Super-Charged Porphyrazines: Synthesis and Physical Properties of Octacationic Tetraazaporphyrins.

We report the synthesis of octacationic macrocycles based on the porphyrazine (tetraazaporphyrin) core. The first stage is the synthesis of Mg(II) 2,3,7,8,12,13,17,18-octakis(4-pyridyl)porphyrazine, [Mgpz(pyr)(8)], and the parent [H(2)pz(pyr)(8)]. Both compounds are freely soluble in aqueous acid due to protonation of the pyridyl groups but with concomitant loss of Mg(2+) by the latter. Methylation of [H(2)pz(pyr)(8)] gives the octacation, 2,3,7,8,12,13,17,18-octakis(N-methyl-4-pyridiniumyl)porphyrazine, [H(2)pz(Me-pyr)(8)](8+), which is freely soluble in water as the chloride salt. The peripheral charges greatly lower the pK(a) values of the pyrrole protons of [H(2)pz(Me-pyr)(8)](8+) in aqueous solution. Global fitting of optical pH titration curves gives pK(a) values for the first and second deprotonations of 6.3 and 8.8 in 2.0 M NaCl and 4.1 and 5.6 at 0.1 M NaCl. The analysis further gives the UV-visible spectra of the [H(2)pz(Me-pyr)(8)](8+), [Hpz(Me-pyr)(8)](7+), and [pz(Me-pyr)(8)](6+) species. The free-base compound [H(2)pz(Me-pyr)(8)](8+) rapidly incorporates a variety of metal ions (Ba(II), Cd(II), Co(II), Cu(II), Mn(III), Ni(II), Pd(II), and Zn(II)) to form [Mpz(Me-pyr)(8)](8+). We present the complete synthesis and characterization of the [Nipz(Me-pyr)(8)](8+) and [Cupz(Me-pyr)(8)](8+) derivatives.

Porphyrazinehexamines and Dinitroporphyrazines: Synthesis, Characterization, and Complementary Electrochemistry.

Unsymmetrical porphyrazines, M[pz(H)(2)(R)(6)], with three substituted pyrroles (substituents, R = -NMe(2), propyl, or 4-tert-butylphenyl) at the periphery and one unsubstituted pyrrole have been synthesized from inexpensive and readily available materials. Linstead crossover macrocyclization of 8 with 9, 11, and 12 gave pigments 1, 3, and 5, respectively. Subsequent demetalation by treatment with TFA gave free-base porphyrazines 2, 4, and 6. Generally, pyrrolines 11 and 12 are better co-cyclization partners and give higher yields for the desired unsymmetrical porphyrazines compared to maleonitriles 9 and 10 when co-cyclized with 8. Nickel porphyrazine 7 was obtained by reaction of 6 with Ni(OAc)(2) in chlorobenzene and dimethylformamide. Both free-base and nickel porphyrazines (6 and 7) were readily nitrated by nitrogen dioxide in CH(2)Cl(2), yielding dinitroporphyrazines 18 and 19, respectively. Electrochemical studies have shown that porphyrazinehexamine 2 is easy to oxidize, having the first oxidation at E(1/2) = - 0.18 V, which is 0.91 and 1.04 V lower than that of 4 and 6, respectively. Dinitroporphyrazine 18, however, displays a 0.6 V of cathodic shift for the two reversible reduction waves compared to those observed for the parent porphyrazine 6.

Applications of Crotonyldiisopinocampheylboranes in Synthesis: Total Synthesis of Restrictinol.

The total synthesis of restrictinol, the hydrolysis product of the antifungal natural product restricticin, starting from commercially available methyl (S)-(+)-3-hydroxy-2-methylpropionate is described. Key stages in the strategy involved (i) the use of Brown's allylboration chemistry to construct an acyclic intermediate bearing three of the four stereogenic centers of the natural product, (ii) formation of a C-glycosidic vinyl iodide, and (iii) introduction of the triene side chain via a Suzuki coupling reaction.

Peripheral Palladium(II) and Platinum(II) Complexes of Bis(dimethylamino)porphyrazine.

The unsymmetrical porphyrazine (tetraazaporphyrin) bearing a single peripheral bis(dimethylamino) functionality, Mg[pz(NMe(2))(2)(Pr)(6)], was prepared by base-catalyzed cross condensation of dipropyl maleonitrile (in excess) with dimethylamino maleonitrile. The freebase (2H[pz(NMe(2))(2)(Pr)(6)]) and centrally metalated forms (M[pz(NMe(2))(2)(Pr)(6)]; M = Ni(II), Cu(II), Mn(III)) were prepared by treatment of Mg[pz(NMe(2))(2)(Pr)(6)] with trifluoroacetic acid and then the appropriate metal salt. PdCl(2) and PtCl(2) were coordinated to the peripheral bis(dimethylamino) chelates, yielding the bimetallic complexes, M[pz(NMe(2))(2)(Pr)(6)]M'Cl(2) (M = Ni, Cu; M' = Pd, Pt). The heteroleptic [N(2)-Pd-S(2)]-capped porphyrazines were prepared readily by substituting the chloride ions of M[pz(NMe(2))(2)(Pr)(6)]PdCl(2) with the dithiolene chelates, maleonitriledithiolate (mnt(2)(-)), benzenedithiolate (bdt(2)(-)), and 1,3-dithiole-2-one-4,5-dithiolate (dmid(2)(-)). The [N(2)-Pt-S(2)] complexes were prepared by reaction of M[pz(NMe(2))(2)(Pr)(6)]PtCl(2) with the dialkyltin-protected dithiolates dibutyltin(toluene-3,4-dithiolate) and dibutyltin(dmit). The peripheral heteroleptic [N(2)-M'-S(2)] core was found to be electroactive for electron-rich dithiolene ligands (bdt(2)(-), E(1/2)(Pd(III)/Pd(II)) = 0.22 V; tdt(2)(-), E(1/2)(Pt(III)/Pt(II)) = 0.20 V; dmid(2)(-), E(1/2)(Pd(III)/Pd(II)) = 0.19 V; dmit(2)(-), E(1/2)(Pt(III)/Pt(II)) = 0.19 V) but not for the relatively electron-poor dithiolene, maleonitriledithiolate (mnt(2)(-)). The X-ray structure of Ni[pz(NMe(2))(2)(Pr)(6)]Pd(mnt) (13) was determined. Crystal data for C(85)H(109)Cl(3)N(24)Ni(2)Pd(2)S(4): space group P2(1)/n; a = 17.435(4) Å, b = 17.982(2) Å, c = 30.577(2) Å; beta = 104.27(2) degrees; Z = 4.

