15-Deoxy-Delta12,14-prostaglandin J2 enhances docetaxel anti-tumor activity against A549 and H460 non-small-cell lung cancer cell lines and xenograft tumors.

15-Deoxy-Delta-prostaglandin J2 is a naturally occurring endogenous ligand for peroxisome proliferator-activated receptor-gamma. The current study was aimed to determine the mechanism of anti-proliferative effect of 15-deoxy-Delta-prostaglandin J2+docetaxel against A549 and H460 non-small-cell lung cancer cell lines and xenograft tumors. In-vitro cytotoxicity of 15-deoxy-Delta-prostaglandin J2 alone and in combination with docetaxel was studied against A549 and H460 cell lines. For in-vivo studies, female athymic nu/nu mice were xenografted with A549 and H460 tumors and treated with 15-deoxy-Delta-prostaglandin J2 (1 mg/kg/day; intraperitoneal), docetaxel (10 mg/kg; intravenous on days 14, 18 and 22) and 15-deoxy-Delta-prostaglandin J2+docetaxel. Apoptosis was measured in A549 cells and tumor tissues, following various treatments. Peroxisome proliferator-activated receptor-gamma, caspases, Bcl2 and p53 family proteins or their mRNA expressions were measured by Western blotting, reverse transcription-polymerase chain reaction and real-time polymerase chain reaction in A549 tumors. A possible role of a peroxisome proliferator-activated receptor-gamma-independent mechanism was studied in A549 cells treated with peroxisome proliferator-activated receptor-gamma antagonist, GW9662. Isobolographic analysis demonstrated synergistic interaction (combination index <1.0) between 15-deoxy-Delta-prostaglandin J2 and docetaxel against A549 and H460 cells in vitro. 15-Deoxy-Delta-prostaglandin J2+docetaxel significantly reduced the tumor volume compared with control (P<0.05), 15-deoxy-Delta-prostaglandin J2 (P<0.05) and docetaxel (P<0.05, P<0.01) in both A549 and H460 tumors. 15-Deoxy-Delta-prostaglandin J2+docetaxel showed a significant increase in apoptosis associated with inhibition of the Bcl2 and cyclin D1 expression and overexpression of caspase and p53 pathway genes. Further, enhanced expression of caspase 3 and inhibition of cyclin D1 by 15-deoxy-Delta-prostaglandin J2+docetaxel was not reversed by GW9662, thus suggesting a possible peroxisome proliferator-activated receptor-gamma-independent mechanism. In conclusion, 15-deoxy-Delta-prostaglandin J2 enhanced the anti-tumor action of docetaxel by peroxisome proliferator-activated receptor-gamma-dependent and -independent mechanisms mediated by induction of apoptosis.

Evaluation of metered dose inhaler (MDI) formulations of ciclosporin.

Our purpose was to evaluate metered dose inhaler (MDI) formulations of cyclosporin (cyclosporine) for aerodynamic properties, chemical stability and bioactivity. Ciclosporin formulations (0.1, 0.5 and 1.0% w/w) were prepared in hydrofluoroalkane (HFA) propellants (134a and 227) containing 3 and 6% ethanol. Aerodynamic properties of the MDI formulations were analysed using an eight-stage Andersen cascade impactor and respirable mass and non-respirable mass, mass median aerodynamic diameter (MMAD) and geometric standard deviation (GSD) were determined from the impaction profiles. The chemical stability of 0.1% ciclosporin in HFA 227 containing 3% ethanol formulation stored at room temperature and 40 degrees C was evaluated by HPLC at 0, 14, 30 and 90 days. The bioactivity of ciclosporin MDI formulations was evaluated by determining the ciclosporin-mediated inhibition of interleukin-2 (IL-2) release from human Jurkat cells stimulated with phorbol 12-myristate 13-acetate (PMA). As ethanol concentration increased from 3 to 6%, respirable mass decreased from 2.3 mg per five actuations to 0.04 mg per five actuations for HFA 227 formulations, and from 1.5 mg to 0.09 mg per five actuations for HFA 134a formulations. The MMAD for both HFA 134a and 227 formulations increased with an increase in ciclosporin concentration. HPLC analysis showed ciclosporin to be extremely stable in HFA 227 at room temperature and 40 degrees C. Stimulation of Jurkat cells with PMA released significant amounts of IL-2, which was inhibited by ciclosporin in a dose-dependent manner. This study shows the feasibility of developing chemically stable and bioactive HFA-based MDI formulations of ciclosporin.

