ROM polymerization-capture-release: application to the synthesis of unsymmetrical porphyrazinedithiols and peripherally metalated derivatives.

Crossover Linstead macrocyclization of a doubly norbornenyl-functionalized dimercaptomaleonitrile with dipropylmaleonitrile gave a crude mixture of porphyrazines containing the hexapropylporphyrazinedithiol magnesium complex. The mixture was subjected to ring-opening metathesis polymerization to yield the insoluble porphyrazinedithiol-functionalized polymers. Cleavage from the polymer backbone using mercury(II) acetate followed by reaction with electrophiles gave access to a range of thioporphyrazinedithiol derivatives including solitaire porphyrazines. Studies into the possible uses of hexapropyl-2,3-di-(carboxymethylthio)porphyrazine in sensing metal cations in solution are described.

1,4-Difluoro-2,5-dimethoxybenzene as a precursor for iterative double benzyne-furan Diels-Alder reactions.

[reaction: see text] The use of 1,4-difluoro-2,5-dimethoxybenzene as a novel precursor for iterative two-directional benzyne-furan Diels-Alder reactions, using a range of 2- and 3-substituted furans, is reported. Substituted oxabenzonorbornadienes were synthesized following the initial Diels-Alder reaction, which upon ring opening under acidic conditions gave substituted naphthol derivatives. Highly substituted anthracenols were generated in the second benzyne-furan Diels-Alder reaction following acid-catalyzed isomerization of the cycloadducts.

ROM polymerization-capture-release strategy for the chromatography-free synthesis of novel unsymmetrical porphyrazines.

[reaction: see text] Crossover-Linstead macrocyclization reactions of two norbornenyl-tagged diaminomaleonitriles with dipropylmaleonitrile gave access to crude mixtures of porphyrazines containing diamino-hexapropyl-porphyrazine magnesium complexes. The mixtures were subjected to ring-opening metathesis polymerization to yield the insoluble diaminoporphyrazine-functionalized polymers. Acid-mediated cleavage from the polymer backbone followed by acylation of the resultant sensitive macrocyclic diamines gave monoacetyl-, monotrifluoroacetyl-, and ditrifluoroacetyl-substituted porphyrazinediamines. Conversion of these amido-porphyrazines to the corresponding zinc macrocycles and studies of their electronic absorption and emission spectra, electrochemistry, and photophysics are described.

Surface-bound porphyrazines: controlling reduction potentials of self-assembled monolayers through molecular proximity/orientation to a metal surface.

We report the preparation of two novel H2[pz(An;B(4-n))] porphyrazines (pzs) which were designed to position themselves quite differently when attached to a surface: one to form a standard self-assembled monolayer (SAM) roughly perpendicular to a surface, the other to lie horizontally along a surface. As the former, we synthesized a pz, 1, where one pyrrole group is functionalized with two thioethers terminated in mercaptides (SR, R = (CH2)3CONH(CH2)2S-), each protected as a disulfide, and -S-Me is attached to the other pyrrole sites; the latter is a pz, 2, with dialkoxybenzo groups fused to two trans-pyrroles of the pz ring, and SR groups are attached to the other pair of pyrroles. Nanostructures of 1 and 2 were successfully patterned on gold surfaces via dip-pen nanolithography, and the predicted molecular orientation of the resulting structures was confirmed by topographic AFM images. The two pzs exhibit similar reduction potentials in solution. Both show large shifts in potential upon surface binding, with the magnitude of the shift depending on the proximity/orientation of the pz to the surface. The first reduction potential of the "vertically" aligned 1 shifts by ca. +430 mV when incorporated in a binary pz/hexanethiol SAM, while that for 2, which lies flat, shifts by ca. +800 mV; the potential thus shifts by ca. +370 mV upon taking a given pz that stands atop a two-legged insulating "standoff" in a traditional SAM and "laying it down". We suggest these observed effects can be explained by image-charge energetics, and this is supported by a simple model.

Full stereochemical assignment and synthesis of the potent anthelmintic pyrrolobenzoxazine natural product CJ-12662.

The structure of the unusual anthelmintic pyrrolobenzoxazine terpenoid natural product CJ-12662 was established by X-ray crystallography and partial synthesis from 2-chloronitrobenzene. An unusual Meisenheimer-type rearrangement was used to provide the core pyrrolobenzoxazine heterocycle, and coupling of a tetracyclic pyrrolobenzoxazine lactone with the terpene alcohol was used to complete the synthesis of CJ-12662.

