Nucleotide sequence of the replication region of plasmid R401 and its incompatibility function.

The plasmids R401 and Rtsl belong to the same incompatibility group, IncT. The nucleotide sequence of the basic replicon of R401 consisting of 1,857 base pairs was determined and compared with that of mini-Rtsl previously reported. The mini-R401 was found to be composed of two clusters of direct repeated sequences flanking a large open reading frame that could encode a 33,000 Mr protein (RepA protein) consisting of 288 amino acids. This structure of mini-R401 is quite similar to that of mini-Rtsl. Furthermore, the nucleotide sequence of mini-R401 is identical to that of mini-Rtsl except for eleven nucleotides; three are located near the carboxyl terminus portion of the RepA coding region (repA) and four are in the repeated sequences (incI) located downstream from repA. Incompatibility study showed that mini-R401 plasmid coexisted stably with the cloned incI repeats of mini-Rtsl, suggesting that mini-R401 RepA protein binds to incI repeats of mini-Rtsl less efficiently than does mini-Rtsl RepA protein.

Three siblings with Peters' anomaly.

Clinical findings of three siblings with Peters' anomaly and congenital heart disease, and histopathological findings of the second child were reported. These three cases all underwent operation for secondary glaucoma. Schlemm's canal was not observed in the first and second child. Autosomal recessive inheritance was considered the possible cause. As for the mechanism by which this disease is produced, it is surmised that abnormal development of the eye is caused by hypoxia due to ophthalmic circulatory insufficiency in the embryogenic period, and that the already formed lens shifts forward causing this disease, judging from the fact that there was severe tetralogy of Fallot in the first and the third case, and VSD + PDA in the second case, and from the histopathological findings.

Dynamic topography of visual evoked potential in children: a study of the development of the visual system.

To study the development of the visual system, the dynamic topography of the visual evoked potential (VDT) was investigated in 30 normal children aged from two weeks to four years. Twenty-three of the 30 children (77%) showed reliable VDT. The results showed three steps in the development of the visual system in these young children. The first step was seen during the first 4 months after birth, and it was represented by a shortening of duration and an increasing of amplitude in N70 in the occipital region. The second step was seen between 9 months and 2 years of age, and it was represented by a shortening of duration and an increasing of amplitude in P100 and P150 over a widespread area in the occipito-parieto-temporal region. The third step was not clearly seen until 3 years of age, and was represented by the appearance of P200, the so called "vertex potential", around the occipitoparietal region. These three steps in the development of the visual system seemed to be closely related to the time-course of the development in the myelination and maturation of the neuron synapses in the visual pathway and visual cortex. With the aid of VDT, it was possible to clarify that the visual system in children develops step by step from a low level to a high level of the visual function.

Complete nucleotide sequence of mini-Rts1 and its copy mutant.

The nucleotide sequence of the whole mini-Rts1 genome consisting of 1,855 base pairs was determined. In addition to the cluster of five 24-base-pair direct repeats previously detected (Y. Kamio and Y. Terawaki, J. Bacteriol. 155:1185-1191, 1983), another cluster of three 21-base-pair direct repeats was found. All eight repeats were located in one direction and contained a consensus sequence TTCCCCPyPyPuPuCACACACC. Between the two clusters, a large open reading frame that could encode a 32,980-dalton polypeptide consisting of 288 amino acids was assigned. The molecular size predicted from the amino acid composition was close to the value of a unique mini-Rts1 product, the RepA protein, newly determined in minicells. A copy mutant of mini-Rts1 obtained in vitro by hydroxylamine treatment contained two-base-pair substitutions, one of which was located in the RepA protein coding region, and the other was close to the region where the oriC homologous sequences exist.
