Isolation and subcloning of chitinase clone from chickpea genomic library.

Chickpea genomic library constructed earlier in phage lambda (EMBL-3) was screened for the presence of chitinase clone using tobacco chitinase cDNA as a probe. Positive clones obtained by primary screening of plaques (2 x 10(6)) were ascertained by secondary and tertiary screening. Presence of chitinase insert in the positive clones obtained, was further confirmed by restricting phage DNA with Sal I and then doing southern with tobacco chitinase. The insert band was eluted out and subcloned in puc 19 plasmid.

Paralytic poliomyelitis (1989-1994): report from a sentinel center.

OBJECTIVE: To study the trends of paralytic poliomyelitis in pre Pulse Polio Immunization period. SETTING: Hospital based sentinel surveillance. METHODS: Analysis of 6704 line-listed poliomyelitis cases from January 1989 to December 1994 attending the Department of Physical Medicine and Rehabilitation. RESULTS: 85% of all cases reported in Delhi were from this center. A decline in alternate year peaks was observed from 1621 cases in 1990 to 1062 cases in 1994. There was an increase in proportion of cases with poliomyelitis in fully vaccinated children from 14% to 22.9%. Polio type I virus was the commonest isolated virus in all the years except 1993, when type II was isolated in 38.7% of cases. CONCLUSION: Despite improvement in immunization coverage, the study reveals that a large number of children (67.2%) who suffer from poliomyelitis are unvaccinated. This stresses need to intensify and sustain high level of immunization coverage with effective vaccine.

A study of childhood tetanus in post-neonatal age group in Delhi.

A study was conducted in tetanus in the postneonatal pediatric age group between January, 1989 and June, 1991. Clinical profile of 55 patients was studied and outcome during this period was compared with previous years (1986-88). Although, an overall decline in total admissions was noted, only a modest decline in mortality was observed. All the patients but one were either unimmunized or incompletely immunized. More than 40% patients were from Delhi. Otorrhea was common (49.1%) in these patients. Primary immunization with booster dose in the community is stressed. Children with aural discharge should be taken as a high-risk group for tetanus and be evaluated for immunization at first visit.

A 42 bp promoter fragment of the gene for subunit III of photosystem I (psaF) is crucial for its activity.

The promoter of the gene for the subunit III of photosystem I reaction center (psaF) from spinach has been dissected and studied via promoter/GUS gene fusions in transgenic tobacco. It possesses an architecture that differs from any other spinach promoter of genes encoding proteins involved in photosynthesis studied to date. A 42 bp region located between -220 and -179 bp upstream of the transcription start site has been identified that is indispensable for expression and binds a trans-acting factor. Maximal light-response is obtained with a -220/+ 163 bp segment, whereas longer promoter sequences are significantly less effective, indicating the existence of upstream elements with silencer characteristics. F1 seedlings show different spatial expression patterns in darkness or light. Etiolated seedlings display high GUS activity in the upper hypocotyl, the hook region and the vascular tissue of the cotyledons, whereas in light-grown seedlings no activity was detected in the hypocotyl and almost all cells of the cotyledons express the GUS gene.

Regulation of hordein synthesis in barley high lysine mutant Notch-2.

Genomic DNA isolated from barley cv. NP 113 and its high lysine mutant Notch-2, and restricted with different restriction enzymes was hybridized with B1 and C-hordein DNA probes. Similar Southern hybridization patterns were observed between NP 113 and Notch-2. Dot blot hybridization analysis of RNA isolated at different developmental stages and from different tissues of seed showed temporal as well as tissue specific expression. The results obtained indicate that regulation at the level of transcription/post transcription may be responsible for lower accumulation of hordein in mutant Notch-2.

Cytochrome b-559 genes from Oenothera hookeri and Nicotiana tabacum show a remarkably high degree of conservation as compared to spinach. The enigma of cytochrome b-559: highly conserved genes and proteins but no known function.

Recent data suggest that cytochrome b-559, an intrinsic membrane protein of the oxygen-evolving photosystem II in chloroplasts, is a heme cross-linked heteromeric polypeptide unit (Herrmann et al. 1984, FEBS Lett 176:239-244). The genes for this cytochrome, designated psbE and psbF, have been located on the chloroplast chromosome of Oenothera hookeri and Nicotiana tabacum by hybridization with fragments of the corresponding spinach genes, and characterized. In both cases, the nucleotide sequence discloses 2 uninterrupted reading frames of 83 and 39 codons separated by a few nucleotides, as in spinach. The amber translation stop codon of psbE overlaps the putative ribosome-binding site for psbF in all cases. The predicted molecular weights of the proteins are 9.2 and 4.3 kd, respectively. In each of the three plant species, and the direction of transcription is opposite to that of the gene for cytochrome f which is located distal to the b-559 genes. Comparison of the deduced amino acid sequences with those from the corresponding spinach genes shows 97% homology. The ubiquitous presence and remarkably high degree of structural and functional conservation of this gene cluster supports the hypothesis of a heteromeric assembly and an important functional role for cytochrome b-559.

Plastocyanin is encoded by an uninterrupted nuclear gene in spinach.

Plastocyanin is a member of photosynthetic electron transport chains that transfers electrons from cytochrome f to the oxidized P700 chlorophyll a pigment of the photosystem I reaction center. We have isolated and characterized cDNA- and genomic clones from spinach (Spinacia oleracea) encoding the complete plastocyanin-precursor polypeptide. The amino acid sequence derived from the nucleotide sequence shows that the precursor consists of 168 amino acid residues including a transit sequence of 69 residues. The precursor polypeptide has a predicted Mr of 16,917, the mature protein of 10,413. The available data indicate that plastocyanin derives probably from a single-copy gene. The coding region contains no intron. The size of the mRNA as determined by S1 nuclease protection experiments is approximately 660 nucleotides, although analysis of different cDNA clones suggests that longer RNA species do exist, approaching the size of the mRNA (850 bases) estimated by Northern blot techniques.
