Application of lactate amperometric sol-gel biosensor to sequential injection determination of L-lactate.

This work describes the construction and evaluation of lactate sol-gel biosensors to accomplish the determination of lactate in pharmaceutical products. Lactate oxidase was incorporated in a porous sol-gel film placed onto a platinum-based electrode. Acid and basic catalysis were assessed. When coupled to a sequential injection system (SIA) the biosensor, based on (3-aminopropyl)trimethoxysilane, 2-(3,4-epoxycyclohexyl)ethyl-trimethoxysilane, deionised water, polyethylene glycol 6000 and acid catalyst, presented a range of linearity of 5x10(-5) to 5x10(-3)M. The analytical usefulness of the developed biosensor was evaluated through analysis of commercial pharmaceutical products containing lactate with a sampling rate of 40 samples h(-1). The enzyme remained active for at least 30 days, enabling about 700 determinations without sensitivity decrease.

Potentiometric behaviour of ion selective electrodes based on iron porphyrins: the influence of porphyrin substituents on the response properties and analytical determination of diclofenac in pharmaceutical formulations.

The potentiometric response characteristics of diclofenac selective electrodes based on Fe(III) tetraphenylporphyrin-chloride (Fe(III)TPP-Cl) and Fe(III) tetrakis(pentafluorophenyl)porphyrin-chloride (Fe(III)TPFPP-Cl) in different mediator solvents and ionic additives are compared. The sensitivity, working range, detection limit, response mechanism, and selectivity of the membrane sensor show a significant dependence on the type of carrier substituent and on the pH value of the sample solution. Studies performed with different amounts of cationic additive (tetra-n-octylammoniumbromide (TOABr)) and anionic additive (sodium tetraphenylborate (NaTPB)) in the membranes allowed the determination of the potentiometric mechanism of action of the used metalloporphyrins. For the analysis of real samples, Fe(III)TPFPP-Cl (type G), prepared in o-NPOE, incorporating 10 mol% of TOABr, was used. This potentiometric unit presented a linear response towards diclofenac concentrations between 10(-5) and 10(-2)mol l(-1) ( I=0.1 mol l(-1)) and slopes of about -59 mV dec(-1), exhibiting a response time of 10s in a buffered solution of ammonia-ammonium sulphate with pH 9.9. The potentiometric analysis of sodium diclofenac in pharmaceutical formulations was carried out by direct potentiometry and the obtained results were compared to those provided by HPLC, presenting relative errors inferior to 1.0%.

Construction and evaluation of PVC and sol-gel sensor membranes based on Mn(III)TPP-Cl. Application to valproate determination in pharmaceutical preparations.

The construction and general performance of new valproate-selective electrodes based on manganese(III) tetraphenylporphyrin [Mn(III)TPP-Cl], as an ionophore, are presented. The ionophore was incorporated into PVC and ceramic membranes (sol-gel) based on methyltriethoxysilane. The influence of membrane composition and pH and the effect of lipophilic cationic and anionic additives in PVC membranes were investigated concerning their influence on the slope, response time, selectivity and lifetime of the electrodes. The PVC membrane without additive and the sol-gel membrane presented slopes and practical limits of detection of -60.8 mV dec(-1) and 5x10(-6) mol l(-1) and -60.3 mV dec(-1) and 1x10(-4) mol l(-1), respectively. The sol-gel membranes displayed higher selectivity for valproate when compared with PVC membranes. These two types of electrodes were coupled to a sequential-injection analysis (SIA) system for the direct determination of valproate in pharmaceutical formulations. The association of Mn(III)TPP-Cl with the sol-gel support inserted in a SIA system provided potentiometric sensors with an analytical range of 1x10(-3)-5x10(-2) mol l(-1), with a sample rate of 55 samples per hour and a sample and carrier consumption of 140 and 2,500 microl per determination, respectively.

Extraction and recovery of chromium from electroplating sludge.

