Expression of zebrafish Lc3 synthase gene in embryonic lens requires hedgehog signaling.

Glycosyltransferases are involved in synthesis of various glycolipids and glycoproteins that play important roles in many biological processes. We have identified a zebrafish gene encoding a member of beta1,3-N-acetylglucosaminyltransferase family, the Lc3 synthase/bGn-T5. Whole-mount in situ hybridization reveals that the Lc3 synthase gene is expressed in two distinct phases during the zebrafish embryogenesis. The early phase extends from late blastulation to the completion of epiboly, during which the expression occurs in the superficial layer of the embryos. The second phase of expression starts during mid-segmentation and persists until day 3, during which the expression occurs prominently in the developing lens. The expression of the Lc3 synthase gene in the lens is inhibited in you-too (yot) mutant embryos that are defective in Hedgehog signaling. The expression in the lens also decreases in cyclops (cyc) and one-eyed-pinhead (oep) mutant embryos and lefty1-injected embryos, which are deficient in Nodal signaling and lack Hedgehog activity in the ventral brain. These results suggest that Hedgehog signaling is required for the Lc3 synthase expression in embryonic lens.

Zebrafish sox9b is an early neural crest marker.

Sox9 is a transcription factor related to campomelic dysplasia and sex reversal in human patients. Earlier studies in zebrafish led to the identification of two homologues of mammalian sox9, sox9a and sox9b. The present study represents the first evaluation of expression patterns of zebrafish sox9b during early embryogenesis. Our analyses reveal that sox9b transcripts are present throughout the life-cycle of the zebrafish, but exhibit tissue-specific distribution during embryogenesis. Zygotic expression of sox9b occurs in the anterolateral margins and the midline of the prospective dorsal neuroectoderm during late gastrulation. During early segmentation, the transcript is expressed in pairs of longitudinal bands in the prospective midbrain, hindbrain, and trunk, which identify the cranial and trunk neural crest progenitors. Neural crest cells cease expression of sox9b during migration, but some of their derivatives resume sox9b expression. Sox9b can serve as a marker for neural crest precursors.

The expression of gbx-2 during zebrafish embryogenesis.

Gbx-2 is a homeobox gene essential for normal development of the midbrain and the anterior hindbrain. Zebrafish gbx-2 shares an overall similarity of 67.8, 68.1, 60.6 and 66.5% in amino acid sequence to human, mouse, chick and Xenopus Gbx-2, respectively. The expression of zebrafish gbx-2 is initially, before the completion of epiboly, restricted to the prospective posterior midbrain. The expression remains detectable until the end of pharyngula period. The gbx-2 mRNA is also detected in the otic vesicles, the dorsoposterior telencephalon, the rostral branchial arches, the pronephric duct and median fin fold.

A novel zinc finger transcription factor resembles krox-20 in structure and in expression pattern in zebrafish.

Krx-20, a zinc finger protein, plays an essential role in hindbrain segmentation of vertebrates. Zebrafish gene frb35 encodes a novel zinc finger protein that shares a high homology with Krx-20. The expression domains of frb35 overlap those of krx-20. The expression of frb35 first occurs in the 3rd rhombomere primordium at the onset of segmentation, which is soon followed by expression in the 5th rhombomere. From mid-segmentation onward, the 5th rhombomere has a higher frb35 expression level than the 3rd rhombomere. Unlike krx20, frb35 expression persists in the 5th rhombomere until the end of pharyngula period.
