Changes in the contents of fat-soluble vitamins and provitamins during tempe fermentation.

The content and formation of fat-soluble vitamins and provitamins during tempe fermentation was studied with 14 different strains of Rhizopus sp. All strains investigated form carotenoids in small amounts during fermentation, but beta-carotene formation was only found in fermentations with six Rhizopus strains. The highest increase of beta-carotene production was to be seen from 34 to 48 h of fermentation. Soybean seeds did not contain ergosterol in detectable amounts. Ergosterol was produced by all Rhizopus strains in concentrations of up to 750 microg g(-1) tempe dry weight (dw) during a 34 h fermentation period and of up to 1610 microg g(-1) tempe dw during a 96 h fermentation period. The amounts of gamma-tocopherol in soybeans ranged from 192.4 to 231.8 microg g(-1) dw. Soybean seeds contained only free, not esterified tocopherols. During fermentation the total amount of vitamin E remained constant but the content of free tocopherols decreased. The content of vitamin K1 in soybeans is not strongly effected by tempe fermentation with pure cultures of Rhizopus sp.

Formation of B-vitamins by bacteria during the soaking process of soybeans for tempe fermentation.

The formation of B-vitamins (nicotinic acid and nicotinamide, thiamine, vitamin B6 and vitamin B12) during the soaking of soybeans by bacteria, isolated from tempe, was investigated. Among the isolates examined no vitamin B6 producer was found. After inoculation of the soaking soybeans with Citrobacter freundii, Klebsiella pneumoniae, Pseudomonas fluorescens and Streptococcus spp. the concentrations of vitamin B12 increased significantly. For the first time it was found that soakings inoculated with C. freundii showed an increased vitamin B12 content. Nicotinic acid and nicotinamide were produced by Lactobacillus spp. and C. freundii. C. freundii synthesized thiamine as well.

Vitamin B12 production by Citrobacter freundii or Klebsiella pneumoniae during tempeh fermentation and proof of enterotoxin absence by PCR.

The influence of some fermentation parameters on vitamin B12 formation by strains of Citrobacter freundii and Klebsiella pneumoniae isolated from Indonesian tempeh samples during tempeh fermentation was investigated. A decrease in fermentation temperature from 32 to 24 degrees C led to a decrease in vitamin B12 formation. Inoculation of soybeans with different numbers of cells of C. freundii at the beginning of solid-substrate fermentation showed that only the velocity of vitamin formation and not the final amount of vitamin formed depended on the number of cells. The addition of cobalt and 5,6-dimethylbenzimidazole increased the vitamin B12 content of tempeh. Nevertheless, levels of incorporation of the two precursors into the vitamin B12 molecule were very low. Neither C. freundii nor K. pneumoniae possessed the genes encoding the enterotoxins Shiga-like toxin SLT IIA, heat-labile enterotoxin LT Ih, and heat-stable enterotoxin ST Ih, as indicated by PCR. This result supports the suggested use of these two strains to form vitamin B12 during tempeh fermentation in Indonesia.

Tempe fermentation: some aspects of formation of gamma-linolenic acid, proteases and vitamins.

During a tempe fermentation the concentrations of linoleic, and alpha-linolenic acids (ALA) decreased while the concentration of oleic acid increased. During fatty acid synthesis Rhizopus sp. produced only gamma-linolenic acid (GLA) instead of ALA. The amount of GLA in tempe were influenced by varying external parameters. The proteolytic capacity of 36 strains of the genus Rhizopus isolated from Indonesian tempe or tempe inocula was examined. There was a distinct increase in the amount of free amino acids during tempe fermentation. Fermentations with mixed populations of bacteria and Rhizopus yielded a lower level of free amino acids, but an increase in total amount of amino acids. In comparison to intracellular, and extracellular proteases the proteases of the cell wall fraction are most responsible for proteolytic capacity of the different Rhizopus strains. Two isolated strains of Citrobacter freundii were found to be the best vitamin B(12) producers during the soaking of soybeans. In the solid substrate fermentation the Rhizopus molds formed vitamin B(6), riboflavin, and nicotinic acid. The addition of bacteria to the solid substrate fermentation resulted in a strong increase of active vitamin B(12) in tempe. In the presence of the Rhizopus mold, the vitamin B(12) formation by C. freundii was three times higher than that of a fermentation without the mold.

Formation of vitamins by pure cultures of tempe moulds and bacteria during the tempe solid substrate fermentation.

The formation of water soluble vitamins (vitamin B12, vitamin B6, riboflavin, thiamine, nicotinic acid and nicotinamide) during the tempe solid substrate fermentation was investigated. The role of several strains of Rhizopus oligosporus, R. arrhizus, and R. stolonifer and the role of several bacteria in the vitamin formation process were checked. All fungal and bacterial strains were isolated from Indonesian tempe and soaking water samples. The Rhizopus strains formed riboflavin, nicotinic acid, nicotinamide and vitamin B6. The final concentrations of these substances depended on the different strains involved and on the fermentation time. Isolates of R. oligosporus were generally the best vitamin formers. The moulds did not produce physiologically active vitamin B12. The thiamine content decreased during fermentation. The addition of bacteria, which had been selected in a screening for vitamin B12 production, resulted in an increase of physiologically active vitamin B12. Citrobacter freundii and Klebsiella pneumoniae showed the best formation capabilities. Furthermore, the bacteria produced riboflavin and vitamin B6 in addition to the moulds. The influence of Rhizopus on the vitamin B12 formation of Cit. freundii was also investigated. The vitamin content of tempe that was fermented with the mould and the bacterium was three times as high as a control fermentation with Cit. freundii only.

Multistep reactions with immobilized microorganisms.

This review presents some examples of new semicontinuous and continuous processes for product formation and for degradation of xenobiotics with immobilized microorganisms. A semicontinuous process for glycerol formation with Saccharomyces cerevisiae cells, adsorbed on sintered glass, and a continuous production of citric acid with Aspergillus niger, entrapped in calcium alginate, are given as examples of the production of primary metabolites. The production of ergot alkaloids with entrapped Claviceps purpurea demonstrates the possibilities of production of secondary metabolites. The continuous degradation of phenolic substances and that of 4-chlorophenol by entrapped and adsorbed microorganisms are given as examples of the possibility of a continuous degradation of xenobiotics by immobilized microorganisms. The continuous degradation of these substances was successful not only in artificial solutions but also in sterile and nonsterile wastewater.

[The value of renal phlebography in the differential diagnosis of arteriographically hypo- or avascular lesions in the kidney (author's transl)].

The present paper is an investigation into the diagnostic value of renal phlebography for differentiating between arteriographically hypo- and avascular malignant lesions in the kidney and benign renal masses. In three examinations carried out on eight patients with renal masses, phlebography was of crucial diagnostic importance. In ten renal cysts a presumptive diagnosis made by arteriography was confirmed by phlebography. In another eight renal cysts, a case of haemorrhagic pyelitis and in two cases of unilateral pseudotumerous pyelonephritis, phlebography was of no value.
