentration determinations.

Cefotaxime: clinical and bacteriologic evaluation.

Cefotaxime, a new parenteral beta-lactamase-resistant cephalosporin, was tested against 78 cephalothin-resistant clinical isolates of Enterobacteriaceae and Pseudomonas species. The anti-bacterial activity of the drug was compared with activities of cefazolin and cefuroxime and was found considerably more active. Most of the cephalothin-resistant strains were resistant to cephazin, too. The antipseudomonal activity of cefotaxime was found limited. The drug was found effective against 23 infections in hospitalized patients with only one clinical failure and two colonizations with cefotaxime-resistant bacteria. Cephalosporinases hydrolyzing cefotaxime were detected in two cefotaxime-resistant strains. Also, beta-lactamases hydrolyzing cefoxitin and cefuroxime were produced by the cefotaxime-resistant isolates studied.

Occult intralabyrinthine schwannoma.

Report of a case of an intralabynrinthine schwannoma discovered unexpectedly during a labyrinthectomy for intractable vertigo, and description of the light and electron microscopic appearances. Although only 18 of these tumors have been previously reported, it is possible that some cases which are categorized as peripheral vertigo of unknown origin, may be due to an occult Schwannoma. All material removed during surgical procedures on the inner ear should therefore be submitted for histopathological examination.

The effect of pre- and postnatal undernutrition on axonal growth and myelination of central motor fibers. A morphometric study on rat cortico-spinal tract.

A morphometric study of the axon circumference and myelin sheath thickness in the rat corticospinal tract was undertaken to establish the effect of pre- and postnatal undernutrition and subsequent nutritional rehabilitation. Both axonal growth and myelin deposition were affected by undernutrition. Rats subjected to postnatal nutritional rehabilitation revealed a catch-up in axonal growth; whereas the process of myelination remained affected.

The effect of pre- and postnatal undernutrition on the calibre growth of central motor fibres: a morphometric ultrastructural study on rat cortico-spinal tract.

The effect of early in life instituted undernutrition on the calibre growth of cortico-spinal motor fibres was studied electron microscopically at three different developmental stages in the rat. The total diameters of these motor fibres were measured from a defined level of the medulla oblongata and plotted in histograms. In undernourished rats. Early initiated nutritional rehabilitation through controlled cross-fostering revealed that the impaired calibre growth was reversible, and thus completely restituted at the age of 180 days. It is concluded that early initiated protein-calorie undernutrition affects this central motor pathway in a similar way, as it affects a peripheral motor nerve (cf. Sima, 1974).

Effect of pre- and postnatal protein-calorie undernutrition on RNA-content in rat motor neurons.

Changes in the RNA-content of isolated anterior horn motor neurons from the 7th cervical segment were studied during the postnatal development of pre- and postnatally undernourished, and normally nourished rats. The motor neurons of the anterior horn were isolated by micromanipulation from Carnoy-fixed sections. The neuronal RNA-content was determined by microchemical methods according to Edström (1964). The observation was made that moderate pre- and postnatal undernutrition does not alter the normal RNA-accumulation in anterior horn motor neurons during the preweaning period. After weaning the cellular RNA incorporation decreases but again reaches normal values at the age of 39 days. The present results are compared with those of a previous study (Haltia 1970) in which rats were severely undernourished postnatally.

Liposarcoma of the meninges. A case report.

A report on a liposarcoma in the meninges of a 70-year-old woman is presented. Pre-operative clinical and angiographic findings indicated that the tumour might be a meningeoma. Pathological examination revealed a predominantly lipoma-like, well-differentiated liposarcoma with round-cell and pleomorphic areas. The angiographic and pathological differential diagnosis are discussed.

Central pontine myelinolysis. A case report.

The first case of central pontine myelinolysis in Scandinavia is presented. A premortem diagnosis was established on the basis of the history of chronic alcoholism, the picture of severe electrolyte disturbance and the development of the neurological syndrome. The postmortem findings verified the clinical diagnosis. A possible aetiological mechanism is postulated. The cause is believed to be a damage to the energy dependent electrolyte transport across the cell membrane, resulting in a cytotoxic oedema and death of the oligodendroglial cells of the brainstem which are particularly vulnerable.

The effect of pre- and postnatal undernutrition on the development of the rat cerebellar cortex. I. Morphological observations.

A morphologic study of the postnatal development of cerebellum in normal and pre- and postnatally undernourished rats was initiated in order to evaluate the effect of undernutrition. Defined areas of the cerebellar hemisphere were studied light-microscopically. Parallel to this study a histochemical investigation was performed on the same cerebellar areas (see part II). It was found that pre- and postnatal undernutrition causes a retarded outgrowth of the apical dendritic tree of the Purkinje cells. Furthermore, undernourished rats displayed a delayed persistence of the external granular layer and a retarded differentiation and migration of presumed basket, stellate and internal granular cells which is in accordance with previous studies on cerebellar development in experimental undernutrition.

The effect of pre- and postnatal undernutrition on the development of the cerebellar cortex in the rat. II. Histochemical observations.

The effect of pre- and postnatal undernutrition on the neuronal enzymatic maturation of the neocerebellar hemispheral lobules VI b, VI c and VII (according to Larsell 1952) was studied. Postnatally oxidative enzyme histochemistry (LDH, SDH, NADH, NADPH, G6PDH) revealed a delayed enzymatic development in the cerebellum between days 8 and 22, as an effect of pre- and postnatal undernutrition. In the Purkinje cell apical cone a delayed increase in enzyme activities could be seen at days 8 to 11 as well as a delayed decrease in Purkinje cell perikaryal enzyme activities at days 11 to 14 post-partum. The retarded enzymatic development seemed to parallel the retarded morphological development (cf. Sima & Persson 1975).
