[Prophylaxis, diagnosis and surgical treatment of adhesions in the abdominal cavity of children].

The clinical and experimental study of postoperative complications of suppurative peritonitis (the pathologic commissure formation) has been done. The authors believe that the combination of the break of cytodynamic correlation in peritonitis centre and GIT paresis is a basis of pathogenesis of the process. The authors advocate combination of anticomissural preventive stern with cytomodulation and GIT paresis treatment. The abdominal ultrasonography has been used for the diagnosis. The main role in surgical treatment belongs to laparoscopy that made it possible to decrease the amount of relaparotomy cases. There were no lethal complications or cases of recurrency.

[Effect of nitrogen-containing compounds on the biosynthesis of avermectins].

The influence of complex nitrogen-containing substrates (Difko yeast extract, EKD nutrient yeast extract, soy bean flour and cotton seed meal), ammonium salts and some amino acids (alanine, methionine, valine, isoleucine and threonine) on the biosynthesis of avermectins in the cultures of two mutants of Streptomyces avermitilis was studied. It was shown that an excess of the nitrogen compounds in the fermentation medium induced a decrease in the antibiotic biosynthesis and the relative content of the group B avermectins.

[The current methods for the prevention, diagnosis and surgical treatment of the adhesive complications of peritonitis in children].

The paper is based on clinical and experimental findings of one of the postoperative complications of purulent peritonitis, namely adhesion formation. The authors consider that impaired cytodynamic relationship in the focus of peritonitis concurrent with gastrointestinal paresis underlie the pathogenesis of the above-mentioned complication. In this connection, the authors propose a set of anti-adhesive measures, which involves prodigiosane cytomodulation and differential treatment of enteroparesis. Intestinal ultrasonic echocardiography was used for diagnostic purposes. Laparoscopy was employed in the surgical treatment of adhesive complications of peritonitis, which allowed relaparotomy to be eliminated or its traumaticity to be reduced to a great extent. There were no deaths and relapses of adhesive disease.

[The use of highly purified Iletin-II-type porcine insulins and human insulin preparations in clinical practice].

The paper presents the results of treatment with insulins of iletin-II and humulin series. Decreased insulin doses, absence of complications, stable compensation of diabetes mellitus, reduction of insulin antibodies titer suggest high clinical effectiveness of the above preparations. Because of low immunogenicity humulin insulins are preferable when compared to iletin-II ones.

[Scanning electron microscopic method for studying the interaction of normal peritoneal macrophages with tumor cells].

It is shown that the damage action of normal peritoneal macrophages of mice includes inhibition of the proliferating activity of the mastocytoma P-815 cells and their complete destruction accompanied by the breaks of their cytoplasmic membrane. Scanning electron microscopy was used to reveal two types of contacts between peritoneal macrophages and cells P-815; the loss of microvilli and the appearance of blebs on cell surface in the case of adsorption of P-815 cells on macrophages; the contact through long filopodia which are sent forth by macrophages in the case when contacting cells are at some distance from each other.

[Immunomodulating and antitumor activity of polysaccharides of plant origin].

The effect of the polysaccharides tagetan, palustran, and zymosan injections to BALB/c mice on the formation of cytotoxic T-lymphocytes in allogenic mixed culture has been studied. Lymphocytes from a 5-day-old mixed culture were characterized by the greatest cytotoxic activity, with spleenocytes serving as reacting cells in mice injected 12-50 mg/kg of tagetan, 20 mg/kg of zymosan and 10-100 mg/kg of palustran, 7, 3 and 5 days, respectively, before the beginning of allogeneic stimulation in vitro. To determine correlation between immunostimulating and antitumor activity 25 mg/kg of tagetan every 14 days, 20 mg/kg of zymosan every 6 days, and 10 mg/kg of palustran every 6 days, were injected to BALB/c and BALB/c nu/nu mice prior to and following subcutaneous implantation of Crocker sarcoma cells. By the end of the first month of immunotherapy the tumour size in mice on tagetan or zymosan was 2-2,5 times smaller than in control animals. Tumour growth inhibition with palustran was about 30%. Polysaccharide administration to BALB/c nu/nu mice was not accompanied by tumour growth inhibition. The data obtained suggest that inhibition of Crocker sarcoma growth in mice injected the above polysaccharides is mediated by stimulation of antitumour T-cell activity.

