Activity of antioxidant enzymes in children from families at high risk of premature coronary heart disease.

A positive family history of coronary heart disease (CHD) is one of the most predictive risk factors of CHD. Many children with increased risk of CHD because of their positive family history of CHD do not present other risk factors, such as altered serum lipid profile. Oxidative stress plays an important part in the pathogenesis of atherosclerosis. Serum antioxidants and intracellular enzymatic antioxidants composed mainly of glutathione peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD) and glutathione reductase counterbalance oxidative stress. Diminished activity of this system may lead to accelerated progression of atherosclerosis. The aim of this study was to assess the activity of CAT, GSH-Px, SOD and glutathione reductase in children with a family history of premature CHD who did not present any other major risk factors of CHD (diabetes, obesity, dyslipidaemia or hypertension). Twenty-two healthy children from high-risk families, selected according to the National Cholesterol Education Program definition, were enrolled in the study. The control group comprised 18 children without a family history of CHD. All the children were healthy and had been screened for hyperlipidaemia, diabetes, hypertension and obesity prior to the study. The erythrocyte activity of CAT, GSH-Px, SOD and glutathione reductase was assessed. Children at high risk of CHD had a statistically significant lower level of GSH-Px and CAT activity than the children in the control group. There were no statistically significant differences in the activity of SOD and glutathione reductase.

Characterisation of epithelial cell line from rat cornea.

PURPOSE: Characterisation of RtCE-1 cells, newly established rat corneal epithelial cell line. METHODS: Morphology of RtCE-1 cells was characterised by light and electron microscopy. Expression of cytokeratins was studied by polyacrylamide electrophoresis and Western blotting. Characterisation of RtCE-1 cells also included karyotype analysis and in vitro study of growth kinetics. RESULTS: The line shows morphological similarities to normal corneal epithelium. The cells express cornea-specific cytokeratins. Karyotype analysis revealed that the cells are polyploid with modal number of chromosomes at passage 30 equalling 72 +/- 14. Growth of the line is partially dependent on EGF. CONCLUSION: RtCE-1 cells may constitute a model for the evaluation of proliferation, function and differentiation of corneal epithelium in vitro.

Lead levels in body fluids of workers of an automobile factory with clinically diagnosed arterial hypertension.

The aim of the study is the evaluation of lead levels in body fluids in a chosen study group with diagnosed arterial hypertension employed in a car factory. The study was carried out on workers diagnosed with various stages of arterial hypertension who had been subjected to long-term lead exposure at different workplaces. The examination of the patients included medical history, physical examination and biochemical tests of blood and urine, following routine in the laboratory diagnosis in order to assess medical condition in terms of subclinical symptoms of lead exposure. Mean values of the lead level in blood and urine of studied population are insignificantly higher than in the control group. The obtained results seem insufficient to confirm the effect of occupational exposure to lead in subthreshold doses on the development of arterial hypertension, and are within the recommended hygienic standards.

The evaluation of noise level in different points of urban area.

The aim of the study was measuring noise intensity in the selected points of traffic routes in the city of Lublin. Investigations were carried out at 11 points located in the centre of the city and at escape roads. Noise value levels showed variable intensity, the highest being observed in the morning and afternoon hours. The highest mean level of noise intensity was recorded at the crossing of Solidarności Avenue and Sikorskiego Street, and the lowest in Narutowicza Street.

The evaluation of intensity of traffic noise in different points of town routes.

The aim of the study was measuring noise intensity in the selected points of traffic routes in the town of Lublin. Investigations were carried out at 11 points located in the centre of the town and at escape roads. Noise value levels showed variable intensity, the highest being observed in the morning and afternoon hours. Effect of Tn10/Tn5 transposons on the survival and mutation frequency of halogen light-irradiated AB1157 Escherichia coli K-12.

We show here that the Tn10/Tn5 transposon when inserted into the chromosome of strain AB1157 makes the bacteria more sensitive to and less mutable by halogen light irradiation. These effects are most probably caused by depletion of UmuD and UmuC proteins since: (i) transformation of the transposon-bearing bacteria with plasmids harbouring umuD'C (or umuDC) leads to recovery of the original survival and mutation frequencies; (ii) insertion of Tn10/Tn5 into chromosomal DNA has no effect on the level of mutation induced by ethyl methane-sulphonate treatment, a mutagen whose activity is umuDC-independent; (iii) the decline in survival is in about the same range for Tn10-bearing bacteria as for bacteria with deleted umuDC. However, whereas transformation of bacteria deleted in umuDC with plasmids carrying umuD'C/umuDC leads to full recovery of halogen light-induced mutability, recovery of survival is poor. This suggests that the mechanisms leading to umuDC-dependent mutagenesis and umuDC-dependent protection of cell survival are different. None of these effects occurs in bacteria bearing the Tn9 transposon in their DNA.