ROMPgel-supported thiazolium iodide: an efficient supported organic catalyst for parallel stetter reactions.

[reaction: see text] A high-loading ROMPgel-supported thiazolium iodide was prepared via ROMPolymerization of the corresponding norbornene-derived monomer. The resulting ionic ROMPgel proved to be an efficient organic catalyst for Stetter reactions. The 1,4-dicarbonyl products, important intermediates in the synthesis of cyclopentenones and heterocycles, were obtained in high yields and excellent purities after minimal purification. The ROMPgel could be reused in up to four consecutive reaction cycles without significant loss of catalytic activity.

Synthesis and properties of dimetallic M1[Pz]-M2[Schiff base] complexes.

We report the synthesis and physical characterization of a series of peripherally functionalized porphyrazines (pz's) 1[M(1); M(2); R], where M(1) is a metal ion incorporated into the pz core, M(2) is a metal ion bound to a bis(5-tert-butyl-salicylidenimine) chelate built onto two amino nitrogen attached to the pz periphery, and R is a solubilizing group (either n-propyl (Pr) or 3,4,5-trimethoxyphenyl (TMP) group) attached to the remaining carbons of the pz periphery. The 1[M(1); M(2); R] species are prepared from precursor pz's with a selenodiazole ring; they are deprotected to form the diamino pz, which reacts with two moles of 5-tert-butyl-2-hydroxybenzaldehyde to form the Schiff base pz. This is metalated to form 1[M(1); M(2); R]. The crystal structures of 1[2H; Ni; Pr] and 1[Cu; ClMn; Pr] are presented. The EPR spectra of the M(1)-M(2) "isomers" prepared with Cu(II) (S = (1)/(2)) and ClMn(III) (S = 2) ions, 1[ClMn; Cu; Pr] and 1[Cu; ClMn; TMP], are a superposition of spectra expected for the S = (3)/(2) and S = (5)/(2) total-spin manifolds that result from strong Heisenberg coupling between the partner spins. The exchange splitting between the two manifolds, as determined by temperature-dependent magnetic susceptibility measurements, is equivalent for the two M(1)-M(2) "isomers", Delta/k(B) approximately 20-25 K, which suggests a sigma-pathway for exchange coupling.

Synthesis of diverse macrocyclic peptidomimetics utilizing ring-closing metathesis and solid-phase synthesis.

The synthesis of a range of highly functionalized peptidomimetic macrocycles has been accomplished using ring-closing metathesis and enyne tandem cross-metathesis-ring-closing metathesis reactions. This approach gives access to rigidified macrocycles modeled on the structures of cyclic peptides and designed to be biologically stable. The potential for peripheral functionalization of these templates has been demonstrated using Diels-Alder reactions, palladium(0) coupling reactions, and amide formation both in the solution phase and using polymer-supported syntheses.

Synthesis and reactions of aminoporphyrazines with annulated five- and seven-membered rings.

The novel five- and seven-membered ring appended aminoporphyrazines 3 and 12 have been prepared via mixed Linstead macrocyclization. The structures of both have been unequivocally established by X-ray crystallographic studies. Reductive deselenation of selenadiazole 3 in the presence of 9,10-phenanthrenequinone or 2,3-butanedione results in the formation of pyrazines 6a,b, whereas oxidation of porphyrazine 12 gave the corresponding seco derivative 14. seco-Porphyrazine 14 mediates the generation of singlet oxygen with a quantum yield of 0.74.

Peripheral metal-ion binding to tris(thia-oxo crown) porphyrazines.

