Flow cytometric analysis of high grade B-cell non-Hodgkin's lymphoma: correlation between histology, immunophenotyping and cell cycle analysis.

The immunophenotype and the DNA parameters of 21 high grade B-cell non-Hodgkin's lymphomas considered in terms of the Kiel Classification were studied by flow cytometry. The centroblastic and immunoblastic lymphomas differed in the expression of lymphocyte markers, however no characteristic immunophenotype for each lymphoma type could be distinguished. Assessment of the FSC/SSC dot plots with histology enables to distinguish subpopulations of the lymphoma cells. The immunoblastic and centroblastic lymphomas were characterized by high proliferative activity although no significant differences in S-phase values between both groups could be found.

Flow cytometric analysis of immunophenotypic and DNA values in non-Hodgkin's B-cell lymphomas classified according to the Kiel Classification and Working Formulation.

B-cell non-Hodgkin's lymphomas classified previously according to the Kiel Classification were reclassified according to the Working Formulation. The comparative analysis of flow cytometric data was done for these two classification systems. It was established that flow cytometric data describe defined groups of malignancy more precisely in the Working Formulation, especially in case of the low grade lymphomas.

Ultrastructure of diffuse malignant mesothelioma of the pleura. An analysis of ten cases.

An electron-microscopic examination of ten cases of malignant pleural mesothelioma is presented. The most characteristic features for diagnosis are: presence of microvilli and desmosomes, abundant intermediate filaments and direct contact between microvilli and collagen fibres. Microvilli were present not only on the luminal surface of the cells, but also on the abluminal surfaces. Differentiation towards the cells characteristic for rhabdomyosarcoma was found in one case.

Alveolar soft part sarcoma: an analysis of 8 cases. Review of the literature.

Eight cases of alveolar soft part carcinoma (ASPS) are presented. The average age of the patients was 28 years. There were 6 women and 2 men. The left sided and right sided tumours were equal in number. Local recurrence was not observed in any of the patient treated, but lung metastases occurred in 6 of them. Three patients died with disease the average 35 months after treatment. Histologically in one case the uniform sheets of cells were observed without alveolar arrangement. The presence of beta glucuronidase (BG), acid phosphatase (AP) and non specific esterase (NE) in tumor cells was seen in one case studied. Immunohistochemical and ultrastructural examination confirm the thesis, that ASPS consist a distinct clinico-morphologic entity, the histogenesis of which remains uncertain.

Mesenchymoma malignum of the buttock.

A case of a soft-tissue tumor of the left gluteal region in a 33-year-old woman is presented. Immunohistochemical study and electron microscopic examination confirmed multidirectional differentiation of the tumor. The patient died 9 months after surgery, despite complementary multimodal therapy. No autopsy was performed.

[Evaluation of hearing in patients with diabetes].

Hearing in 60 diabetics aged 20-70 years were studied by means of tonal, verbal and B'ekèsy audiometry, SISI tests, Fowler tests, stapedius reflex and tone decay tests. The material was grouped according to the diabetes type, chronicity of disease, age, sex and presence of retino- and polyneuropathy. The results proved that diabetes may cause the progressive form of hearing loss which depend of chronicity of disease and age of patients. Sometimes the hypo-acuity may be found in the recent form of diabetes, rarely as sudden deafness. The hearing impairment is more frequent among the retino- and polyneuropathy patients. The localisation of lesions may be in all part of hearing pathways from the labyrinth to cortical areas.

A study of the thermic responses to a meal and to a sympathomimetic drug (ephedrine) in relation to energy balance in man.

1. Sixteen adult male volunteers were selected on the basis of body size and customary food intake: half could be described as "lean' and habitually consuming large amounts of food (group mean +/- SEM: 15.03 +/- 1.13 MJ/d), the high-energy-intake group (HEI group), and half though "lean' admitted to a weight problem and regularly consumed a lower than average food intake (group mean +/- SEM: 6.90 +/- 0.39 MJ/d), the low-energy-intake group (LEI group). 2. Energy expenditure was measured by open-circuit indirect calorimetry. Resting metabolic rate (RMR) was recorded. A meal (Complan, either 2.1 MJ or 4.2 MJ), ephedrine hydrochloride (0.25 mg and 0.50 mg/kg body-weight) or a water control were then administered and metabolic rate (MR) was measured for 4 h. Blood was collected before and 1 h after the meal or drug, and the serum analysed for various hormones and blood metabolites. 3. The size of the thermic response to feeding but not the time-course was related to meal size in both groups. MR increased by 21.6 and 28.6% in the HEI group and by 8.2 and 20.0% in the LEI group in response to the 2.1 and 4.2 MJ Complan meals respectively. Fasting insulin levels were similar in both groups but showed a significantly higher level in the LEI than HEI group after the Complan meals. 4. The mean RMR increased by 5.2 and 10.3% in the LEI in response to ephedrine and by 15.7 and 11.2% in the HEI groups after 0.25 mg and 0.50 mg ephedrine/kg respectively. The rise in serum-free fatty acids in response to ephedrine was significantly higher in the HEI group than in the LEI group. 5. These results suggest (1) the meal size required to promote a maximum thermic effect is smaller in energetically-inefficient individuals (2) the sensitivity to a sympathomimetic drug is also increased in energetically-inefficient individuals. 6. We conclude that in energetically-efficient individuals both the thermic response to a meal and the sympathetic-mediated thermogenesis are lower than in energetically-inefficient ones.

Calmodulin antagonist reduces release of malondialdehyde from isolated ischemic/reperfused rat heart.

To investigate the role of Ca-calmodulin complex in the development of myocardial injury caused by ischemia and reperfusion, an isolated working rat hearts were subjected to ischemia/reperfusion with and without calmodulin antagonist, trifluoperazine (TFP 5 x 10(-7)M) in the perfusion medium. In TFP-treated hearts postischemic recovery of hemodynamic function was markedly improved and the release of LDH and malondialdehyde (MDA) significantly reduced as compared with control hearts. It is concluded that: 1) calmodulin-dependent mechanism seems to be involved in peroxidative injury of cellular membranes, 2) cardioprotective effect of TFP results at least in part from prevention of membrane damage caused by lipid peroxidation.