Preferential segregation of marker chromosomes 14 and 18 in mouse recombinant inbred strains derived from the KE and CBA/Kw strains.

The segregation pattern of chromosomes 14 and 18 were analyzed in recombinant inbred strains of mice developed from KE and CBA/Kw strains. The analysis was possible owing to the fact that the C-band on chromosomes 14 of the CBA/Kw strain and that of chromosome 18 of the KE strain show size polymorphism: while the CBA/Kw mice have a small sized C-band on chromosome 14, the KE mice show small C-bands on chromosome 18. Chromosomes were identified by G-banding and FISH. The results show that the chromosomes with small centromeric chromatin segregate preferentially.

Mutation induction and mutation frequency decline in halogen light-irradiated Escherichia coli K-12 AB1157 strains.

The effects of halogen light irradiation on reversion of argE3-->Arg+ in E. coli K12 strain AB1157 and its mfd- mutant, and on mutation frequency decline (MFD) after transiently incubating irradiated bacteria under non-growing conditions were studied. The induction of mutations, the mutational specificity, and the MFD effect had the same characteristic features as those seen in E. coli B strains after irradiation with 254 nm UV light. MFD which is due to repair of premutagenic lesion in the transcribed strand of glnU gene and prevents mutations leading to supB formation, was not observed in halogen light-induced mutations in the mfd-1 strain. Overproduction of UmuD'C proteins led to a large increase in mutation frequency, which was much greater in mfd- than in mfd+ strains. In bacteria irradiated with halogen light and incubated immediately in a rich medium to express mutations, the formation of supB predominated strongly over that of supE(ochre) in mfd- cells but was at a similar level in mfd+ cells. Introduction of zcf117::Tn10 to AB1157 strain makes cells more sensitive to halogen light irradiation, whereas introduction of mfd-1 does not.

Conformation of plasmid DNA from Escherichia coli deficient in the repair systems protecting DNA from 8-oxyguanine lesions.

8-Oxyguanine (8ohG) is a major oxidation product of guanine and a biomarker of oxidative stress in mammal. We have attempted to estimate the level of 8ohG residues in plasmid DNA (pGW2123 and pBR322) grown in various bacterial strains (fpg, mutY, or mutT, plus mutT fpg and mutT mutY double mutants) differing in the system protecting cells against the mutagenic effects of 8ohG in DNA. The method was based on digestion of plasmid DNA with Fpg, and agarose gel electrophoresis. Fpg converts pDNA from covalently closed circular to the open circular (ccc-->oc) form of pDNA when there is at least one 8ohG, or apurinic site, per ccc pDNA molecule. It was found that: i) the content of 8ohG in pDNA grown in any of the tested bacteria is below one 8ohG per 10(4) base pairs; ii) a substantial part of pGW2123 is isolated from the bacteria in the oc form; iii) the ratio of oc/ccc in pGW2123 depends on the bacterial host and is the lowest when the plasmid was harvested from mutY- deficient cells; iv) pBR322, unlike pGW2123, is isolated predominantly in the ccc form; and v) of the pBR322 grown in the tested bacteria apparently the most resistant to Fpg digestion was pBR322 grown in the mutY strain. It is proposed that this reflects the compact structure of pDNAs when they are grown in bacteria deficient in mutY gene product.

[A study of erythrocyte oxidative enzyme activity in patients with laryngeal cancer before and after treatment with a selenium preparation CEROSEL].

The preliminary results of activity changes of antioxidant enzymes (glutathione peroxidase, catalase and superoxide dismutase) in erythrocyte in 19 patients with laryngeal cancer were presented. Enzymes activity in patients was signified three times: before surgery, before and after radiotherapy. During Co-60-therapy patients received leaven preparation of selenium CE-ROSEL in dose 100 micrograms Se/day. Obtained results showed increase of enzymes activity after CE-ROSEL supplementation. This changes are probably the result of selenium influence on protected mechanism before the free radicals.

Proteolysis inhibitors from Ascaris lumbricoides: fact or fiction?

The results of our own 25 years investigations on biological activity of proteolytic inhibitors of Ascaris lumbricoides with the help of various experimental models were discussed. As it results from these studies alpha-chymotrypsin and trypsin inhibitors from A. lumbricoides are not fiction but are substances with significant biological activity, which may be important in regulation of the host-parasite system as well as parasite pathogenic factor.