Molecular cloning and sequence analysis of the cDNA encoding thyroid-stimulating hormone beta-subunit of common duck and mule duck pituitaries: in vitro regulation of steady-state TSHbeta mRNA level.

For better understanding of phylogenetic diversity and evolution of pituitary thyroid-stimulating hormone (TSH) in birds, we have cloned the cDNAs encoding TSH beta subunit (TSHbeta), by reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE) from two species of domestic ducks, common duck (Tsaiya duck and Pekin duck) (Anas platyrhynchos domesticus) and mule duck (hybrid of male muscovy duck Cairina moschata and female A. platyrhynchos domesticus). The nucleotide sequences of isolated TSHbeta cDNAs of the two species of ducks are identical, with each including 66 bp of 5'-untranslated region (UTR), 402 bp of coding region, and 82 bp 3'-UTR followed by 18 bp poly A tract. The deduced TSHbeta subunit of the ducks contains 134 amino acids consisting of a putative signal peptide of 19 amino acids and a putative mature protein of 115 amino acids. However, the TSHbetas of common duck and mule duck differ from the TSHbeta of muscovy duck in one amino acid at position 97 of the mature protein: isoleucine for common duck and mule duck, and valine for muscovy duck. Our findings thus demonstrate that inter-genus variation of TSHbeta exists in Family Anatidae, and that TSHbeta gene in the mule duck is preferentially transcribed from the maternal genome rather than from the paternal genome. TSHbeta mRNA expressions were investigated by culturing common duck pituitaries with various doses of hormones. Thyrotropin-releasing hormone (TRH) stimulated, while thyroid hormones, triiodothyronine (T(3)) and thyroxine (T(4)), inhibited the TSHbeta mRNA levels, in dose-related manners. The findings thus support that the mode of regulation of TSH gene expression in hypothalamo-pituitary-thyroid axis in birds is similar to that in mammals. Cortisol and corticosterone decreased the steady-state TSHbeta mRNA levels at the pituitary level, in a dose-related manner, the first-time demonstration in vertebrates. The results may suggest that glucocorticoids exert suppressive action directly at pituitary level in modulation of steady-state TSHbeta mRNA level.

In vitro and in vivo comparison of dermal irritancy of jet fuel exposure using EpiDerm (EPI-200) cultured human skin and hairless rats.

The purpose of this study was to evaluate an in vitro EpiDerm human skin model (EPI-200) to study the irritation potential of jet fuels (JP-8 and JP-8+100). Parallel in vivo studies on hairless rats on the dermal irritancy of jet fuels were also conducted. Cytokines are an important part of an irritation and inflammatory cascade, which are expressed in upon dermal exposures of irritant chemicals even when there are no obvious visible marks of irritation on the skin. We have chosen two primary cytokines (IL-1alpha and TNF-1alpha) as markers of irritation response of jet fuels. Initially, the EPI-200 was treated with different quantities of JP-8 and JP-8+100 to determine quantities which did not cause significant cytotoxicity, as monitored using the MTT assay and paraffin embedded histological cross-sections. Volumes of 2.5-50 microl/tissue (approximately 4.0-78 microl/cm2) of JP-8 and JP-8+100 showed a dose dependent loss of tissue viability and morphological alterations of the tissue. At a quantity of 1.25 microl/tissue (approximately 2.0 microl/cm2), no significant change in tissue viability or morphology was observed for exposure time extending to 48 h. Nonetheless, this dose induced significant increase in IL-1alpha and TNF-alpha release versus non-treated controls after 24 and 48 h. In addition, IL-1alpha release for JP-8+100 was significantly higher than that observed for JP-8, but TNF-alpha release after 48 h exposure to these two jet fuels was the same. These findings parallel in vivo studies on hairless rats, which indicated higher irritation levels due to JP-8+100 versus JP-8. In vivo, transepidermal water loss (TEWL) and IL-1alpha expression levels followed the order JP-8+100 > JP-8 > control. Further, in vivo TNF-alpha levels for JP-8 and JP-8+100 were also elevated but not significantly different from one another. In aggregate, these findings indicate that EPI-200 tissue model can be utilized as an alternative to the use of animals in evaluating dermal irritation.

