CYP19 gene polymorphism in endometrial cancer patients.

PURPOSE: Initiation/promotion of endometrial cancer is known to be associated with estrogenic influence. Therefore, it is possible that some allelic polymorphisms of the genes involved in steroidogenesis or steroid metabolism contribute to endometrial cancer susceptibility. METHODS: Here, we compared CYP19 (aromatase) gene polymorphism in 85 endometrial cancer patients and in 110 non-affected women. RESULTS: The genotypes containing the longest alleles (A6 and A7) of CYP19 were found to be over-represented in patients as compared to controls. In addition, these genotypes demonstrated a tendency to be associated with increased concentrations of estradiol and testosterone in postmenopausal patients. CONCLUSIONS: Thus, CYP19 polymorphism might be one of the genetic risk factors for endometrial cancer development.

Effects of indomethacin and hyperventilation on cerebral hemodynamics and oxygenation in newborn piglets.

We investigated the effect of indomethacin (INDO) and hyperventilation (HV) on cerebral hemodynamics and oxygenation in newborn piglets using near-infrared spectroscopy (NIRS). A total of 10 piglets aged 7-10 days were divided into 2 groups. INDO (Experimental group; 0.2 mg/kg i.v.) or saline (Control group) was administered after steady state, then each group was hyperventilated (pCO2; 20 mm Hg). Cerebral oxyhemoglobin concentrations (HbO2) significantly decreased immediately after INDO administration compared to controls (p < 0.01). This decrease continued for 15 min. During HV, HbO2 remained low compared to the steady state (p < 0.05) in the experimental group. Total hemoglobin concentrations (HbT) decreased simultaneously with HbO2. In the control group, no changes in HbO2 or HbT were observed after saline administration, and only a slight decrease in HbO2 was observed during the 5 minutes' HV compared to the steady state. No changes in cytochrome aa3 (Cytaa3) were observed in either group during 100% O2 inhalation. We conclude that INDO decreased HbO2 and HbT by vasoconstriction, but that HV did not further aggravate this tendency. Great care should be taken when using INDO for newborn neonates.

Heat stroke-like episode in a child caused by zonisamide.

A 2-year-old mentally retarded boy with frontal lobe epilepsy presented with an episode that resembled heat stroke during the administration of zonisamide. He developed hyperpyrexia with oligohidrosis and central neurological symptoms, including, chorea-like involuntary movements, resting tremor, and cogwheel rigidity. A sweat test using pilocarpine iontophoresis revealed a marked reduction in the sweat response, which suggested a postganglionic sweating dysfunction. A skin biopsy examined by light and electron microscopy showed no morphological abnormality in the sweat glands. The oligohidrosis caused by zonisamide was reversible in that the patient regained the ability to sweat within 2 weeks of the cessation of drug administration. Children receiving zonisamide should be monitored for oligohidrosis and the development of neurological symptoms associated with an elevation of body temperature.