We report the preparation and solution properties of metal-free [1b(H(2))] and cobalt [1c(Co)] porphyrazines (pz's), where three pyrroles are functionalized with a S(2)O(3) crown ether for metal-ion binding and the fourth pyrrole is appended with two long-chain alcohols (bis((11-hydroxyundecyl)thio)) for potential use as surface anchors. Compounds 1b(H(2) and 1c(Co) exhibit ion-specific optical changes in the presence of Ag(+) and Hg(2+). Binding of Ag(+) to 1b(H(2)) is described by a 1:1 binding isotherm, with K(D) approximately 147 microM, whereas binding of Hg(2+) to 1b(H(2)) appears more complex. For 1c(Co), binding of Ag(+) and Hg(2+) also can be fit to a 1:1 isotherm, with K(D) approximately 109 and 83 microM, respectively. All four titrations show nonisosbesticity, including those apparently describable by the 1:1 isotherm, which indicates that there are intermediate stages as multiple ions bind. Neither 1b(H(2)) or 1c(Co) gives optical responses to Ni(2+), Zn(2+), Pb(2+), or Cu(2+) or to alkali (Li(+), Na(+), K(+), and Cs(+)) and alkaline earth (Mg(2+), Ca(2+), and Ba(2+)) metal ions. Nonetheless, "hard" ions are sensed electrochemically: Na(+) and Li(+) strongly shift the pz/pz(-) couple of 1b(H(2)()) and Co(III)/Co(II) couple of 1c(Co). For the addition of 4 equiv of Li(+) to 1c(Co), the Co(III)/Co(II) shifts +40 mV, and for the addition of 4 equiv of Na(+) to 1c(Co), the shift is +155 mV. The shifted redox waves of 1c(Co) all retain their reversibility. In contrast, for the addition of either 4 equiv of Li(+) or Na(+) to 1b(H(2)), the shifts of the pz/pz(-) couple are essentially the same, approximately 170 mV, and the shifted redox waves become broadened and less reversible, due to ion-induced aggregation. For 1c(Co) the shape of the titration curves [M]/[1b(H(2))] versus E(1/2) is concave to the x-axis, implying cooperative binding of multiple M(+) ions as part of the redox/binding system of equilibria.

Tuning the singlet oxygen quantum yield of near-IR-absorbing porphyrazines.

We report the quantum yields for singlet oxygen production by a series of porphyrazines (pz) of the form M[pz(An;B4-n)] (Scheme 1), where the peripheral substituent A is [S-R]2 with R = (CH2CH2O)3H, B is a fused alpha,alpha'-dialkoxybenzo group and M = 2H, Mg or Zn. These compounds show intense near-IR absorbance/emission (longest wavelength emission, approximately 830 nm). Their solubilities vary with R, whereas their optical properties do not. We show that singlet oxygen sensitization by these luminescent compounds can be "tuned" from essentially off to on by varying n and selection among M = 2H, Mg or Zn. The quantum yields vary ca 60-fold within the set of compounds studied, from phidelta = 0.007 for compound 3 to phidelta = approximately 0.4 for compound 11.

ROMPgel-supported triphenylphosphine with potential application in parallel synthesis.

[reaction: see text] ROMPgel-supported triphenylphosphine was synthesized in three steps (67%) from norbornadiene, 4-bromoiodobenzene, and chlorodiphenylphosphine. The supported reagent has a high loading (2.5 mmol/g) and favorable swelling properties in organic solvents. It has been utilized for the conversion of alcohols to halides, the reduction of ozonides, and the isomerization of alpha,beta-acetylenic esters and in the Staudinger reaction. In general, filtration of the resin from the reaction mixtures and evaporation gave the corresponding products in high yield and purity.

Unified route to the palmarumycin and preussomerin natural products. Enantioselective synthesis of (-)-preussomerin G.

The total syntheses of eight members of the palmarumycin family have been achieved, with identification of the absolute stereochemistry for three of these natural products. In addition, the ras-farnesyl transferase inhibitor (-)-preussomerin G has been synthesized, achieving the first enantioselective route for accessing this family of natural products. Highlights of the synthetic work include an asymmetric epoxidation of a cyclic enone in excellent yield and enantiomeric excess and a potentially biomimetic oxidative spirocyclization for the introduction of the bis-spiroketal array unique to the preussomerin natural products.

Peripherally metalated secoporphyrazines: a new generation of photoactive pigments.

Base-catalyzed cross condensation of dipropylmaleonitrile 1 with bis(dimethylamino)maleonitrile 2 in an equimolar ratio afforded the porphyrazines 3a, 4a, 5a, 6a and 7a. Subsequent demetalation of 5a with TFA followed by remetalation with Zn(OAc)(2) gave ligand 5c in good yield. Compound 5c was, in turn, selectively oxidized and further peripherally functionalized using Pt(PhCN)(2)Cl(2) and PdCl(2) to yield the novel seco solitaire porphyrazines 10a and 10b. The photophysical profiles of the seco solitaire porphyrazines 10a and 10b were evaluated by means of absorption, emission, and transient absorption spectroscopy. The new pigments 10a and 10b were found to be photochemically more stable than the solitaire complexes 3d and 3e and mediated the generation of singlet oxygen with quantum yields of 0.59 and 0.45, respectively.