Inhalation delivery and anti-tumor activity of celecoxib in human orthotopic non-small cell lung cancer xenograft model.

PURPOSE: To determine the in vivo anti-tumor effect of aerosolized Celecoxib (Cxb) in combination with i.v Docetaxel (Doc) and compare the anti-tumor effect with oral Cxb combined with i.v Doc in human orthotopic non-small cell lung cancer (NSCLC) xenograft model. MATERIALS AND METHODS: Female Nu/Nu mice were implanted with orthotopic tumors by injecting A549 cells into the lung parenchyma. Seven day after tumor implantation the mice were treated with aerosolized Cxb (30 min exposure/day, 5 mg/ml solution) + i.v Doc (10 mg/kg) and the effect was compared with oral Cxb (150 mg/kg/day) + i.v Doc (10 mg/kg), for 28 days. Small-animal nose only inhalation chamber (CH Technologies, Westwood, NJ) was utilized for aerosol exposure. Therapeutic activity of Cxb (aerosol/oral) + Doc was estimated by differences in lung weight, tumor area and animal body weight. Lung tumor samples isolated from mice were analyzed for (a) PGE2 levels by enzyme immunoassay (EIA) (b) expression of Fas and Factor VIII by immunohistochemistry (c) IL-8 expression using EIA kits and (d) mRNA expression for caspase-3 by Real-Time PCR. RESULTS: Mice treated with Cxb (aerosol/oral) + Doc showed significant reduction (P < 0.001) in lung weight and tumor area as compared to Cxb or Doc treatments. Cxb (aerosol/oral) + Doc showed increased apoptosis mediated via increased Fas and caspase-3 (P < 0.001) expression as compared to untreated control. Further, the combination treatment showed antiangiogenic effect as demonstrated by reduced expression of Factor VIII, IL-8 (P < 0.001) and PGE2 (P < 0.001) in lung tumors as compared to untreated control. Aerosolized Cxb at a significantly lower therapeutic dose (4.56 mg/kg/day) demonstrated comparable anti-tumor efficacy to orally administered Cxb (150 mg/kg/day). CONCLUSION: Cxb was formulated and effectively delivered via aerosolization to treat orthotopic lung tumors in combination with i.v Doc. Cxb when administered by aerosol produced same therapeutic effect as oral Cxb, but at lower therapeutic dose and thus shows promise for the treatment of lung cancer.

Multiscale spatial Monte Carlo simulations: multigriding, computational singular perturbation, and hierarchical stochastic closures.

Monte Carlo (MC) simulation of most spatially distributed systems is plagued by several problems, namely, execution of one process at a time, large separation of time scales of various processes, and large length scales. Recently, a coarse-grained Monte Carlo (CGMC) method was introduced that can capture large length scales at reasonable computational times. An inherent assumption in this CGMC method revolves around a mean-field closure invoked in each coarse cell that is inaccurate for short-ranged interactions. Two new approaches are explored to improve upon this closure. The first employs the local quasichemical approximation, which is applicable to first nearest-neighbor interactions. The second, termed multiscale CGMC method, employs singular perturbation ideas on multiple grids to capture the entire cluster probability distribution function via short microscopic MC simulations on small, fine-grid lattices by taking advantage of the time scale separation of multiple processes. Computational strategies for coupling the fast process at small length scales (fine grid) with the slow processes at large length scales (coarse grid) are discussed. Finally, the binomial tau-leap method is combined with the multiscale CGMC method to execute multiple processes over the entire lattice and provide additional computational acceleration. Numerical simulations demonstrate that in the presence of fast diffusion and slow adsorption and desorption processes the two new approaches provide more accurate solutions in comparison to the previously introduced CGMC method.

A reactivity index study to monitor the role of solvation on the interaction of the chromophores with amino-functional silanol surface for colorimetric sensors.

