Multivariate analysis of temporal descriptions of open-field behavior in wild-derived mouse strains.

The open-field test is a commonly used apparatus in many behavioral studies. However, in most studies, temporal changes of details of behavior have been ignored. We thus examined open-field behavior as measured by both conventional indices and 12 ethograms supported by detailed temporal observation. To obtain a broader understanding, we used genetically diverse mouse strains: 10 wild-derived mouse strains (PGN2, BFM/2, HMI, CAST/Ei, NJL, BLG2, CHD, SWN, KJR, MSM), one strain derived from the so-called fancy mouse (JFI), and one standard laboratory strain, C57BL/6. Conventional measurements revealed a variety of relationships: some strains did not show the hypothesized association between high ambulation, longer stay in the central area, and low defecation. Our ethological approach revealed that some behaviors, such as freezing and jumping, were not observed in C57BL/6 but were seen in some wild-derived strains. Principal component analysis which included temporal information indicated that these strains had varied temporal patterns of habituation to novelty.

Use of a standard strain for external calibration in behavioral phenotyping.

The present paper evaluates the inclusion of a standard strain or outbred stock in multi-strain behavioral phenotyping protocols to perform the same role as the external standard in biochemical assay procedures. As potential standards, the F344 inbred strain and an outbred stock of Long Evans were tested with three other inbred strains. To evaluate the influence of rearing conditions on phenotype stability, one group of F344s was born at the University of Tsukuba, another, bred elsewhere and delivered to Tsukuba at 4 weeks of age. All animals were tested in open-field (OF), runway emergence (RE) and digging tests as adults. The results showed no influence of breeding or transportation history on OF and RE behavior of the two F344 groups, while there was evidence that digging behavior may be affected by the different rearing experience. The inclusion of a 'standard strain or stock' in phenotyping protocols involving multiple inbred strains or lines of rats, mice and flies has obvious advantages by providing a reference point for inter-laboratory comparisons. The properties of inbred strains and outbred stocks favorable to their use as standards are discussed.

Conditioned effects of kindling three different sites in the hippocampal complex of the rat.

Rats received kindling stimulations to the perirhinal cortex (PRh), ventral hippocampus (VH), or dorsal hippocampus (DH) in 1 environment and an equivalent number of sham stimulations in a 2nd environment. The PRh-kindled rats displayed rapid kindling and a swift emergence of conditioned interictal defensiveness. In contrast, the VH- and DH-kindled rats displayed much slower kindling and slow or no conditioning, respectively. No effects of conditioning on the convulsions, comparable with those associated with amygdala kindling, were observed. These results establish the generality of some of the previously reported kindling-related conditioned effects, confirm the site specificity of some of these effects, and suggest that the convulsions, rather than the stimulations, function as the unconditioned stimuli for the conditioning of interictal behavior.

Anticipating the attack: temporal conditioning during amygdala kindling in rats.

The present study showed that amygdala-kindled rats use short-interval timing superimposed on phase or ordinal timing to predict when a convulsion will occur. In 2 experiments, rats received 1 stimulation and 1 sham stimulation each day, always at the same times (conditioned stimulus [CS]+ and CS- times, respectively) and 150 s after rats had been placed in the testing chamber (the preadministration interval). As kindling progressed, the rats displayed more defensive behavior at the CS+ time than at the CS- time. Then, a stimulation-free peak-procedure test was conducted: At the CS+ time, but not at the CS- time, defensive behavior increased progressively as the 150-s preadministration interval elapsed, and then it gradually declined.

Squamous metaplasia mimicking papillary carcinoma in the upper urinary tract.

We experienced a case of squamous metaplasia mimicking papillary urothelial cell carcinoma in the upper urinary tract. A 69-year-old woman, who complained of gross hematuria and intermittent left flank dull pain underwent nephrectomy with the clinical diagnosis of papillary urothelial carcinomas in the left upper urinary tract according to positive split urine cytology and tumorous filling defects of contrast media by the abdominal CT scan. Pathological diagnosis was squamous metaplasia and concomitant foreign body granuloma. Those changes were judged due to a tiny calculus in the ureter. Our presented case implies that a tiny calculus can cause the metaplastic change in the urothelial epithelium and the combination of radiographical and cytological diagnoses would not be enough to lead the correct diagnosis and the definitive surgical treatment against protruding lesions in the upper urinary tract requires more reliable diagnostic modalities.

Single-nucleotide polymorphism analysis by hybridization protection assay on solid support.

The clinical need for high-throughput typing methods of single-nucleotide polymorphisms (SNPs) has been increasing. Conventional methods do not perform well enough in terms of speed and accuracy to process a large number of samples, as in clinical testing. We report a new DNA microarray method that uses hybridization protection assay (HPA) by acridinium-ester-labeled DNA probes. Probes were immobilized on the bottom of streptavidin-coated microtiter plates by streptavidin-biotin binding. We studied aldehyde dehydrogenase 2 (ALDH2) genotyping using two probes, discriminating A/G polymorphism. We also designed four probes to type the Alzheimer's disease-related gene ApoE, which has three genotypes (ApoE2, 3, and 4) determined by two SNP loci (C/T polymorphism). SNP analysis of the ALDH2 gene or the ApoE gene from human genome samples by solid-phase HPA was successful. Unlike other methods, the microarray by HPA does not require a washing step and can be completed within 30min. It also has advantages in discriminating one-base mismatch in targets. These characteristics make it a good candidate for practical SNP analysis of disease-related genes or drug-metabolizing enzymes in large numbers of samples.

Simultaneous determination of endogenous and orally administered (15)N-labeled polyamines in rat organs.

A method for the simultaneous determination of polyamines (putrescine, spermidine, and spermine) by ionspray ionization-mass spectrometry was modified to determine (15)N-labeled polyamines together with unlabeled polyamines using (13)C,(15)N double-labeled polyamines as internal standards. This technique permitted the use of (15)N-labeled polyamines as tracer compounds to follow polyamine biosynthesis, interconversion, and absorption. The method was used to examine the organ distribution of orally administered (15)N-labeled polyamines in rats. Each (15)N-labeled polyamine was taken up by the three organs tested: the small intestine, liver, and kidney. The uptake of (15)N-labeled spermidine was greater than that of (15)N-labeled spermine and putrescine. Administration of a mixture of (15)N-labeled polyamines was useful for determining the disposition of each (15)N-polyamine absorbed from the intestinal tract.
