Accelerated decrease in bone mineral density in women aged 52-57 years.

Bone mineral density (BMD) has been known to decline in middle-aged and elderly individuals, but when this decline begins and the rate at which it occurs remain unclear. We thus undertook this study to examine the association between BMD and age by their mean values in women visiting the Shimane Institute of Health Science for medical examination. We performed dual energy x-ray absorptiometry measurement of lumbar vertebrae in 1,167 women, and of the entire skeleton in 1,038 women. The ages of subjects ranged from 30 to 70 years. We found that the mean value of whole-body and lumbar BMD changed little in the age range of 30-51 years, and any change after 58 years was a gradual decrease, unlike the sharp decrease found between 52 and 57 years of age. The effects of endocrine kinetics may be reflected in women by the decrease of bone density relative to age. In conclusion, BMD declines more rapidly in women within the age range of 52-57 years than in those 58 years and over. This regression line is considered useful in predicting BMD of whole-body skeleton and lumbar vertebrae relative to age for the prevention of osteoporosis in women.

Genetic mosaics in the massive persisting rhizosphere colony "shiro" of the ectomycorrhizal basidiomycete Tricholoma matsutake.

The ectomycorrhizal basidiomycete Tricholoma matsutake produces commercially valuable fruit bodies "matsutake" on a massive persisting rhizosphere aggregate of mycelia and mycorrhizas called "shiro." Using inter-retrotransposon amplified polymorphism analysis, we attempted to explore the potential diversity within the population of T. matsutake isolated from small Pinus densiflora woodlands located in various parts of Japan. In general, random phylogenetic relationship was noted among T. matsutake tested. The population from each limited sampling area was highly heterogeneous. Even some isolates from fruit bodies produced in the same shiro and those from spores in the same fruit bodies were found to be genetically diverse, indicating the occurrence of genetic mosaics in shiro. In a mosaic shiro, heterologous genets produced their fruit bodies concurrently. Data suggested that the dispersal of spores through sexual reproduction may have been more prevalent than generally accepted in T. matsutake to bring mosaicism and coordination of heterologous genets within the shiro. Implementation of management taking such diversity into consideration is urgently needed for the restoration of devastated matsutake fields in Japan. Exploration of individual clones in mosaic fungal resources that promote colonization and fruit body production is necessary for it.

Specific distribution in homobasidiomycete hymenophores of the transcripts of Ras protein and G-protein alpha-subunit genes.

Results of in situ RNA-RNA hybridization showed a complementary distribution of the transcripts of Ras protein gene (ras) and trimeric G-protein alpha-subunit gene (ga1) in the hymenophores of white-rot basidiomycete Lentinula edodes and ectomycorrhizal basidiomycete Lyophyllum shimeji. The ras gene is present mostly in outer region of trama (the region branching out into the subhymenium) and in trams cells, while the ga1 gene is present mostly in hymenium (on which a large number of basidiospores are formed) and in subhymenium (on the top of which hymenium is formed). This suggests that the ras and ga1 genes play distinct physiological roles in the hymenophores of both basidiomycete fungi.

Preparation of new nitrogen-bridged heterocycles. 56. Syntheses and reactions of 1-[2,2-bis(alkylthio)-1-(ethoxycarbonylacetyl)vinyl]pyridinium salts.

The title compounds, readily available from the S-alkylation of pyridinium 1-[alkylthio(thiocarbonyl)](ethoxycarbonylacetyl)methylides with alkyl halides or alkyl bromoacetates, were treated with a base and then a dehydrogenating agent to provide some unique products such as 3-[bis(alkylthio)methylene]-2(3H)-indolizinones and dialkyl 7-methyl-4-oxo-1,4,8,8a-tetrahydro-1,4-thiazino[3,4,5-cd]indolizine-1,5-dicarboxylates. On the other hand, similar reaction of these pyridinium salts in the absence of the dehydrogenating agent afforded alkyl 2-hydroxyindlizine-3-carboxythiolates, whose yields were increased by adding trifluoroacetic acid to the reaction mixture. The structures of some products were confirmed by the X-ray analyses.

Deterioration in learning and memory of inferential tasks for evaluation of transitivity and symmetry in aged SAMP8 mice.

This study examined age-dependent deficits in the learning and memory of inferential tasks, using an established senescence-accelerated mouse model in age-related brain dysfunction (SAMP8) and its genetically related inbred strain (SAMR1). The mice learned two sets of nonspatial odor-odor pairs by association learning successively (i.e., A-->B, X-->Y, then B-->C, Y-->Z). They were tested in transitive inference (i.e., A-->C, X-->Z) and symmetrical inference (i.e., C-->B, Z-->Y). In the probe test of A-->C, X-->Z transitive inference, 1-month-old SAMP8 and control SAMR1 at the same age significantly chose the alternative based on transitive inference, but 4- and 7-month-old SAMP8 performed at a random chance level, in comparison with unambiguous inference by control SAMR1 at the same ages. During the test of C-->B, Z-->Y symmetrical inference, SAMP8 at 1 month of age made errors as frequently as control SAMR1 at the same age, but SAMP8 at 4 and 7 months of age made more errors than SAMR1 at the same ages. At 4 and 7 months of age, SAMP8 made more errors than 1-month-old SAMP8. Control SAMR1 did not show such an age-related deficient. These results indicate that SAMP8 mice have age-related learning and memory deficits in the ability to perform inferential tasks. Age-related hippocampal dysfunction is suggested to be the cause of these age-related deficits in old SAMP8 mice during the performance of inferential tasks mediated by declarative memory.
