Cryptosporidium muris: prevalence, persistency, and detrimental effect on milk production in a drylot dairy.

A total of 1746 individual fecal samples were obtained from milking cows during three separate visits to a drylot dairy farm. In addition, 1240 fecal samples were also obtained from cows in four additional farms. Cryptosporidium muris was prevalent in all five herds sampled. Cows that were sampled more than once invariably remained in the same shedding category. Cows shedding C. muris oocysts produced significantly less milk (approximately 3.2 kg/d). After corrections for the effects of age, parity, pen, and DIM in a logistic regression model, mean daily milk production was significantly associated with shedding status.

Ovine lentivirus expression and disease. Virus replication, but not entry, is restricted to macrophages of specific tissues.

To better define the relationship between lentivirus infection and lymphoproliferative or inflammatory disease, we studied postmortem specimens of 38 sheep naturally infected with ovine lentivirus (OvLV) and with different clinical manifestations of OvLV-associated disease. Immunohistochemistry, in situ hybridization, and virus isolation were used to localize viral protein, viral RNA, and infectious virus to specific cells and tissues. Viral protein or infectious virus was found in cells morphologically and histochemically compatible with macrophages (M phi s), but only in lung, bone marrow, mammary gland, lymph node, spleen, synovium, brain, and spinal cord, frequently in association with lymphocyte infiltrates. In contrast, viral RNA was found in a variety of cell types, including epithelium, M phi s, and M phi-like cells, and in a wider range of tissues, with or without OvLV-associated lesions. In summary, these findings suggest that in vivo: 1), OvLV can enter a variety of cell types, 2), productive infection is restricted to cells of M phi lineage, and 3), cells expressing viral proteins are limited to specific tissues, those associated with OvLV-induced diseases.

Immunochemiluminescent Southern blot assay for polymerase chain reaction detection of human parvovirus B19 DNA.

Human parvovirus B19 is not only an acute self-limited infection causing erythema infectiosum, transient aplastic crisis, foetal hydrops and arthritis but can also be a chronic infection causing chronic anaemia and associated with chronic neuropathy and vasculitis. Serologic studies have proven to be the most sensitive way to detect acute infection in the immunologically normal patient while polymerase chain reaction (PCR) assays for B19 DNA are the most sensitive way to detect chronic infection. The ability to detect B19 in clinical specimens can be further increased with a second amplification step using nested primers. However, nested PCR is both time consuming and enhances the risk of false-positive results due to contaminating DNA. In this study, we developed a sensitive immunochemiluminescent Southern blot assay for detecting PCR amplified B19 DNA with a digoxigenin labelled primer. The sensitivity and specificity of this assay were comparable to nested PCR and at least 100-fold more sensitive than a single PCR amplification.

Hematologic, serologic values, histopathologic and fecal evaluations of bison from Yellowstone Park.

Hematologic and blood chemistry parameters were measured in 149 free-ranging American bison (Bison bison) from Yellowstone National Park, Wyoming (USA). Additionally, histopathologic evaluations of lung, liver, spleen, kidney, and mesenteric and bronchial lymph nodes were made from ten animals. Forty-five fecal samples were screened for the presence of helminth ova. Leukopenia and markedly low blood urea nitrogen concentrations were the most notable differences observed from other bison populations. All tissues examined were essentially normal; there was evidence of moderate intestinal parasite burdens.

Effects of virus load in the pathogenesis of lentivirus-induced lymphoid interstitial pneumonia.

