Comparative biochemical and genetic studies of the GOX-1 (HAO-1) and the XDH-1 polymorphism in the rat (Rattus norvegicus).

Biochemical and genetic studies have been carried out to test whether the GOX-1 (HAO-1) and the XDH-1 polymorphism in the rat are determined by one locus or by two different closely linked loci. Besides similar migration behaviour upon electrophoresis the phenotypes of both polymorphisms share major biochemical properties such as responses to heat inactivation at 50 and 70 degrees C and isoelectric points. This suggests that they represent only one enzyme. The one-enzyme hypothesis is supported by linkage analyses performed in 50 offspring of a backcross population [(SPRD-Cu3 x BN)F1 x SPRD Cu3] and 14 LXB recombinant inbred strains (progenitor strains LEW and BN) in which not a single recombinational event can be observed. The latter finding agrees well with the GOX-1/XDH-1 distribution pattern investigated in 93 inbred strains and substrains (BENDER et al. 1984). Because of these results and due to a preferential reactivity with glycolic acid we propose to designate this polymorphism only as glycolate oxidase (GOX-1).

Pulse and steady-state labelled and actinomycin D chased 5,6-dichloro-1-beta-D-ribofuranosylbenzimidazole (DRB)-resistant hnRNA from uninfected HeLa cells.

The fraction of hnRNA synthesized in the presence of DRB in uninfected HeLa cells ranges in size from 4S to over 45S. High molecular weight DRB-resistant hnRNA has been demonstrated to decay after 2 h of actinomycin D chase. This fraction is composed of both poly(A)(+) and poly(A)(-) RNA molecules. The synthesis in the presence of DRB of short polyadenylated hnRNA was also observed. The nature of both hnRNA subfractions is discussed.

Stimulatory effect of hnRNA on template activity of isolated rat liver chromatin.

Transcription of chromatin isolated from rat liver with E. coli RNA polymerase was stimulated up to five-fold with heterogeneous nuclear RNA (hnRNA) from rat liver. The transcription product had features of DNA-primed RNA. Neither cytoplasmic poly(A)-containing RNA nor high or low molecular weight cytoplasmic poly(A)-lacking RNA from rat liver exhibited stimulation of transcription. The stimulatory effect seems to be confined to middle repetitive sequences of hnRNA. With purified DNA templates, addition of hnRNA resulted in complete inhibition of transcription. The stimulatory effect of hnRNA on chromatin transcription was also observed with endogenous RNA polymerase. This effect was dependent ionic strength and was most pronounced under conditions optimal for RNA-polymerase B activity.

Comparison of cytoplasmic RNA species from rat liver and Morris hepatoma 5123D.

1. In a highly differentiated Morris hepatoma 5123D the fraction of repetitive, cytoplasmic poly(A)-containing RNA hybridizing with DNA to the Cot values below 10(2) mol X sec/l is present in a higher proportion than in normal rat liver. 2. Unique sequences of cytoplasmic RNA containing poly(A) both in Morris hepatoma and normal rat liver exhibit similar hybridization kinetics. 3. No sequence differences in cytoplasmic poly(A)-free RNA between Morris hepatoma 5123D and normal rat liver were found using hybridization-competition technique.

Isolation and properties of poly(A)-containing RNA from the cytoplasm of rat liver cells.

1. The described technique for preparative isolation of poly(A)-containing RNA by complexing with poly(U)-cellulose and elution at 45 degrees C, is simple, reproducible, and the poly(U)-cellulose preparation is suitable for repeated use. 2. The poly(A)-containing RNA isolated from the poly(U)-cellulose complex is still contaminated by ribosomal RNA which can be removed by recomplexing. The purified preparation is heterogeneous on polyacrylamide-gel electrophoresis and is located over the region from 5-6S to 28S. Poly(A) segments of cytoplasmic RNA show heterogeneous size distribution. 3. In acidic medium, the poly(A) segments form ordered double-stranded structure. 4. The rapid labelling of rat liver and fibroblast cytoplasmic poly(A)-containing RNAs and their base composition resemble closely the corresponding properties of mRNA.