This work reports a study of the extraction and recovery of chromium from the wastes (class I dangerous) generated by a galvanic manufacturer. Commercial HCl at room temperature was employed, and the conditions of the extraction process were optimized according to a sequential experimental design, which also included the acid concentration and contact time as variables. The best extraction conditions (80% v/v; 30 min; 97.6% Cr) for the chromic sludge were chosen in order to make the recovery process economically feasible. After each extraction, the residue was submitted to leaching essays, to assess environmental risks. It was found that sludge could be characterized as no longer dangerous. In the recovery study, a simple and low-cost technique was evaluated for selectivity based on an oxidation step with hydrogen peroxide. A 2(3) factorial design to assess the influence of oxidation time (min), temperature ( degrees C) and peroxide amount (mol/L) was employed. The best conditions, yielding a chromium recovery of about 92%, were a time of 60 min, a temperature of 60 degrees C and 2.1 mol/L peroxide. Additional essays revealed that the same result could be obtained with more economic conditions (40 min, 1.4 mol/L peroxide and 60 degrees C). This technique proved not only effective in comparison with existing alternatives, but also low costing.

Flow-through sol-gel optical biosensor for the colorimetric determination of acetazolamide.

An optical biosensor based on immobilised carbonic anhydrase and its application to the determination of the anti-glaucoma agent acetazolamide by enzyme inhibition measurements, is described. The enzyme and a pH indicator dye, cresol red, were physically immobilised in overlapped sol-gel thin films, in a dual-layer format. Carbonic anhydrase catalyses the dehydration of HCO3-, which in turn causes a change of pH in the microenvironment of the sensor. By following the colour transition of cresol red, the enzymatic reaction as well as its inhibition by acetazolamide can be monitored. The sensor was integrated in a flow cell and coupled to a continuous flow system operating on a multicommutation and binary sampling approach. Measurements were made at pH 6.0 at the wavelength of 570 nm. Linear response was obtained for acetazolamide concentrations between 1.0 and 10.0 mmol l(-1), with a sampling frequency of 22 samples h(-1) and a detection limit of 0.2 mmol l(-1). The results obtained in the analysis of real samples were in good agreement with those obtained by a reference method, showing no significant differences at a confidence level of 95%.

Ion selective electrodes for penicillin-G based on Mn(III)TPP-Cl and their application in pharmaceutical formulations control by sequential injection analysis.

The work describes the construction, evaluation and analytical application of ion selective electrodes sensitive to penicillin-G antibiotics for pharmaceutical products analysis. Different types of polymeric membranes based on PVC (poly(vinyl chloride)) and EVA (ethyl-vinyl-acetate), without internal reference solution, were prepared using 5,10,15,20-tetraphenylporphyrinate (TPP) manganese(III) (Mn(III)TPP-Cl) as electroactive material. Different additives such as tetra-n-octylammoniumbromide (cationic additive) and sodium tetraphenylborate (anionic additive) were incorporated into the membranes to evaluate their influence on electrodes performance. The comparison of the developed detectors was based on general analytical characteristics, selectivity and lifetime. To accomplish the analysis of real samples, two selective membranes composed of 33.0% (w/w) of PVC, 66.0% (w/w) of o-NPOE and 1.0% (w/w) of Mn(III)TPP-Cl (type A) and 33.0% (w/w) of PVC, 66.0% (w/w) of o-NPOE, 1.0% (w/w) of Mn(III)TPP-Cl and 10% mol (relative to the molar concentration of Mn(III)TPP-Cl) of sodium tetraphenylborate (type B) were used. Type A electrode presented a linear response between 2 x 10(-5) and 10(-1) moll(-1) for penicillin-G, a slope of about -59 mVdec(-1) and a reproducibility of about +/-0.5 mVday(-1), while type B exhibited a linear response between 5 x 10(-5) and 10(-1) moll(-1) for penicillin-G, a slope of about -61 mVdec(-1) and a reproducibility of about +/-0.3 mV day(-1). The potentiometric analysis of penicillin-G in pharmaceutical products was carried out by direct potentiometry and the results obtained were compared with those provided by the HPLC reference method. These membranes (type A and type B) were used to prepare tubular electrodes that were coupled to a sequential injection system (SIA) and presented a linear range between 2 x 10(-4) and 1 x 10(-2) moll(-1) and slopes of -59.3 +/- 0.8 and -57.3 +/- 1.2 mVdec(-1), respectively. The tubular electrode constructed using type B membrane (type TB) was used to carry out the potentiometric analysis of penicillin-G in pharmaceutical formulations. The proposed procedure enabled relative errors between 0.1% and 1.2% (n = 4) and a sampling-rate of about 25 samples per h.