To better define the relationship between ovine lentivirus (OvLV) infection and respiratory disease, pulmonary leukocytes and postmortem lung specimens from 42 sheep seropositive or at risk for OvLV infection were obtained. The lungs were examined for lesions of lymphoid interstitial pneumonia (LIP), and animals were categorized into five groups by severity of LIP and OvLV serologic status. The presence of OvLV in alveolar macrophages was established by proviral DNA amplification using the polymerase chain reaction (PCR), and the proportion of infected cells was determined by a quantitative focal immunoassay (FIA) and by immunohistochemistry. The concentration of OvLV p25 in serum was measured by capture ELISA. In contrast to animals with mild or no pulmonary lesions, sheep with moderate or severe LIP (17/42) were all seropositive, 71% had antigenemia (greater than 2 ng/mL), and 82% had proviral DNA in 1.5 x 10(5) alveolar macrophages. Of sheep positive by PCR, those with moderate or severe LIP (79%) had an average of 3 infected cells/10(3) alveolar macrophages by FIA. These results implicate alveolar macrophages as important target cells in the pathogenesis of OvLV-induced respiratory diseases.

Treatment of De Quervain's tenosynovitis with corticosteroids. A prospective study of the response to local injection.

Fifty-six cases of De Quervain's tenosynovitis (in 55 patients) were treated with a "long-acting" corticosteroid, methylprednisolone acetate, and followed prospectively over a 4-year period. Approximately 90% of these patients were effectively managed either with a single injection (58%) or with multiple injections (33%) of this compound. Seventeen patients experienced recurrence a mean of 11.9 months after the initial injection. Three had minor flares and were not reinjected; the others responded to reinjections. Ten percent of the cases could not be controlled with local injection, and these patients were referred for surgery. Adverse reactions were self-limited and relatively minor; no tendon ruptures or local infections occurred. We present a discussion of our review of the literature regarding medical therapy and surgical release for this condition. Treatment of De Quervain's tenosynovitis with methylprednisolone acetate injection rapidly controls the signs and symptoms, does not lead to serious adverse reactions, and should be the preferred initial treatment.

Facilitated isolation, purification, and analysis of glucuronoxylomannan of Cryptococcus neoformans.

Cryptococcus neoformans was cultured in a chemically defined medium. The culture was adjusted to 0.25% formaldehyde or autoclaved after 5 days of growth at 35 degrees C, and a cell-free supernatant was obtained by centrifugation. Solid calcium acetate was added to the supernatant to give a 5% solution, and the pH was adjusted to approximately 5 with glacial acetic acid. The polysaccharide (PS) was precipitated by the addition of 3 volumes of 95% ethanol. The PS was dissolved in 0.2 M NaCl, and insoluble calcium salts were solubilized by the addition of several drops of glacial acetic acid. The PS solution was treated by ultrasonic irradiation for 15 min. This concurrently decreased the molecular weight of the PS and reduced the viscosity of the solution. The ultrasonically irradiated PS was precipitated by differential complexation with hexadecyltrimethylammonium bromide at 23 degrees C, the complex was dissolved in 1 M NaCl, and the glucuronoxylomannan was precipitated by adding 3 volumes of ethanol. The glucuronoxylomannan was dissolved in 1 M NaCl and then ultrasonically irradiated for 2 h to reduce the molecular mass to a limiting value of approximately 100 kDa (GXMS). The purified GXMS was centrifuged, dialyzed, and finally recovered by lyophilization. GXMS was chromatographed on DEAE-cellulose at reasonable concentrations without the complication of high solution viscosity. The sugar composition and structure of GXMS were determined by gas-liquid chromatography, permethylation gas-liquid chromatography-mass spectrometry, and 13C nuclear magnetic resonance spectroscopy. The improved solution characteristics of GXMS were ideal for the determination of its chemical and serological properties.

Identification of a 15-kilodalton surface glycoprotein on sporozoites of Cryptosporidium parvum.