Nucleophilic Substitution Reactions of (Alkoxymethylene)dimethylammonium Chloride.

The use of imidate esters as potential replacements for diethyl azodicarboxylate and triphenylphosphine in the Mitsunobu reaction is described. A series of secondary alcohols were allowed to react with (chloromethylene)dimethylammonium chloride, generated from dimethylformamide (DMF) and oxalyl chloride, to give imidate esters. Reaction of these salts with potassium benzoate or potassium phthalimide gave the products of S(N)2 substitution in excellent yields with clean inversion of stereochemistry. Optimization of reaction conditions is discussed as a means to increase the atom economy of the process by minimizing the quantity of nucleophile required.

Porphyrazines and Norphthalocyanines Bearing Nitrogen Donor Pockets: Metal Sensor Properties.

The amino-functionalized porphyrazines 1, 2, and 4 and norphthalocyanines 3 and 5, which were prepared via Linstead macrocyclization reactions of dinitrile precusors, were subject to UV-vis titrations in the presence of inorganic ions. These polydentate macrocycles are able to bind metal ions by the peripheral ligating diazacrown or vicinal diamine entities in addition to metal ion binding within the porphyrazine (norphthalocyanine) cavity. Although 1 only weakly bound Ag(I) and Hg(II), 2 to 5 were superior polydentate ligands. Surprisingly, porphyrazine 4 was a superior ligand for binding 4 equiv of Co(II), Cu(II), Zn(II), and Cd(II) than the corresponding tetracrowned porphyrazine 2. Both 2 and 4 were excellent ligands for peripherally complexing 4 equiv of Ag(I) and Hg(II) and only for one metal, Pb(II), was the crowned system 2 superior to 4. The crowned norphthalocyanine 3 was generally inefficient in the peripheral complexation of inorganic cations, whereas 5 was superior for the peripheral binding of Co(II), Cu(II), Zn(II), Cd(II), and Hg(II) (1 equiv). Again, only with Pb(II) was the crowned system 3 a superior ligand to 5. X-ray crystallographic characterization of diazacrown 12a, porphyrazine 1 (M = Mg), and the complexes of crown 12b with Cu(BF(4))(2) and with HClO(4) are reported.

Diastereoselective Conjugate Addition to (+)-Camphorsulfonic Acid Derived Nitroalkenes: Synthesis of alpha-Hydroxy and alpha-Amino Acids.

Diastereoselective tandem conjugate addition of both oxygen- and nitrogen-centered nucleophiles to the novel (1S)-10-camphorsulfonic acid derived nitroalkenes 9, 10, and 11 and ozonolysis gave the alpha-hydroxy and alpha-amino thiol acid derivatives 12, 13, and 14. In all cases, the (R)-diastereomer was formed as the major component albeit with only modest levels of selectivity (33-71% de). The structures of the products and the stereochemistry of the Michael addition step were unequivocally established by X-ray crystallographic studies of nitroalkenes 9 and 10 and (2S)-12c and (2R)-13a and by alternative syntheses from (S)-alanine, (S)-valine, and ethyl (S)-lactate.

Convenient Asymmetric Syntheses of anti-beta-Amino Alcohols.

Condensation of allylborane reagents 9 and 12 with aldehydes gave anti-3-[(diphenylmethylene)amino]-1-alken-4-ols 10 and 13 with high relative and absolute stereocontrol. Subsequent deprotection gave the corresponding free anti-3-amino-1-alken-4-ols 11 and 14. Alternatively, reaction of imines 13a, 13f, and 13g with trifluoromethanesulfonic anhydride and acidic methanol gave, via rearrangement, double inversion, and hydrolysis, the isomeric anti-4-amino-1-alken-3-ols 22, 38a, and 38b in good yield. The stereochemistry of the rearrangement products has been established by a single crystal X-ray study of compound 37 and by chemical correlation.

Total Synthesis of (+)-Papuamine: An Antifungal Pentacyclic Alkaloid from a Marine Sponge, Haliclona sp.

The total synthesis of (+)-papuamine, the antipode of the C(2)-symmetric, optically active, pentacyclic diamine natural product, starting from a chiral diol is described. The diol is available via an asymmetric Diels-Alder reaction between 1,3-butadiene and di-(-)-menthyl fumarate. The key transformation in the synthesis is an intramolecular Pd(0)-catalyzed (Stille) coupling reaction to form the central 13-membered diazadiene macrocyclic ring.
