Fluorescence quenching immunoassay performed in an ionic liquid.

We describe the first example of immunoanalysis performed within an ionic liquid with minimal deleterious effect; our results bode well for the development of second-generation biosensors, particularly in applications involving poorly water soluble analytes including pesticides, phospholipids, and illicit drugs.

Validation of a standardized portable fluorescence method for determining trace beryllium in workplace air and wipe samples.

Beryllium is widely used in industry for its unique properties; however, occupational exposure to beryllium particles can cause potentially fatal disease. Consequently, exposure limits for beryllium particles in air and action levels on surfaces have been established to reduce exposure risks for workers. Field-portable monitoring methods for beryllium are desired in order to facilitate on-site measurement of beryllium in the workplace, so that immediate action can be taken to protect human health. In this work, a standardized, portable fluorescence method for the determination of trace beryllium in workplace samples, i.e., air filters and dust wipes, was validated through intra- and inter-laboratory testing. The procedure entails extraction of beryllium in 1% ammonium bifluoride (NH(4)HF(2), aqueous), followed by fluorescence measurement of the complex formed between beryllium ion and hydroxybenzoquinoline sulfonate (HBQS). The method detection limit was estimated to be less than 0.02 microg Be per air filter or wipe sample, with a dynamic range up to greater than 10 microg. The overall method accuracy was shown to satisfy the accuracy criterion (A< or = +/-25%) for analytical methods promulgated by the US National Institute for Occupational Safety and Health (NIOSH). Interferences from numerous metals tested (in >400-fold excess concentration compared to that of beryllium) were negligible or minimal. The procedure was shown to be effective for the dissolution and quantitative detection of beryllium extracted from refractory beryllium oxide particles. An American Society for Testing and Materials (ASTM) International voluntary consensus standard based on the methodology has recently been published.

Formation of an unusual charge-transfer network from an ionic liquid.

An intriguing and novel charge-transfer complex between dimethyldihydrophenazine and diethylviologen has been crystallized from an ionic liquid at room temperature, resulting in an interesting stacking motif of interrupted D***A***D type triads: efficient formation of the complex is seen within an ionic liquid and acetone, with the complex absorbing strongly across nearly the entire visible-NIR spectral region.

Structural changes upon photoexcitation into the metal-to-ligand charge-transfer state of [Cu(pqx)(PPh3)2]+ probed by resonance Raman spectroscopy and density functional theory.

The structural changes that occur when [Cu(pqx)(PPh(3))(2)](+) (pqx is 2-(2'-pyridyl)quinoxaline) undergoes excitation through a metal-to-ligand charge-transfer (MLCT) transition are investigated using resonance Raman excitation profiles coupled with density functional theory (DFT). The DFT calculations predict bond lengths to within 3 pm and absolute deviations of 7 cm(-1) for the vibrational frequencies of [Cu(pqx)(PPh(3))(2)](+). TD-DFT calculations of oscillator strengths (f = 0.089) and band positions (419 nm) showed close agreement with experiment (f = 0.07, 431 nm). Resonance Raman spectra show the 527 cm(-1) (nu(29)) and 1476 cm(-1) (nu(75)) modes undergo the largest dimensionless displacement (Delta = 1.5 and 1.1, respectively) following photoexcitation into the MLCT Franck-Condon region. The solvent couples strongly to the MLCT transition and resonance Raman intensity analysis (RRIA) gives a solvent reorganization energy of 3400 cm(-1) for dichloromethane and 2800 cm(-1) for chloroform solutions. A large inner-sphere reorganization of 3430 cm(-1) in dichloromethane solution (3520 cm(-1) in chloroform solution) was found for [Cu(pqx)(PPh(3))(2)](+), indicating that the molecule as a whole undergoes significant distortion following MLCT excitation.

Novel binding of beryllium to dicarboxyimidazole-based model compounds and polymers.

The ligand 4,5-dicarboxyimidazole (H(2)DCI) and its methyl derivative 1-methyl-4,5-dicarboxyimidazole (H(2)MDCI) have been shown to bind to Be(II) forming a zwitterionic species that has been structurally characterized. A new dicarboxyimidazole-based polymer has been prepared and its Be-binding properties have been studied using NMR ((1)H and (9)Be) and fluorescence spectroscopy; it represents a rare example of beryllium binding to a polymer. Models of the mononuclear and polymeric Be(II)-binding sites have been studied using density functional theory (DFT), and the (9)Be NMR chemical shifts of these model materials have been calculated for the purpose of direct comparison to experimentally observed values. Differences in the binding modes of the mononuclear and polymeric species are discussed.

The design and synthesis of porphyrin/oligiothiophene hybrid monomers.

In an effort to build effective photovoltaic cells based on porphyrin-functionalised polythiophenes we have focused on synthetic routes to three monomer types. By controlling the geometric structure of the monomer, oxidation of these materials should produce polymers with different architectural structures, and as a result, different opto-electronic properties. Employing Wittig protocols allowed access to monomers in which the porphyrin moiety is connected to the beta-position of the thiophene via an alkene linkage. In addition, monomers were constructed using porphyrin condensation methods to afford alpha-thiophene meso-substituted porphryins. Another set of monomers was also prepared via porphyrin condensation routes, but instead utilising beta-formylthiophenes. By utilising different formyloligothiophenes we were able to generate a series of monomers that can be used to control the loading of the porphyrin in the polythiophene matrix.