Amino-functional silanol surface are mostly used for the immobilization of inorganic ions, molecules, organic or biochemical molecules onto the mesopore surface. In analytical chemistry, the metal ion uptake was visualized through colorimetric sensors using chromophore molecules. One needs to know the structure-property correlation between the chromophore and silylating agent while choosing chromophore, which is very important to design the sensors. We have used two chromophores representative of hydrophobic and hydrophilic type. We used density functional calculation on all the interacting molecules in both the unsolvated phase and solvated medium within the domain of hard soft acid base principle (HSAB) to look at the localized activity of the interacting atoms of these reacting molecules to formulate a priori rule to choose of the best chromophore. We have as well postulated the mechanism of interaction between chromophore and the silylating agent. The results were compared with experiment and it is observed that solvation plays a detrimental role in the binding of chromophore with silylating agent. The results also show that, the range of reactivity index can be used as a suitable property to scale activity of chromophore molecules suitable for the sensing process. It is observed that the hydrophobic chromophore binds stronger with both the metal and the silylating agent; whereas for the hydrophilic one, it binds only with the silylating agent when solvated and in all cases the metal ion binding is weaker compared to that of the hydrophobic one.

Enhancement of antitumor activity of docetaxel by celecoxib in lung tumors.

Our study investigates the effect of a highly selective cyclooxygenase-2 (COX-2) inhibitor, celecoxib, on the cytotoxicity of docetaxel in nude mice bearing A549 tumor xenografts and elucidates the molecular mechanisms of the antitumor effect of this combination. Female nu/nu mice, xenografted with s.c. A549 tumors were treated with either celecoxib (150 mg/kg/day), docetaxel (10 mg/kg) or a combination of both. The tumor tissues were quantified for the induction of apoptosis, intratumor levels/expressions of prostaglandin E2 (PGE2), 15 deoxy prostaglandin J2 (15-d PGJ2), microsomal prostaglandin E synthase (mPGES) and cytoplasmic phospholipase A2 (cPLA2). The combination of celecoxib with docetaxel significantly inhibited the tumor growth (p < 0.03) as compared to celecoxib or docetaxel alone, decreased the levels of PGE2 by 10-fold and increased the 15-d PGJ2 levels by 4-fold as compared to control. The combination also enhanced the peroxisome proliferator-activated receptor (PPAR)-gamma expression, decreased the expression of cPLA2, mPGES and vascular endothelial growth factor (VEGF), but had no effect on the expression of COX-1 or COX-2 in tumor tissues. TUNEL staining of the tumor tissues showed a marked increase in the apoptosis in the combination group as compared to the celecoxib- or docetaxel-treated groups and this was associated with an increase in the intratumor p53 expression. In conclusion, the combination of celecoxib with docetaxel produces a greater antitumor effect in s.c. A549 tumors as compared to celecoxib or docetaxel alone and this effect is associated with concomitant alterations in the intratumor levels of PGE2 and 15-d PGJ2.

Anti-cancer effect of celecoxib and aerosolized docetaxel against human non-small cell lung cancer cell line, A549.

Direct delivery of chemotherapeutic agents to the lung can increase both the drug concentration and exposure period to lung tumours. The objective of this study was to formulate docetaxel (DOC) into a metered dose inhaler (MDI), assess its aerodynamic characteristics and to evaluate the effect of celecoxib (CXB), a cyclooxygenase-2 (COX-2) inhibitor, on the in-vitro cytotoxicity and apoptotic response of aerosolized DOC against human lung adenocarcinoma cell line A549. A stable solution-type MDI formulation was developed with 0.25% DOC and 15% w/w ethyl alcohol using HFA 134a propellant. The formulation was evaluated for medication delivery, mass median aerodynamic diameter (MMAD), geometric standard deviation (GSD), percent throat deposition, respirable mass and respirable fraction. A six-stage viable impactor was used to assess the in-vitro cytotoxicity of DOC-MDI alone or in combination with CXB. Induction of apoptosis in A549 cells by DOC (non-aerosolized and aerosolized) in combination with CXB was evaluated by established techniques, such as caspase-3 estimation and terminal deoxynucleotidyl transferase-mediated nick end labelling (TUNEL) staining. The influence of different treatments on the expression of COX-2 and peroxisome proliferator-activated receptor-gamma(PPAR-gamma) in A549 cells was studied by RT-PCR. The DOC-MDI formulation had a MMAD of 1.58 microm, (GSD = 3.2) and a medication delivery of 80 microg/shot. DOC-MDI (one shot) in combination with CXB (10 microg mL(-1)) had a cell kill of more than 80% as determined by in-vitro cytotoxicity assay. The specific caspase-3 activity in A549 cells treated with DOC (0.01 microg mL(-1)) and CXB (10.0 microg mL(-1)) combination was 4 times higher than CXB and untreated control group, respectively. Further, TUNEL staining showed significant apoptosis of A549 cells treated with aerosolized DOC alone or in combination with CXB when compared with CXB and untreated cells. The RT-PCR experiments showed similar expression of COX-2 in both control and treated groups. PPAR-gamma expression was increased in the combination treatment (0.01 microg mL(-1) DOC and 10 microg mL(-1) CXB) as compared with control (untreated), DOC (0.01 microg mL(-1)) and CXB (10 microg mL(-1)) treatments. Our results indicate the potential of inhalation delivery of DOC in the treatment of lung cancer.