[Immunomodulating activity of p-hydroxyphenyllactic and ascorbic acids].

p-Hydroxyphenyl lactic acid (PHA) in a concentration of 5 . 10(-5) M produced a significant inhibition of cell proliferation in response to alloantigens in a one-way mixed lymphocyte culture (MLC) in colonic cancer patients and in blast transformation in response to suboptimal doses of Con A. Multiple administration of ascorbic acid in an optimal concentration to the culture increased the proliferative response of lymphocytes to alloantigens and Con A. PHA and ascorbic acid did not exhibit any immunomodulating action during the use of healthy donors' lymphocytes or lymphocytes from colonic cancer patients, transformed with optimal mitogen doses. PHA did not affect the production of cytotoxic T lymphocytes in the MLC of the spleens of allogeneic mice but inhibited lymphocyte proliferation in response to alloantigens in the MLC of the spleens obtained from B6 and vitamin A deficient animals.

[Correction of biochemical and immunological indices in colonic cancer using optimal doses of retinyl acetate and ascorbic acid].

Blood plasma retinol level in normal donors and patients with colonic carcinoma was measured by high-pressure liquid chromatography and the concentration of p-hydroxyphenyl lactic and homogentizine acids by Gas Chromatograph Mass Spectrometer MAT-311A using 2H4-p-hydroxyphenylacetic acid as internal reference. The functional activity of lymphocytes was estimated from the proliferative response to alloantigens in a one-way mixed lymphocyte culture and in blast transformation reaction to Con A and pokeweed mitogen. After systematic intake of retinyl acetate and ascorbic acid in optimally high doses, the patients manifested an increase in vitamin C level in plasma and lymphocytes and a lowering of p-hydroxyphenyl lactic acid excretion. Blood plasma retinol remained unchanged. Daily intake of retinyl acetate and ascorbic acid for 8-12 days produced a significant increase of lymphocyte proliferation in response to alloantigens in mixed lymphocyte culture and blast transformation reaction to suboptimal mitogen doses.

[Effect of avitaminosis B6 in mice on the function of the T-lymphocytes in vitro].

The influence of different degrees of avitaminosis B6 in mice on the cytolytic activity of T-lymphocytes measured by the amount of Na2Cr51O4 released from the lysed target cells was studied on a model of primary immune response in a mixed lymphocyte culture in vitro. Keeping of the animals for 3 weeks on pyridoxine-free diet failed to influence the capacity of lymphocytes to proliferate in vitro and their cytolytic activity. In animals on pyridoxine-free diet for 45 days the amount of pyridoxal 5(1)-phosphate in the spleen decreased by 55% in comparison with control. Lymphocytes obtained from these animals and cultivated in vitro had a markedly decreased capacity to 3H-thymidine incorporation into DNA in response to alloantigen. The cytolytic activity of these lymphocytes also diminished. The capacity of various pyridoxine forms to restore T-lymphocyte functions disturbed by avitaminosis B6 was studied.

[Electron microscopic study of the interaction between cytolytic T-lymphocytes and target cells].

Thymocytes stimulated in vitro in a mixed culture was sorbed by centrifugation on the surface of target cells for electron microscope study of cytology of immune T-lymphocytes and early cytolysis periods. A well developed Golgi apparatus was revealed in the cytoplasm of lymphocytes; there was also accumulation of tubular structures 50 to 60 nm in diameter which communicated with the cysterns of the granular endoplasmic reticulum, with "descended" vesiculi and plasmatic membrane of lymphocyte. This membrane formed numerous contacts with the membrane of target cells thus producing closed clefts. Taking into consideration these data and also current views on the intertransformation of membranes and intracellular transport a hypothetic scheme of the mechanism involved in the cytolysis of the target cell by immune T-lymphocyte was put forward.

[Proliferation and cytotoxicity of thymus lymphocytes in vitro].

Proliferative and cytollytical activity of lymphocytes was compared in lymphocyte alloimmunization of the spleen and intact thymus. The count of live cells and DNA-synthesizing cells in the thymocyte monoculture was 10--15-fold, and in mixed thymus cell culture--about 5-fold lower than the corresponding amounts of spleen cells. The index of immune thymocyte stimulation was several times greater than that of the immune cells of the spleen. The cytotoxicity peak was observed on the 4th--5th day of stimulation when the cytolytic activity of the immune thymocytes approached the action of the immune cells of the spleen. Low DNA synthesis and a marked cytotoxic activity of immune thymocytes signified that stimulation of the thymus cells in vitro permitted to obtain cell population with a high content of cytolytic T-lymphocytes.