Chloride-selective membrane electrodes and optodes based on an indium(III) porphyrin for the determination of chloride in a sequential injection analysis system.

Two quasi-independent methods for potentiometric and optical determination of chloride were simultaneously implemented in a flow system, providing real-time assessment of the quality of results. A potentiometric and an optical polymeric membrane doped with the same indium(III) octaethyl-porphyrin were used as sensor ionophore. The working mechanism and the analytical characteristics of these porphyrin-based sensors with respect to dynamic range, selectivity, repeatability and lifetime are discussed. These sensors, utilised as detectors in a flow system, were applied for the analysis of chloride in pharmaceutical solutions. The quality of the results obtained was evaluated by comparison with those provided by the reference method and no significant statistical differences at the 95% confidence level were observed. The simultaneous attainment of two measurements permitted the standardisation of results in real time and the detection of failures in the procedure.

Direct determination of copper in urine using a sol-gel optical sensor coupled to a multicommutated flow system.

In this work, a multicommutated flow system incorporating a sol-gel optical sensor is proposed for direct spectrophotometric determination of Cu(II) in urine. The optical sensor was developed by physical entrapment of 4-(2-pyridylazo)resorcinol (PAR) in sol-gel thin films by means of a base-catalysed process. The immobilised PAR formed a red 2:1 complex with Cu(II) with maximum absorbance at 500 nm. Optical transduction was based on a dual-colour light-emitting diode (LED) (green/red) light source and a photodiode detector. The sensor had optimum response and good selectivity towards Cu(II) at pH 7.0 and its regeneration was accomplished with picolinic acid. Linear response was obtained for Cu(II) concentrations between 5.0 and 80.0 microg L(-1), with a detection limit of 3.0 microg L(-1) and sampling frequency of 14 samples h(-1). Interference from foreign ions was studied at a 10:1 ( w/ w) ion:Cu(II) ratio. Results obtained from analysis of urine samples were in very good agreement with those obtained by inductively coupled plasma mass spectrometry (ICP-MS); there was no significant differences at a confidence level of 95%.

Sequential injection chromatographic determination of paracetamol, caffeine, and acetylsalicylic acid in pharmaceutical tablets.

In this contribution, a new separation method for simultaneous determination of paracetamol, caffeine, acetylsalicylic acid, and internal standard benzoic acid was developed based on a novel reversed-phase sequential injection chromatography (SIC) technique with UV detection. A Chromolith Flash RP-18e, 25-4.6mm column (Merck, Germany) and a FIAlab 3000 system (USA) with an 8-port selection valve and a 5 mL syringe were used for sequential injection chromatographic separations in our study. The mobile phase used was acetonitrile-(0.01 M) phosphate buffer (10:90, v/v) pH 4.05, flow rate 0.6 mL min(-1). UV detection was at 210 and 230 nm. The validation parameters showed good results: linearity (r >0.999) for all compounds, detection limits in the range 0.3-0.8 microg mL(-1), repeatability (RSD) of peak heights between runs in the range 1.10-4.30% at three concentration levels and intra-day repeatability of the retention times in the range 0.28-0.43%. The analysis time was <6 min. The method was found to be applicable for the routine analysis of the active compounds paracetamol, caffeine, and acetylsalicylic acid in pharmaceutical tablets.