A reactivity index study to rationalize the effect of dopants on Brönsted and Lewis acidity occurring in MeAlPOs.

The influence of both bivalent and trivalent metal substituents from a range of metal cation (Co, Mn, Mg, Fe and Cr) on the acidic property (both Brönsted and Lewis) of metal substituted aluminum phosphate MeAlPOs is monitored. The influence of the environment of the acid site is studied both by localized cluster and periodic calculations to propose that the acidity of AlPOs can be predictable with accuracy so that AlPO material with desired acidity can be designed. A semi-quantitative reactivity scale within the domain of hard soft acid-base (HSAB) principle is proposed in terms of the metal substitutions using density functional theory (DFT). It is observed that for the bivalent metal cations Lewis acidity linearly increases with ionic size, where as the Brönsted acidity is solely dependent on the nearest oxygen environment. Intramolecular and intermolecular interactions show that once active site of the interacting species is identified, the influence of the environment can be prescribed. Mg(II)-doped AlPO-34 shows highest Brönsted acidity and whereas Cr(III)-doped species shows lowest acidity. Fe(II)/Fe(III)-doped AlPO-34 shows highest Lewis acidity, whereas Mn(III), Mg (II) shows lowest acidity.

Molecular cloning of cDNAs and structural model analysis of two gonadotropin beta-subunits of snakehead fish (Channa maculata).

The cDNAs encoding beta-subunits of follicle-stimulating hormone (FSH) and luteinizing hormone (LH) have been cloned from the pituitary of snakehead fish, Channa maculata, and the three-dimensional structural models of the encoded FSH and LH were investigated. The cloned cDNAs, including 5'-untranslated region (UTR), open-reading frame, and 3'-UTR followed by a poly(A) tail, were obtained by reverse transcription-polymerase chain reaction and rapid amplification of cDNA end methods. The open-reading frames of FSH-beta cDNA encodes a 120-amino acid protein with a signal peptide of 18 amino acids and a mature protein of 102 amino acids; while LH-beta cDNA encodes a 140-amino acid protein with a signal peptide of 33 amino acids and a mature protein of 115 amino acids. The amino acid sequence identities of snakehead fish FSH-beta and LH-beta in comparison with other fish are 27.8-81.9% and 45.2-88.8%, respectively; while in comparison with tetrapods are 26.2-28.9% and 37.5-51.2%, respectively. Both FSH-beta and LH-beta of snakehead fish resemble most to those of Perciformes, implying their closer phylogenetic relationship. All 12 cysteine residues are conserved in snakehead fish LH-beta; while 11 cysteine residues are conserved in its FSH-beta. The third cysteine is absent in snakehead fish FSH-beta; instead, a positionally shifted cysteine residue is present at the N-terminus, as found in some phylogenetic related fish. The structure models of snakehead fish FSH and LH, constructed by using the crystal structures of human FSH and human chorionic gonadotropin as respective template, showed that the positionally shifted N-terminal cysteine residue of snakehead fish FSH-beta likely can substitute the third cysteine to form a disulfide bond with the 12th cysteine.

Effects of water on the structure and bonding of resorcinol in the interlayer of montmorillonite nanocomposite: a periodic first principle study.

