Detection of peptaibols and partial cloning of a putative peptaibol synthetase gene from T. harzianum CECT 2413.

The characterization of 11- and 18-residue peptaibols (peptides synthesized by peptide synthetases) at Trichoderma harzianum CECT 2413 (a filamentous fungus) was performed. Using a heterologous probe from tex1, the only peptaibol synthetase cloned and characterized so far in Trichoderma species, was cloned; a region that comprised 11676 bp of a second peptide synthetase gene detected in these strain (called salps2) and sequenced. The deduced sequence of Salps2 (3891 amino acids) contained three complete and a fourth incomplete module of a peptide synthetase, in which the typical adenylation, thiolation and condensation domains were found, but also an additional dehydrogenase/reductase domain in the C-terminus of the last module. Based on sequence similarity and analysis of its modular structure, it is proposed that Salps2 is a peptaibol synthetase. Additionally, analysis of =4.4-kb sequence downstream of salps2 was done and the signature sequences of Salps2 were identified and compared with those of available sequences of the other Trichoderma peptaibol synthetases.

The in-vitro antimicrobial activities of some medicinal plants from Cameroon.

The antimicrobial activities of 10 plant species (Voacanga africana, Crepis cameroonica, Plagiostyles africana, Crotalaria retusa, Mammea africana, Lophira lanceolata, Ochna afzelii, Ouratea elongata, Ou. flava and Ou. sulcata), each of which is currently used in the traditional medicine of Cameroon, were investigated in vitro. The activities of a methanol extract of each plant were tested, in disc-diffusion assays, against 37 reference or laboratory strains of seven species of microorganism (Staphylococcus aureus, S. epidermidis, Enterococcus hirae, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa and Candida albicans). The minimal inhibitory concentrations of each extract were then estimated, against each of the more susceptible microorganisms (i.e. those giving an inhibition zone measuring at least 9 mm in diameter in the disc-diffusion assays), by agar dilution. Although, in the disc-diffusion assays, each of the 10 methanol extracts investigated displayed some degree of antimicrobial activity against at least one species of microorganism, no activity against the Gram-negative bacteria (Es. coli, K. pneumoniae and Ps. aeruginosa) was observed. The extract with the greatest antimicrobial activity was that of Pl. africana (Euphorbiaceae).

New pigments from the terrestrial cyanobacterium Scytonema sp. collected on the Mitaraka inselberg, French Guyana.

Inselbergs are hills rising abruptly from the surrounding plains where cyanobacteria are the only living organisms under conditions of intense solar radiation. A survival mechanism to prevent UV-damage has been associated with synthesis of the ultraviolet-screening, photostable sheath pigment scytonemin. The organic extract of Scytonema sp., collected on the Mitaraka inselberg, French Guyana, yielded three new pigments, tetramethoxyscytonemin (1), dimethoxyscytonemin (2), and scytonine (3), derived from the scytoneman skeleton of scytonemin. These structures were assigned mainly on the basis of (1)H and (13)C NMR and MS experiments.

Cytotoxic and antimicrobial coumarins from Mammea africana.

Six coumarin derivatives [three 4-phenylcoumarins (Mammea A/AA, Mammea A/BA and MAB 3), two 4-n-propylcoumarins (Mammea B/BB and Mammea B/BA) and one 4-n-pentylcoumarin (Mammea C/OB)], 1,5-dihydroxyxanthone and 1-methoxy-5-hydroxyxanthone have been isolated from the stem bark of Mammea africana Sabine collected in Cameroon. Although known, the structures of the coumarin derivatives were confirmed by spectral analysis, including two-dimensional nuclear magnetic resonance. All the coumarin compounds showed noteworthy cytotoxicity against the human 9-KB cell line. Both of the 4-n-propylcoumarins were also found to exhibit significant activity against Staphylococcus aureus.

Identification of an anti-inflammatory principle from the stem bark of Millettia versicolor.

The dichloromethane-soluble fraction of the methanol extract of the stem bark of Millettia versicolor Welw. (Leguminosae) has been shown to possess anti-inflammatory activity. The chromatographic fractionation and subsequent analysis of the spectroscopic data of this extract led to the isolation and identification of 2-acetyl-7-methoxynaphtho[2,3- b]furan-4,9-quinone (1) along with two known quinones. Pharmacological data demonstrate that compound 1 has relevant anti-inflammatory properties whereas the other two isolated compounds do not.

A new biflavonoid from Ochna beddomei.

A new biflavonoid, 2,3-dihydroochnaflavone 7,4',7"-tri-O-methyl ether (1) together with two known biflavonoids namely, 2,3-dihydroochnaflavone (2) and ochnaflavone (3) were isolated from the stem bark of Ochna beddomei. The structures were determined by means of spectral and chemical studies.

Flavones from Andrographis viscosula.