Sequential injection analysis of lead using time-based colorimetric detection and preconcentration on an anionic-exchange resin.

The development of a sequential injection analysis manifold for the colorimetric determination of lead in water samples is described The concentration of lead was assessed from its catalytic effect on the reaction of resazurine reduction caused by sulfide in an alkali medium. To that effect, the reaction zone was stopped at the detector, and the time interval required for the attainment of an absorbance decrease of 0.800 at the wavelength of 610 nm was estimated. Interference of other transition metals of the samples was minimized by adding potassium iodide to the sample and retaining the iodocomplexes formed in an on-line anionic resin (AGI X8). Elution was made with a 2 mol/L sodium hydroxide solution. The relationship [SIA] microg/L = 0.99 (+/- 0.11) x [ETAAS] microg/L + 0 (+/- 4) was obtained upon comparing the results given by the proposed system and by electrothermal atomization atomic absorption spectrometry (ETAAS) after the analysis of ten water samples.

Amperometric biosensor based on monoamine oxidase (MAO) immobilized in sol-gel film for benzydamine determination in pharmaceuticals.

The development and application of a flow-through amperometric biosensor for benzydamine determination in anti-inflammatory drugs is described. The biosensor was obtained by physical entrapment of monoamine oxidase in a sol-gel film applied on platinum or carbon paste conducting support. The sol-gel membranes were prepared using an optimum concentration of 3-aminopropyltriethoxy silane, 2-(3,4-epoxycyclohexyl)ethyl-trimethoxy silane, double distilled water saturated with polyethylene glycol 6000 and HCl. The developed biosensors were incorporated in a single channel flow injection system to enable the determination of benzydamine in the concentration range of 0.05-2.5 mmol l(-1) (with platinum based electrode) or within 0.1-2.5 mmol l(-1) (carbon paste based electrode). The operational stability of the bioanalytical system developed was about 3 months permitting approximately 4700 substrate measurements. The flow injection system developed enables a sampling rate of 20-25 samples h(-1) and relative S.D. of results less than 4%. The analytical usefulness of the proposed procedure was evaluated through analysis of commercial pharmaceutical products containing benzydamine, available on the Portuguese market. The results obtained did not differ significantly from the values resulting from analysis of the same products by the method described in the BP Pharmacopoeia.

Flow-injection amperometric determination of dopamine in pharmaceuticals using a polyphenol oxidase biosensor obtained from soursop pulp.

The amperometric determination of dopamine (Do) in pharmaceuticals formulations by flow injection analysis (FIA) is proposed. An enzymatically modified carbon paste electrode constituted by 25% (w/w) of polyphenol oxidase obtained from Annona muricata L. tissue, 30% (w/w) of graphite, 30% (w/w) of silicone and 15% (w/w) of 7,7,8,8 tetracyanoquinodimethane (TCNQ), was used as flow-through detector. The flow amperometric detection was carried out at a potential of 0.10 V (vs. Ag/AgCl) when an injected sample volume of 250 microl was inserted on a 0.3 M phosphate buffer carrier solution (pH 7.8) flowing at 2.5 ml/min. The developed biosensor showed good stability and reproducibility, enabling up to 500 determinations in 60 days, without considerable loss of enzymatic activity. The FIA system presented a linear response to Do concentrations in the interval from 2 x 10(-2) to 2 x 10(-4) M, with relative standard deviations lower than 1.5%. The kinetic parameter K(M) for the soluble and immobilized enzyme was 1.45 x 10(-2) and 1.91 x 10(-2) M, respectively. In the analyses of different commercially pharmaceutical formulations a relative deviation lower than about 3.4% was obtained.

Determination of bopindolol using the flow injection technique coupled with solid phase extraction.

