Clustering of polarity reversals of the geomagnetic field.

Often in nature the temporal distribution of inhomogeneous stochastic point processes can be modeled as a realization of renewal Poisson processes with a variable rate. Here we investigate one of the classical examples, namely, the temporal distribution of polarity reversals of the geomagnetic field. In spite of the commonly used underlying hypothesis, we show that this process strongly departs from a Poisson statistics, the origin of this failure stemming from the presence of temporal clustering. We find that a Lévy statistics is able to reproduce paleomagnetic data, thus suggesting the presence of long-range correlations in the underlying dynamo process.

Periodic and aperiodic traveling pulses in population dynamics: an example from the occurrence of epidemic infections.

The dynamics of the occurrence of the dengue hemorrhagic fever in the 72 provinces of Thailand is investigated by performing a proper orthogonal decomposition (POD) on spatiotemporal data. Using this technique, we are able to identify and select the contribution of different modes, selected according to the energy content, to the evolution of the epidemic during 14 years. We found that the phenomenon is characterized by periodic cycles of yearly occurrence characterized by spatial scales of about 420 km. Superimposed on this basic mode, POD analysis is able to reveal the presence of high-energetic aperiodic traveling pulses of the epidemic, which extend spatially for about 510 km from Bangkok.

Proper orthogonal decomposition of solar photospheric motions.

The spatiotemporal dynamics of the solar photosphere is studied by performing a proper orthogonal decomposition (POD) of line of sight velocity fields computed from high resolution data coming from the MDI/SOHO instrument. Using this technique, we are able to identify and characterize the different dynamical regimes acting in the system. Low-frequency oscillations, with frequencies in the range 20-130 microHz, dominate the most energetic POD modes (excluding solar rotation), and are characterized by spatial patterns with typical scales of about 3 Mm. Patterns with larger typical scales of approximately 10 Mm, are associated to p-modes oscillations at frequencies of about 3000 microHz.

Roquefortine C occurrence in blue cheese.

Several strains of Penicillium are used for the production of mold-ripened cheeses, and some of them are able to produce mycotoxins. The aims of the research were the determination of roquefortine C and PR toxin in domestic and imported blue cheeses, the identification of the penicillia used as starter, and the investigation of their capacity for producing toxins in culture media. Roquefortine C was always found in the cheeses at levels ranging from 0.05 to 1.47 mg/kg, whereas the PR toxin was never found. The identification of the fungal strains present in the domestic cheeses included Penicillium glabrum, Penicillium roqueforti, and Penicillium cyclopium in the Gorgonzola "dolce" and Penicillium roqueforti in the Gorgonzola "naturale"; in one case, the presence of Penicillium crustosum was observed. The strains isolated from the foreign cheeses belonged to P. roqueforti. The strains were able to produce between 0.18 and 8.44 mg/liter of roquefortine in yeast extract sucrose medium and between 0.06 and 3.08 mg/liter and less than 0.05 mg/liter when inoculated in milk at 20 degrees C for 14 days and 4 degrees C for 24 days, respectively. Linear relations between production of roquefortine in culture media and cheeses did not emerge. PR toxin ranged from less than 0.05 to 60.30 mg/liter in yeast extract sucrose medium and was produced in milk at 20 degrees C from only one strain. The low levels and the relatively low toxicity of roquefortine make the consumption of blue cheese safe for the consumer.

Production of cyclopiazonic acid by molds isolated from Taleggio cheese.

Twenty-seven strains of Penicillium were isolated from the rind of Taleggio, a typical Italian cheese, so that we could test their capacity to produce cyclopiazonic acid (CPA); all strains produced CPA. The production was strongly influenced by the strain variety and growth conditions. Strains incubated at 25 degrees C for 7 days always produced CPA in mannitol broth, with concentrations ranging from 0.02 to 1 microg/ml, whereas only 33% of strains grown in yeast-extract broth produced CPA, with a maximum value of 0.1 microg/ml. In milk, maximum production (1.6 microg/ml) was observed after 14 days of incubation at 25 degrees C. In order to evaluate the presence of the toxin and its capacity for migrating into the cheeses, the rind, the cheese near the rind, and the cores from six Taleggio cheeses were analyzed. CPA was present in five cheeses, with a maximum concentration of 0.25 mg/kg in one rind, and in one cheese, the toxin migrated to the core. A positive correlation between CPA production and surface mold was found.

[Exercise two-dimensional echocardiography in recent myocardial infarction. Is it useful in the detection of multivessel disease?].