An immunoglobulin A monoclonal antibody (MAb5C3) was developed against a 15-kDa surface glycoprotein (GP15) of Cryptosporidium parvum sporozoites. Indirect immunofluorescence and colloidal gold immunoelectron microscopy revealed that the antibody reacted with both the sporozoite and merozoite surface plasma membranes. On Western immunoblots, MAb5C3 binding was found to be strongly inhibited when 200 mM N-acetylglucosamine was used as a competing sugar. N-Acetylgalactosamine inhibited binding of the antibody only slightly, whereas glucose, mannose, and galactose failed to inhibit binding. MAb5C3 was found to recognize a similar 15-kDa epitope associated with a Cryptosporidium sp. isolated from guinea pigs. However, MAb5C3 failed to react with any proteins or glycoproteins associated with C. baileyi from chickens, Cryptosporidium sp. (= bovine C. muris) from cattle, C. serpentis from a rat snake, bradyzoites of Besnoitia darlingi from an opossum, sporozoite/oocyst extracts of Caryospora bigenetica from an eastern diamondback rattlesnake, sporozoites of Eimeria nieschulzi and E. papillata from rats and mice, or tachyzoites of Toxoplasma gondii (RH strain). When hybridoma supernatants containing MAb5C3 were administered orally to suckling mice experimentally infected with C. parvum, a 75% reduction in developmental stages was seen histologically at 72 h postinfection and a 67.5% reduction in mean oocyst output was found at 6 days postinfection.

Methods for control of lamb epididymitis in large purebred flocks.

An autogenous, multivalent, adjuvanted bacterin for epididymitis was tested in flocks of 700 to 800 Suffolk X white-faced lambs. Long-term feeding of low dosages of antibiotics also was tested in the Suffolk flock. Both methods appeared to reduce the incidence of the disease. The incidence of clinical epididymitis and the number of positive culture results from clinically affected rams were significantly reduced.

Identification of outer oocyst wall proteins of three Cryptosporidium (Apicomplexa: Cryptosporidiidae) species by 125I surface labeling.

Autoradiography of oocyst wall surface proteins of three Cryptosporidium spp. revealed common bands at 285 to 290, 145 to 148, 120, 57, and 32 kilodaltons (kDa). Cryptosporidium baileyi and C. muris share proteins at 180, 100, 80 to 81, 29, and 18 to 19 kDa; C. baileyi and C. parvum share one protein at 46 to 47 kDa; and C. muris and C. parvum share a protein at 67 to 69 kDa. Additional protein bands, each unique to one species, were also observed.

Osteocartilaginous metaplasia of the right atrium in male domestic sheep.

Among 53 clinically healthy male sheep with epididymitis, nine had minimal to severe firm nodular thickening of the right atrial wall at necropsy. The two most severely affected atria were about three times normal size. Histologically, the atrial masses consisted of partly mineralized cartilage nodules with lipid-filled cavities and basophilic to eosinophilic trabeculae. Further investigation failed to reveal clues as to etiopathogenesis of these lesions.

Abomasal cryptosporidiosis in cattle.

A 6-week-old calf and nine feedlot steers shed oocysts similar to Cryptosporidium muris-like oocysts. There were massive populations of this Cryptosporidium in the peptic glands of most of these animals. The oocysts were larger and more oval than the frequently reported type which is generated in the intestines of many animal species and thought to be similar to Cryptosporidium parvum. The pattern of shedding of this newly discovered Cryptosporidium in the steers was continuous over a period of months whereas the C. parvum-like oocysts cease to be shed 1 to several weeks after onset. The nature of the host-parasite interaction in abomasal cryptosporidiosis is yet to be determined. Morphologic changes that resulted from the interaction were an approximate 10% increase in abomasal mucosal thickness, widening of gland lumens in the middle region, and atrophy of epithelium in the same region.

Death associated with parenteral administration of copper disodium edetate in calves.

Copper disodium edetate in recommended doses was apparently responsible for the deaths of one calf and clinical signs of toxicosis in 5 others on one farm, and 7 deaths and clinical signs of toxicosis in a number of others on another ranch. Signs of hyperexcitability, hypermetria, hindlimb weakness, head pressing, depression, and opisthotonos occurred 6 to 24 hours after injections and preceded death by 1 to 2 days. Necropsy and histologic examination revealed massive liver necrosis. High blood concentrations of liver enzymes in affected cattle that did not die indicated that they had liver damage. High blood concentration of iron in cattle that died indicated possible interaction of copper and iron.