In the proposed procedure, the determination of bopindolol using a flow injection analysis (FIA) technique, with spectrophotometric detection at 635 nm, is described. The method is based on the production of a green, water-soluble complex with ferric ions in acid medium. The automated lab-made FIA system was used for the direct determination of bopindolol in tablets. Bopindolol was adsorbed onto the solid phase in a mini-column, which was integrated directly into the flow system. The positive feature of the use of solid phase extraction (SPE) was the pre-concentration of bopindolol (seven times). The sample throughput was 50 samples per hour. Using the SPE method, bopindolol was determined with a linear range from 125 to 1000 microg ml(-1) (Relative standard deviation (R.S.D.)=1.87%), with a detection limit (3sigma) of 70 microg ml(-1). The method was applied to the determination of bopindolol in Sandonorm tablets. The results obtained were compared with a conventional HPLC method, both analytical techniques were in good agreement.

Simple and inexpensive flow L-glutamate determination using pumpkin tissue.

This work refers to a very easy to implementate flow injection system with potentiometric detection for l-glutamate determination in food samples. The proposed procedure is based on measurement of carbon dioxide produced by decarboxylation of l-glutamate catalyzed by l-glutamate decarboxylase (E.C. 4.1.1.1.5) from Cucurbita maxima (pumpkin). The FI potentiometric system includes an enzymatic reactor with a length of 8 cm and thickness of 5 mm packed with 200 mg of a C. maxima outer layer cut in to small pieces. The proposed procedure allowed l-glutamate determinations in the concentration interval of 10-100 mmol L(-1) for an injected sample volume of 50 microL. A phosphate buffer (0.1 mol L(-1), pH 5.5) solution flowing at 1.4 mL min(-1) was used as the carrier solution in the system. The results obtained in the analysis of food samples revealed a relative error lower than 5% when compared with those provided by the spectrophometric reference procedure. The immobilized reactor retained its initial activity for 21 days. It was possible to measure 40 samples/h with the flow system proposed.

Determination of dipyrone in pharmaceutical products by flow injection analysis with potentiometric detection.

This work describes an FIA potentiometric procedure for the quantification of dipyrone in pharmaceutical products. For the detector, a tubular electrode comprising a polymeric membrane containing tetraoctylammonium as an electroactive material (5% w/w), dibutylphtalate as a mediator solvent (65% w/w) and PVC (30% w/w) directly applied above a graphite conductor support was used. This unit was incorporated into a monochannel FI-system with a 0.1 mol/L phosphate buffer solution (pH = 5.2) as the carrier solution. The electrode showed a linear response from 8.0 x 10(-4) to 10(-1) mol/L dipyrone, a slope of 62.1 +/- 0.2 mV/dec in pH 5.2 units, an injection volume of 500 microL and a carrier flow-rate of 6 mL/min. This procedure was applied to the analysis of pharmaceutical formulations (oral and injectable) containing dipyrone; the obtained results gave a relative error of less than 3.9% and coefficients of variation less than 1% and 5%, respectively, for the FIA and classical iodometric methods.

A sequential injection analysis system for potassium clavulanate determination using two potentiometric detectors.

Two almost independent potentiometric methods were implemented in a multi-task flow system based on the concept of sequential injection analysis for the analytical control of potassium clavulanate in pharmaceutical formulations. In addition to in-line preparation of the solutions submitted to the analysis, the system simultaneously detected the clavulanate anion and the potassium cation using two potentiometric detectors selective to each of the species referred. For the two determinations, the system developed presented an analytical range of 2 mM-0.1 M. Relative standard deviations of 0.5 and 0.6%, respectively, for clavulanate and potassium, were calculated from the signals obtained for 10 consecutive injections of a solution of 5x10(-3) M potassium clavulanate. The frequency of sampling was 53 samples h(-1). The quality of results supplied was evaluated by comparison with the reference procedure revealing relative errors less than 2% for the two determinations. The simultaneous attainment of two measurements permitted the standardisation of results in real time, the detection of faults in the procedure and monitoring the chemical stability of clavulanate.
