Myristylation of pp60c-src is not required for complex formation with polyomavirus middle-T antigen.

Middle-T antigen (middle-T), the transforming gene product of polyomavirus, associates with several cellular tyrosine kinases, such as pp60c-src. Complex formation leads to kinase activation and is essential for cell transformation. Middle-T-associated as well as uncomplexed pp60c-src is predominantly found in the plasma membrane. We transfected mouse 3T3 fibroblasts with a mutated c-src gene (2Ac-src), allowing the expression of a protein containing alanine instead of glycine in position 2 of the primary translation product. Contrary to the wild-type c-src gene product, pp60c-src(2A) was not myristylated and accumulated in the cytoplasm instead of being transferred to cellular membranes. The mutant protein was able to associate with middle-T and was activated similarly to the wild-type c-src gene product. Both wild-type and 2A mutant protein were membrane associated upon complex formation with middle-T. This finding suggests that the putative carboxy-terminal membrane anchor sequence of middle-T is sufficient to hold middle-T-associated pp60c-src(2A) in the plasma membrane.

Cigarette smoke components are not very effective in directly inactivating alpha 1-proteinase inhibitor.

Cigarette smoke was found to be rather ineffective in inactivating alpha 1-proteinase inhibitor (alpha 1-PI) in aqueous solution, whereas a slow inactivation of alpha 1-PI by a dimethyl sulfoxide extract of whole cigarette smoke condensate was observed. However, this inactivation could only partially be prevented by antioxidants indicating that it is not, or at least not exclusively, due to oxidation. The bulk of inactive alpha 1-PI found in lung lavage fluids from smokers is thus probably generated through endogenous mechanisms and not through smoke components directly.

[Several years' experience with thumb saddle joint prosthesis by the de la Caffinière method].

This type of endoprosthesis was used for four years in chronically painful idiopathic osteoarthritis of the carpometacarpal joint of the thumb. Our results match exactly those published by the originator of the endoprosthesis CAFFINIERE, i.e. 70% excellent results, nearly 30% problem cases. The problem of loosening of the stem or the cup of the prosthesis seems to depend in our cases on the operative technique rather than on the design or other factors. In the rheumatoid patient as well as in polyarthrotic hands we cannot encourage the prosthetic replacement of the carpometacarpal joint of the thumb.

Plate osteosynthesis of femoral shaft fractures in adults. A follow-up study.

In 67 diaphyseal femoral fractures in adults treated by plating, the average follow-up time was 5 years. Sixty per cent of the patients suffered multiple injuries; 69% of the fractures were comminuted. The goal of treatment was rigid internal fixation. However, bridging of the comminuted area with a plate was sometimes necessary to maintain length as well as to encourage reconstitution and revascularization of fragments. Almost all patients had satisfactory end results. Only 2 cases were rated poor. Patients with rigid interfragmentary fixation had few complications, while patients with obvious instability (14 cases) developed 2/3 of the complications. The infection rate was 3%. Some complications might have been avoided by early cancellous bone grafting. The study confirms the need for primary or early secondary bone grafting to ensure bony union if cortical defects, potentially unstable fixation, or devascularized fragments are present.