Two new 2'-oxygenated flavones, 5,7,2'-trimethoxyflavone (1). and 5,7,2',4',6'-pentamethoxyflavone (2). were isolated from the whole plant of Andrographis viscosula along with three known flavones, echioidinin (3). 5,2',6'-trihydroxy-7-methoxyflavone (4). and echioidin (5). The structures of these compounds were elucidated on the basis of 1D and 2D NMR spectral studies.

Biflavonoids from Ochna afzelii.

The stem bark of Ochna afzelii furnished three biflavonoids among which two are new. Their structures were established from spectroscopic and chemical evidences.

Two new fatty acid glucosides from the root bark of Ochna calodendron.

Two new fatty acid glucosides, (3E,8Z)-8-carboxy-1-(O-beta-D-glucopyranosyl)-2,6-dihydroxytritriaconta-3,8-diene (1) and (8Z)-8-carboxy-1-(O-beta-D-glucopyranosyl)-3,4-epoxy-2,6-dihydroxytritriacont-8-ene (2), have been isolated from the root bark of Ochna calodendron and their structures elucidated by extensive two-dimensional NMR methods including HMQC, HMBC and NOESY experiments.

Sequences and antimycoplasmic properties of longibrachins LGB II and LGB III, two novel 20-residue peptaibols from Trichoderma longibrachiatum.

Longibrachins are members of the class of natural Aib-containing peptides designated as peptaibols. Six longibrachins, LGA I-IV and LGB II and III, were purified from a Trichoderma longibrachiatum strain by a procedure employing several chromatography steps including reversed-phase HPLC. The amino acid sequence determination was based on a combination of liquid secondary ion mass spectrometry (LSIMS) and two-dimensional 1H and 13C NMR spectroscopy. Longibrachins are 20-residue peptaibols with a C-terminal phenylalaninol and either neutral (LGA; Gln18) or acidic (LGB; Glu18) character. Longibrachins LGB II and III have novel sequences. Both longibrachins LGA and LGB show significant bactericidal activity against mycoplasmas (Acholeplasma, Mycoplasma, and Spiroplasma), with minimal inhibitory concentrations in the range 1.56-12.5 microM (3-25 micrograms/mL), and also perturb the permeability of membrane bilayers. Longibrachin LGA IV is the most potent of the presently known 18-20-residue peptaibols. The antimicrobial and membrane-perturbing properties of longibrachins, which are described here for the first time, were shown to be correlated.

Androechin, a new chalcone glucoside from Andrographis echioides.

A new chalcone glucoside, androechin, and a known flavone glucoside, echioidinin 5-O-glucoside, were isolated from the whole plant of Andrographis echioides. Androechin was characterized as 2,2',6'-trihydroxy-4'-methoxychalcone 2'-O-beta-D-glucopyranoside by spectral and chemical studies.

Two new 2'-oxygenated flavones from Andrographis elongata.

Two new 2'-oxygenated flavones, 5,2',6'-trihydroxy-7-methoxyflavone (3) and skullcapflavone I 2'-O-beta-D-(4"-E-cinnamyl) glucopyranoside (5), together with three known flavones, 7-O-methylwogonin (1), skullcapflavone I (2) and skullcapflavone I 2'-O-beta-D-glucopyranoside (4) were isolated from the whole plant of Andrographis elongata, and the structures were elucidated by FAB-MS and one- and two-dimensional (1D- and 2D)-NMR spectral studies including 1H-1H correlation spectroscopy (COSY), heteronuclear single quantum coherence (HSQC), heteronuclear multiple bond connectivity (HMBC) and rotating frame Overhauser enhancement spectroscopy (ROESY) experiments, and chemical studies.

Two unprecedented natural Aib-peptides with the (Xaa-Yaa-Aib-Pro) motif and an unusual C-terminus: structures, membrane-modifying and antibacterial properties of pseudokonins KL III and KL VI from the fungus Trichoderma pseudokoningii.

Pseudokonins KL III and KL VI are two natural ten-residue peptides, which both contain the (Xaa-Yaa-Aib-Pro) motif and exhibit an unusual C-terminus. They have been isolated from the fungus Trichoderma pseudokoningii by intensive reversed-phase HPLC, beside peptaibols classically C-ended by a beta-amino alcohol. The amino acid sequences and the chemical structures of the C-ends have been determined by the combined use of positive ion LSI-MS and two-dimensional homo- and heteronuclear NMR, including COSY, TOCSY, ROESY, 13C heteronuclear single quantum correlation (HSQC) and heteronuclear multiple bond correlation (HMBC). Instead of one of the amino alcohols usually found as C-terminal residue in peptaibols, pseudokonins KL III and KL VI are characterized by -Pro-NH2 and cyclo-(Aib-L-Proal) (Proal, prolinal), respectively. Such backbone modifications are described here for the first time for peptaibol antibiotics. The unusual cyclo-(Aib-L-Proal) C-terminus is probably the result of an intramolecular cyclization of the two last Aib and Pro residues of a ten-amino acid precursor, via a Proal intermediate. A secondary structure stabilized by -C=O...H-N-hydrogen bonds of the 1<--4 type has been deduced for both peptides from ROESY data, 3JNHCalphaH couplings and amide proton temperature coefficient values. The (Xaa-Yaa-Aib-Pro) beta-bend ribbon spiral, which has been described for the first time in the case of a 14-residue peptaibol containing three repetitive (Xaa-Yaa-Aib-Pro) motifs (Segalas G et al. Biopolymers 1999; 50: 71-85) appears to be maintained in the two shortened modified peptides. The beta-bend ribbon structure thus appears to be initiated by a single (Xaa-Yaa-Aib-Pro) motif and unaffected by the C-terminal modifications. However, the membrane and antibiotic properties of pseudokonins KL III and KL VI, point to the unfavourable effect of both shortening and cyclization of the peptide chain.