Predicting 9Be nuclear magnetic resonance chemical shielding tensors utilizing density functional theory.

The structures of a series of beryllium containing complexes have been optimized at the B3LYP/6-31G(d) level and their (9)Be magnetic shielding values have been determined using B3LYP/6-311G+g(2d,p) and the gauge-including atomic orbital (GIAO) method. The calculated chemical shifts are in excellent agreement with experimental values. The performance of a variety of NMR methods (SGO, IGAIM, CSGT) were also examined but were found to be inferior to the GIAO method at the chosen level of theory employed. The theoretical method has been utilized to predict the beryllium chemical shifts of structurally characterized complexes for which no measured (9)Be NMR spectrum exists, and to investigate a literature complex with an unusual (9)Be NMR chemical shift. A new standard for beryllium NMR in nonaqueous solvents has been suggested.

Structural changes upon reduction of dipyrido[2,3-a:3',2'-c]phenazine probed by vibrational spectroscopy, ab initio calculations, and deuteration studies.

A series of bridging ligands, dipyrido[2,3-a:3',2'-c]phenazine (ppb), dipyrido[2,3-a:3',2'-c]-6,7-dichlorophenazine (ppbCl2), and dipyrido[2,3-a:3',2'-c]-6,7-dimethylphenazine (ppbMe2), and their binuclear copper(I) complexes have been synthesized, and their spectral properties were measured. The single-crystal structure of the complex, [(PPh3)2Cu(mu-ppbCl2)Cu(PPh3)2](BF4)2 in the monoclinic space group P21/c, 18.2590(1), 21.1833(3), 23.2960(3) A with Z = 4 is reported. The copper(I) complexes are deeply colored through MLCT transitions in the visible region. The vibrational spectra of the ligands have been modeled using ab initio hybrid density functional theory (DFT) methods (B3LYP/6-31G(d)) and compared to experimental FT-Raman and IR data. The DFT calculations are used to interpret the resonance Raman spectra, and thus the electronic spectra, of the complexes. The preferential enhancement of modes associated with the phenanthroline section of the ligands with blue excitation (lambda(exc) = 457.9 nm) over phenazine-based modes with redder excitation (lambda(exc) = 514.5 and 632.8 nm) suggests the 2 MLCT transitions terminated on different unoccupied MOs are present under the visible absorption envelope. The radical anion species of the ligands are prepared by the electrochemical reduction of the binuclear copper(I) complexes; no evidence of dechelation prevalent in other copper(I) complexes is observed. The resonance Raman spectra of the reduced complexes are dramatically different from those of the parent species. Across the series common bands are observed at about 1590 and 1570 cm(-1) which do not shift with reduction but are altered in intensity. The normal-mode analysis of the radical anion species suggests that these normal modes primarily involve bond length distortions that are unaffected by reduction.

Metallation effects on the thermal interconversion of atropisomers of di(orthomethylarene)-substituted porphyrins.

A new series of meso-substituted diaryl free-base and metalloporphyrins have been prepared. Each arene has been substituted with both a methyl group in the ortho position and a formyl group in the meta position. Rotation of the arene units is prevented at room temperature due to the steric restrictions imposed by the flanking methyl groups at the porphyrin beta-pyrrolic positions on the methyl groups at the ortho position on the meso-substituted arene unit. This allowed the alpha alpha and alpha beta atropisomers of this porphyrin to be separated and characterised. X-Ray crystallographic determination of the structure of the free-base porphyrin revealed a very flat porphyrin core. Metallation resulted in the isolation and characterisation of the nickel, zinc and copper derivatives. The assignments of the alpha alpha and alpha beta isomers are confirmed by X-ray crystallographic determination of the structures of the Cu(II) analogues. The copper alpha alpha structure exhibits a very twisted porphyrin core, the copper alpha beta structure is also distorted, but to a lesser degree. The activation energy for rotation has been calculated for each of the 2H, Ni and Zn derivatives. The energy required to rotate the arene ring increases in the order Ni < Zn approximately 2H. No significant difference in the free energy of rotation was observed between experiments carried out with the alpha alpha and small alpha beta isomers.

Toward functionalized conducting polymers: synthesis and characterization of novel beta-(styryl)terthiophenes.

Metal-catalyzed coupling methodologies have been employed in the synthesis of the key building block 3'-formyl-2,2':5',2"-terthiophene. Wittig olefinations with this aldehyde have produced five novel beta-styryl-substituted terthiophene monomers. These materials have been fully characterized by NMR spectroscopy, microanalysis, mass spectrometry, and X-ray crystal structure analysis. The results from the UV/visible spectroscopy and cyclic voltammetric investigations are reported.

7-Bromoquinolin-8-ol.

Structure analysis of the title compound, C(9)H(6)BrNO, has established that bromination of an 8-hydroxyquinoline derivative occurred in the 7-position. Intermolecular and weak intramolecular O-H.N hydrogen bonds are present, the former causing the molecules to pack as hydrogen-bonded dimers in the solid state.