Resorcinol forms a novel nanocomposite in the interlayer of montmorillonite. This resorcinol oligomer is stable inside the clay matrixes even above the boiling point of the monomer. A periodic ab initio calculation was performed with hydrated and nonhydrated montmorillonite before and after intercalation of resorcinol. For the most feasible dimer and tetramer shaped oligomer of resorcinol, the intramolecular and intermolecular hydrogen bonding feasibility has been tested using the DFT-BLYP approach and the DNP basis set in the gas phase and in the presence of aqueous solvent. After locating the active site through Fukui functions within the helm of the hard-soft acid-base principle, the relative nucleophilicity of the active cation sites in their hydrated state has been calculated. A novel quantitative scale in terms of the relative nucleophilicity and electrophilicity of the interacting resorcinol oligomers before and after solvation is proposed. Besides that, a comparison with a hydration situation and also the strength of the hydrogen bridges have been evaluated using mainly the dimer and cyclic tetramer type oligomers of resorcinol. Using periodic ab initio calculations, the formation mechanism was traced by the following two ways: (1) resorcinol molecules combine without any interaction with water or (2) resorcinol oligomerizes through water. Both the mechanism is compared and the effect of water on the process is elucidated. The results show that resorcinol molecules combine after hydration only and hence they are stable at higher temperature. The fittings of the oligomers were also tested as well by periodic calculation to compare the stability of the oligomers inside the newly formed clay nanocomposite.

Structure-property relationship in py-hexahydrocinchonidine diastereomers: ab initio and NMR study.

Two py-hexahydrocinchonidine diastereomers were selectively obtained in the heterogeneous catalytic hydrogenation of cinchonidine over supported Pt catalyst. The two isolated compounds when used as chiral base catalysts in the Michael addition of a beta-keto ester to methyl vinyl ketone gave products of opposite configuration in excess. To trace the reason of this behavior, in the present study, the structures of the two diastereomers were fully optimized by ab initio quantum chemical calculation. These results were then compared with several nuclear Overhauser enhancement spectroscopy (NOESY) signal intensities from the spectra of the two compounds. Further we performed a conformational search on all the optimized geometries independently for the two flexible torsional angles, which are linking the quinuclidine and tetrahydroquinoline moieties present in these molecules. This study allowed us to propose the configuration of the C(4)(') chiral center. Thus, the product mixture resulted in the hydrogenation of cinchonidine containing the 4'-(S)-diastereomer in excess (de = 20%). According to the computation results the 4'-(S)-diastereomer is more stable than the 4'-(R)-diastereomer. The 4'-(S)-conformer obtained by computation has lower electronic energy than the structures obtained for the 4'-(R)-diastereomer, which may explain the excess formation of the first one. The results of the Michael addition catalyzed by these diastereomers were interpreted on the basis of these conclusions.

Time accelerated Monte Carlo simulations of biological networks using the binomial tau-leap method.

UNLABELLED: Developing a quantitative understanding of intracellular networks requires simulations and computational analyses. However, traditional differential equation modeling tools are often inadequate due to the stochasticity of intracellular reaction networks that can potentially influence the phenotypic characteristics. Unfortunately, stochastic simulations are computationally too intense for most biological systems. Herein, we have utilized the recently developed binomial tau-leap method to carry out stochastic simulations of the epidermal growth factor receptor induced mitogen activated protein kinase cascade. Results indicate that the binomial tau-leap method is computationally 100-1000 times more efficient than the exact stochastic simulation algorithm of Gillespie. Furthermore, the binomial tau-leap method avoids negative populations and accurately captures the species populations along with their fluctuations despite the large difference in their size. Recently, Gillespie introduced the tau-leap approximate, accelerated stochastic Monte Carlo method for well-mixed reacting systems [J. Chem. Phys. 115, 1716 (2001)]. In each time increment of that method, one executes a number of reaction events, selected randomly from a Poisson distribution, to enable simulation of long times. Here we introduce a binomial distribution tau-leap algorithm (abbreviated as BD-tau method). This method combines the bounded nature of the binomial distribution variable with the limiting reactant and constrained firing concepts to avoid negative populations encountered in the original tau-leap method of Gillespie for large time increments, and thus conserve mass. Simulations using prototype reaction networks show that the BD-tau method is more accurate than the original method for comparable coarse-graining in time.

Formulation and evaluation of aerosolized celecoxib for the treatment of lung cancer.