A biflavanone from Cycas beddomei.

A new biflavanone, 7,7"-di-O-methyltetrahydrohinokiflavone together with tetrahydrohinokiflavone were isolated from the stems of Cycas beddomei. The structures were established on the basis of spectral and chemical studies.

Indicanines B and C, two isoflavonoid derivatives from the root bark of Erythrina indica.

In addition to two known compounds, 5,4'-di-O-methylalpinumisoflavone and cajanin, a new 3-phenylcoumarin metabolite, named indicanine B, and a new isoflavone derivative, named indicanine C, were isolated from the root bark of Erythrina indica. By means of spectroscopic analysis, the structures of the new compounds were characterized as 4-hydroxy-3-(4'-hydroxyphenyl)-5-methoxy-2",2"-dimethylpyrano [5",6":6,7] coumarin and 4'-hydroxy-5-methoxy-2",2"-dimethylpyrano [5",6":6,7] isoflavone, respectively. The 13C-NMR data of cajanin and the in vitro antimicrobial spectrum and potencies of the isolated compounds are also reported.

7-O-Methyltetrahydroochnaflavone, a new biflavanone from Ochna beddomei.

7-O-Methyltetrahydroochnaflavone (1), a new biflavanone, together with nine known flavonoids, afrormosin (2), 2,3-dihydroochnaflavone 7-O-methyl ether (3), kaempferol (4), 2,3-dihydroochnaflavone (5), ochnaflavone (6), (-)-epicatechin (7), kaempferol 3-O-rhamnoside (8), taxifolin 3-O-rhamnoside (9), and kaempferol 3-O-glucoside (10), were isolated from the leaves of Ochna beddomei, and the structures were elucidated by spectral and chemical studies.

Indicanine A, a new 3-phenylcoumarin from root bark of Erythrina indica.

A new 3-phenylcoumarin, indicanine A (1), has been isolated from the root bark of the African medicinal plant Erythrina indica, together with three known compounds, robustic acid (2), daidzein, and 8-prenyldaidzein. The structure of the new compound was characterized, as 4-hydroxy-5-methoxy-3-(4'-methoxyphenyl)-2" -(1-methylethenyl)dihydrofurano[4",5":6,7]coumarin by means of extensive spectroscopic analyses. The compounds were found to be devoid of in vitro antibacterial activity.

The ion-channel activity of longibrachins LGA I and LGB II: effects of pro-2/Ala and gln-18/Glu substitutions on the alamethicin voltage-gated membrane channels.

Longibrachins LGA I (Ac Aib Ala Aib Ala Aib(5) Ala Gln Aib Val Aib(10) Gly Leu Aib Pro Val(15) Aib Aib Gln Gln Pheol(20), with Aib: alpha-aminoisobutyric acid, pheol: phenylalaninol) and LGB II are two homologous 20-residue long-sequence peptaibols isolated from the fungus Trichoderma longibrachiatum that differ between them by a Gln-18/Glu substitution. They distinguish from alamethicin by a Pro-2 for Ala replacement, which allowed to examine for the first time with natural Aib-containing analogues, the effect of Pro-2 on the ion-channel properties exhibited by alamethicin. The influence of these structural modifications on the voltage-gated ion-channel forming activity of the peptides in planar lipid bilayers were analysed. The general 'barrel-stave' model of ion-channel activity, already described for alamethicin, was preserved with both longibrachins. The negatively charged LGB II promoted higher oligomerisation levels, which could presumably dilute the repulsive effect of the negative Glu ring near the entrance of the channel and resulted in lower lifetimes of the substates, confirming the strong anchor of the peptide C-terminus at the cis-interface. Reduction of the channel lifetimes was observed for the longibrachins, compared to alamethicin. This argues for a better stabilisation of the channels formed by peptaibols having a proline at position 2, which results in better anchoring of the peptide monomer N-terminus at the trans-bilayer interface. Qualitative assays of the temperature dependence on the neutral longibrachin channel properties demonstrated a high increase of channel lifetimes and a markedly reduced voltage-sensitivity when the temperature was decreased, showing that such conditions may allow to study the channel-forming properties of peptides leading to fast current fluctuations.
