Frequency of chromosome aberrations induced by trimethyltin chloride in human peripheral blood lymphocytes in vitro: related to age of donors.

Human peripheral blood lymphocytes of healthy male and female individuals of different age groups were treated with two aqueous doses (0.5 microgram and 1.0 microgram) of trimethyltin chloride in mitogen stimulated and serum supplemented culture medium for 72 h at 37 degrees C. Chromatid and chromosome types of aberrations were observed to be increased in both treated sets in all age groups. Significant variations were observed between age groups (P less than 0.001) and between experimental sets (P less than 0.001). Moreover, interaction of chemical and donors age was statistically highly significant (P less than 0.05-P less than 0.001). However, no linear correlation between the increase of donor's age and aberrations was observed.

Effects of culture media on spontaneous incidence of mitotic index, chromosomal aberrations, micronucleus counts, sister chromatid exchanges and cell cycle kinetics in peripheral blood lymphocytes of male and female donors.

The spontaneous incidence of mitotic index (MI), chromosomal aberrations (CA), micronucleus counts (MNC), sister chromatid exchanges (SCE) and cell cycle kinetics (CCK) were studied in human peripheral blood lymphocytes grown in M199 and RPMI-1640 culture media. Lower frequencies of CAs, MNC and SCEs were observed in lymphocytes cultured in medium RPMI-1640. The reduction of the MI and the replicative index in M199 medium showed delayed cell cycle kinetics.

Frequency of micronuclei induced in peripheral lymphocytes by trimethyltin chloride.

The clastogenicity of trimethyltin chloride was evaluated in human peripheral blood lymphocytes with micronucleus counts (MNC) as the endpoint. Two concentrations (0.5 micrograms and 1.0 microgram) of trimethyltin chloride were added to lymphocytes of healthy male and female subjects of different age groups, in mitogen-stimulated and serum-supplemented culture medium (RPMI 1640, Gibco) for 48 h at 37 degrees C. A significant increase in micronucleus counts was observed with both doses, which was more pronounced with the lower dose. ANOVA in male and female donors revealed significant differences between age groups (P less than 0.001), chemical concentrations (P less than 0.001) and for the interaction of these 2 factors (P less than 0.05 in females only). However, no regular increase or decrease in MNC frequencies was observed with the donor's age. Higher frequencies of MNC enhancement were observed in male individuals than in females.

Size variation of mastoid air cell system in Indian people at different age groups: a radiographic planimetric study.

A radiographic planimetric study of mastoid air cell system was carried out on 100 normal human subjects of which 50 were males and 50 females--who were further subdivided into five age groups. They had no history of past ear disease or any other clinical ENT abnormality. The mean area of the mastoid air cell system was measured planimetrically on X-rays and the data analysed according to age and sex. It was 12.05 +/- 0.67 cm2 in males and 11.45 +/- 0.70 cm2 in females (which are more or less the same as that of Western people). The size of the cranial bones has no apparent role in the size variation of the mastoid air cell system. The development of the latter was very rapid up to 10 years of age in both sexes and it continued even after 20 years of age but at a much slower rate.

Cytogenetic studies in human populations exposed to gas leak at Bhopal, India.

Frequencies of chromosomal abnormalities, sister chromatid exchanges, and replicative index were assessed following peripheral lymphocyte culture in 129 individuals from Bhopal, India. Of these, 83 persons (40 male and 43 female) had been exposed directly to the methyl isocyanate (MIC) gas after the accident at the Union Carbide plant on December 2 and 3, 1984. The remaining 46 samples were taken from age-matched unexposed persons in the same city. Chromosome aberrations were recorded at first cycle metaphase (M1) and sister chromatid exchanges, at second cycle metaphase (M2), following standard schedule. The frequency of chromosomal aberrations was, in general, higher in individuals from the exposed populations, with the females showing a higher incidence. Nondisjunction of chromosomes or laggard was rare. The frequencies of sister chromatid exchanges and depression in mitotic and replicative indices could not be related to exposure or sex. The persistence of chromosomal abnormalities in the form of replicating minutes and exchange configurations, even 1114 days after exposure to the gas, may indicate a residual effect on T-cell precursors.

Frequency of sister chromatid exchanges induced by trimethyltin chloride in human peripheral blood lymphocytes as related to age of donors. A brief report.

Two concentrations (0.5 microgram and 1.0 microgram) of trimethyltin chloride were added to lymphocytes of healthy male and female donors of different age groups and cultured at 37 degrees C for 72 h. The range and mean number of sister chromatid exchanges were significantly increased in the treated cells but could not be related to age or sex of the donor.

Cytotoxic effects of trimethyltin chloride on human peripheral blood lymphocytes in vitro.

Trimethyltin chloride was found to induce cytotoxic damage in vitro in human peripheral blood lymphocytes. Two concentrations (0.5 micrograms and 1.0 micrograms) were added to lymphocytes from male and female subjects in mitogen stimulated and serum supplemented culture medium for 72 h. A dose-related increase of inhibition of replication index (P less than 0.01) and cell division (P less than 0.001) was observed. The frequencies of abnormal cells and chromosomal aberrations such as chromatid and chromosome breaks, dicentrics, triradial and quadriradial configurations were increased significantly (P less than 0.001), as were micronucleus counts (P less than 0.001) and sister chromatid exchanges (P less than 0.001). Endoreduplication, an extremely rare spontaneous event in human lymphocytes, was observed in a few cases at all dose levels. The effects were more pronounced in lymphocytes obtained from habitual smokers.

Effects of stannic chloride on human leucocytes in vitro.

Single dose treatment with stannic chloride (4 micrograms/ml) on human lymphocytes in vitro revealed a significant increase of chromosomal aberrations. These showed a distinct relationship with the donor's age. Single and isochromatid breaks including gaps, premature chromosome condensation, irregular staining, stretching of the centromere and interchange, i.e. quadriradial exchange of chromosome arms, and also aneuploidy, were observed.

Production of methionine and glutamic acid from n-alkanes by Serratia marcescens var. kiliensis.

A hydrocarbon-utilizing Serratia marcescens var. kiliensis grew and accumulated methionine and glutamic acid in a synthetic medium with hydrocarbon as sole carbon source. n-Hexadecane and ammonium phosphate were found as the most suitable carbon and nitrogen sources, respectively. Optimum pH for growth and methionine production was 7.2, and that for glutamic acid accumulation was 7.4. Yeast extract significantly stimulated growth and amino acid production and could be substituted by cyanocobalamine. Benzylpenicillin, Tween 80, SDS or EDTA did not increase amino acid production. Under optimal cultural conditions in the laboratory the organism produced 1.68 g of glutamic acid and 0.78 g of methionine per litre.

Effects of emetine on the metabolism of ascorbic acid.

Treatment with emetine lowered the ascorbic acid concentrations of serum, liver and kidney, while the ascorbic acid concentration of adrenal tissue remained unaffected. The ability of the liver to synthesize (-)-ascorbic acid from (+)-glucuronolactone was also reduced after emetine treatment. The reduced concentration of ascorbic acid in liver and serum after emetine treatment may result from the diminished synthesis of ascorbic acid by the liver.