PURPOSE: We examined the effect of aerosolized celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, on the in vitro cytotoxicity and apoptotic response of docetaxel against the human lung carcinoma cell lines A549 and H460. METHODS: A metered dose inhaler (MDI) formulation of celecoxib was prepared and evaluated for its medication delivery and aerodynamic properties. The in vitro cytotoxicity of the aerosolized celecoxib-MDI alone or in combination with docetaxel was assessed using a six-stage viable impactor by a previously established method. The induction of apoptosis was evaluated by morphologic examination (acridine orange and Hoechst staining) and DNA fragmentation. Furthermore, in an attempt to identify molecular targets involved in the anticancer mechanisms of celecoxib and docetaxel, we examined their effect on the expression of an array of markers involved in the COX-2 dependent and independent pathways. RESULTS: The celecoxib-MDI had a medication delivery of 231.3 microg/shot, mass median aerodynamic diameter (MMAD) of 1.4 microm (GSD = 1.9), and respirable fraction of 50.7%. The celecoxib-MDI (2 shots) in combination with docetaxel had cell kills as high as 81.3% and 67.7% in A549 and H460 cells, respectively. Hoechst and acridine orange staining showed an enhanced induction of apoptosis in A549 and H460 cells exposed to aerosolized celecoxib with docetaxel, which was further confirmed by DNA fragmentation. Western blot analysis showed a significant reduction in cPLA2 expression in both A549 and H460 cells treated with the combination of celecoxib with docetaxel. In the COX-2 independent pathway, there was a significant increase in the expression of PPAR-gamma and p53, whereas pro-caspase-3 expression was significantly decreased, which may contribute to the enhanced apoptotic response observed with the combination treatment. CONCLUSIONS: Our results suggest that aerosolized celecoxib significantly enhances the in vitro cytotoxicity and apoptotic response of docetaxel against A549 and H460 cells, and this enhanced activity is mediated via alterations in expression of various molecular targets involved in apoptosis.

Structure and property correlation for Ag deposition on alpha-Al2O3--a first principle study.

The nature of bonding at the interface between deposited silver and (001) surface of alpha-Al2O3 for both Al-terminated and OH-terminated has been investigated using a periodic ab initio method. Substantial inter-planar relaxations within the alumina were found at both the interfaces and the bulk. The periodic calculation with Ag deposition shows that 10% of Ag loading on alumina results maximum stability. Now, this is known that, the clean alumina surface only exists at UHV condition and normally the alumina surface prefers to stay hydroxylated. We have therefore compared the silver bonding over hydroxylated alumina surface and confirmed the fact that the hydroxylated surface binds silver weakly in comparison to the clean surface and it recommends that the silver cluster over the hydroxylated surface begins to join in to form three-dimensional nuclei. The deposited Ag forms a cluster on top of the alumina surface. The Ag atomic packing was monitored to rationalize the role of packing on activity of silver. Three low-index Ag surfaces (100), (110) and (111) are investigated via the ab initio density functional calculations with ultrasoft potentials. We have monitored the relation between Ag atomic packing and its electronic properties. The results show that the structural and electronic property of Ag deposited on alumina surface depends significantly on atomic packing. Ag110 over clean alumina surface shows highest surface energy and smallest work function, whereas for the OH-terminated surface it is the Ag111. The results are discussed in view of the existing experimental data and models of metal-oxide interface.

Switching of PET fluorescence signals induced by ligand exchange reactions of N-(9-anthrylmethyl)amine on Cu(II) complexes and its application to postcolumn detection of glyphosate.

N-(9-Anthrylmethyl)amines which combine a fluorescent subunit and a chelate forming fragment have revealed a signal switching property in an aqueous solution upon complexation (off) with Cu(II) and liberation (on) of the probe molecule by substitution with an other ligand. The ligand exchange reaction between N-(phosphonomethyl)glycine (glyphosate), a typical herbicide, with N-(9-anthrylmethyl)amines on Cu(II) ion leads the fluorescence signal intensity to revive, providing an indirect detection system of glyphosate available in water of neutral pH region. The present system has been applied to the post column detection in the ion chromatographic separation of glyphosate and its metabolite aminomethylphosphonic acid (AMPA).
