Comparative bioavailability and pharmacokinetics of noscapine hydrogen embonate and noscapine hydrochloride.

The relative bioavailability and pharmacokinetics of two orally administered aqueous suspensions of noscapine hydrogen embonate were compared with those of noscapine hydrochloride solution. Noscapine hydrochloride showed faster absorption and gave a higher peak concentration than the embonate. The average bioavailability of the embonates was 71% of that of the noscapine solution (p less than 0.05). No significant difference was observed when the embonate was administered with water, polyvidone and flavouring agents.

Effect of dosage form and formulation factors on the adherence of drugs to the esophagus.

In recent years, many case reports concerning esophageal injuries caused by drugs have been published. The primary cause has apparently been the adherence of the drug product to the esophagus. In the present study, the adherent tendency of a number of types of tablets and capsules were tested in vitro using a recently developed isolated porcine esophagus preparation. The results showed that the tendency of products to adhere to the esophageal mucosa can be modified to a great extent by shape and formulation. Products with low adherence can be obtained by film coating with aqueous dispersions or by sugarcoating. In contrast, gelatin capsules and some cellulose films appear to have a high tendency to adhere to the esophagus.

Quantitative determination of nitroglycerin in human plasma by capillary gas chromatography negative ion chemical ionization mass spectrometry.

A quantitative method for determination of nitroglycerin in human plasma was developed. Nitroglycerin and the internal standard (butane-1,2,4-triyl trinitrate) were extracted from plasma with pentane. The extracts were analysed by gas chromatography mass spectrometry using fused silica capillary columns and electron capture negative ion chemical ionization. The quantitation limit of the method was about 50 pg ml-1. Linear calibration curves were obtained in the range of 50-1600 pg ml-1. Precision at the level of 100 pg ml-1 was 4%.

Development of a method for study of the tendency of drug products to adhere to the esophagus.

A method to determine the adherence of drugs to the esophagus was developed using isolated swine esophagi. A number of types of tablets and capsules (e.g., of doxycycline) were tested. The results showed that the tendency of products to adhere to the esophagus can be regulated by pharmaceutical properties. It was concluded that the method described is simple, inexpensive, and accurate.

Effect of particle size on the bioavailability of digoxin.

The bioavailability of digoxin in three tablets prepared from materials with different particle sizes was measured in healthy volunteers in a cross-over study using an alcoholic solution of digoxin as a reference standard. Its bioavailability in tablets with particle sizes of 7 mu or 13 mu was 78-97% of that of digoxin in solution. The tablet with largest particle size (102 mu) showed markedly lower bioavailability than the reference solution, namely 39%. It is obvious that particle size is an important determinant of the dissolution rate and bioavailability of digoxin.