Exercise two-dimensional echocardiography (2D-ECHO) can be used to detect coronary artery disease in patients (pts) by the development of stress-induced transient asynergy in areas without wall motion abnormalities when at rest. The aim of the study is to verify the accuracy of exercise 2D-ECHO in the identification of high risk pts with multivessel disease after the first acute myocardial infarction (AMI). Technically adequate 2D-ECHO examinations were obtained in 21 of 28 (75%) consecutive patients after acute myocardial infarction. 30-50 days after acute myocardial infarction, these 21 pts (19 males and 2 females, mean age +/- SD = 54.3 +/- 8.7) underwent 2D-ECHO during bicycle exercise in supine position. The marker of multivessel disease was the development, during the stress test, of new areas of asynergy not adjacent to the infarcted area (i.e. transient remote asynergy). Two months after acute myocardial infarction all pts underwent coronary angiography to verify the severity of coronary obstruction (reduction of luminal diameter greater than or equal to 75% in the non infarct related vessel).(ABSTRACT TRUNCATED AT 250 WORDS)

[Echinococcal cyst of the right ventricle: diagnostic role of 2-dimensional echocardiography].

A case of cardiac echinococcosis in a 22 year-old woman is reported. Clinical findings and cardiac catheterization data offered only presumptive evidence of an intraventricular mass but failed to make a positive diagnosis of cardiac involvement by hydatid disease. Using Two-Dimensional Echocardiography (2-DE) we were able to identify a rounded structure with multiple loculation into the right ventricle, highly suggestive of a hydatid cyst. Surgery confirmed our findings. We confirm the ability of 2-DE to detect and characterise intracardiac masses and we suggest that 2-DE could be considered the procedure of choice in the diagnosis of cardiac hydatid disease.

Current status of activity assays for tissue plasminogen activator.

Tissue plasminogen activator (TPA) is a serine protease involved in the fibrinolytic system that dissolves blood clots. The enzyme catalyzes the conversion of a zymogen, plasminogen, to the enzymatically active form, plasmin, by limited proteolysis. In the course of searching for specific activity assays that might be useful in monitoring the purification of TPA, we have developed several coupled photometric assays. In addition, radioactive, agarose-plate, and other activity assays have also been considered and investigated for this purpose (Table 1). We have previously reported the one-step photometric procedure consisting of a thioester, thiobenzyl benzyloxycarbonyl-lysine (Z-Lys-S-Bzl) and fibrinogen-coated plates. It is simpler and more sensitive than the old two-step method without using immobilized fibrinogen. The new assay has been used successfully for protein purification and can be easily adapted to automated processes. Recently, other chromogenic substrates, D-Val-L-Leu-L-Lys-p-nitroanilide (Val-Leu-Lys-pNA) and D-Ile-L-Pro-L-Arg-p-nitroanilide (Ile-Pro-Arg-pNA) were also used in the one-step assay. It is found that TPA activity is greatly enhanced by immobilized fibrinogen and free fibrinogen when either the thioester. Val-Leu-Lys-pNA, or Ile-Pro-Arg-pNA were used in the colorimetric assay (Fig. 1, A-D). Enzyme kinetics studies indicate that the Km for plasminogen assayed on the thioester and fibrinogen-coated plates is 1.5 micrograms per ml, which is substantially lower than that observed in untreated plates (4.8 micrograms per ml). This is not due to the effect of fibrinogen on the second step of the coupled photometric assay because there is no change in the plasmin activity under these conditions (Fig. 1E). Similar results in TPA activation have also been observed, when fibrin-coated plates were used. Free fibrinogen, which is an activator of TPA, has been included in the standard assay mixture. We are able to detect less than 1 ng of TPA activity within a one-hour incubation time at 20 degrees C (Fig. 2A). In the thioester assay, however, high concentrations of reducing agents and nonspecific proteins cause significant background due to the interaction of DTNB with these reagents. 125I-labeled fibrin-coated plates had been extensively used in the past for urokinase and TPA assays. Although the sensitivity of the radioactive procedure is equivalent to that of the thioester photometric method, it appears that the kinetics of the enzyme are not easy to follow nor is the reproducibility great.(ABSTRACT TRUNCATED AT 400 WORDS)

Desmopressin and imipramine in the management of nocturnal enuresis: a multicentre study.

The efficacy and safety of desmopressin (Minirin/DDAVP) treatment compared with imipramine were investigated in a multicentre, open, cross-over design in 57 patients, aged 6-15 years, affected by nocturnal enuresis to establish the best therapeutic approach to this condition. After a two-weeks observation and control period, patients were randomised to one of two groups: intranasal administration of desmopressin, 30 micrograms/day for three weeks, followed by imipramine, 0.9 mg/kg for a further three weeks, or imipramine 0.9 mg/kg for three weeks, followed by desmopressin, 30 micrograms/day for a further three weeks. Following treatment, all patients were observed for a further two weeks. Administration of either treatment protocol resulted in a statistically significant decline in the number of enuretic episodes per week compared to the control. The greater antidiuretic effect observed in the group receiving imipramine followed by desmopressin suggests the two compounds have different profiles. Also, when the treatment period was compared with the follow-up, the antidiuretic effect had a longer duration in the group initially given imipramine. No further improvement was seen when desmopressin was administered first, with a mild worsening of the effect sometimes occurring, suggesting a different carry-over effect between the two treatments. This suggests that desmopressin offers a better approach to the management of nocturnal enuresis.
