Triterpene glycosides from the seeds of Astragalus sinicus L.

From the seeds of Astragalus sinicus L. (Leguminosae), seven triterpene glycosides were isolated and identified as soyasaponin I-III methyl esters (1-3) which were treated with CH2N2 during the separation procedure, soyasaponin IV (4), soyasapogenol B 3-O-beta-D-glucuronopyranoside (5), 3-O-alpha-L-rhamnopyranosyl(1-->2)-beta-D-xylopyranosyl(1-->2)-beta-D- glucuronopyranosyl 3 beta, 22 beta, 24-trihydroxy-11-oxoolean-12-ene (6), whose sapogenol (8) was obtained by enzymatic hydrolysis using glycyrrhizinic acid hydrolase, unambiguously characterized and designated as complogenin, and 3-O-alpha-L-rhamnopyranosyl(1-->2)-beta-D-galactopyranosyl(1-->2)-beta-D - glucuronopyranosyl complogenin (7).

[Study on protein polymorphism of Chinese KM subcolonies].

This paper presents the results of study on 24 protein markers by means of electrophoresis in 4KM subcolonies maintained in Beijing, Shanghai and Changcun, and compares it with that of one NIH colony introduced from U.S.A. in 1982. The results revealed that: (1) There were few genetic variants on allelic constitutions of Chinese KM mouse colonies, and the genetic distance among KM subcolonies is 0.008-0.027 positively related with the time the colony closed; (2) The unique position of S: KM mouse was shown in phylogenetic diagram of 4 KM subcolonies, which agrees with the result from mandible analysis; (3) The allelic constitutions of KM mice differs from NIH mice a Swiss derivative colony at Es-3, Es-10, Glo-1, Gpt-1, Got-2 and Mpi-1 loci and the average genetic distance between KM and NIH colonies is 0.131 + 0.011, which indicates that Chinese KM mice is one of non-Swiss derivative subspecies.

Arsenic trioxide induces apoptosis in cisplatin-sensitive and -resistant ovarian cancer cell lines.

Arsenic trioxide (As(2)O(3)), has been used for centuries in traditional Chinese medicine; it has considerable efficacy in the treatment of relapsed acute promyelocytic leukemia, inducing partial differentiation and promoting apoptosis of malignant promyelocytes. Although a number of studies have demonstrated that As(2)O(3) has potent activity against cell growth in a series of leukemia cell lines, little information is available regarding this compound's effect on cell growth in solid tumor cell lines. In this study, we investigated the effects of As(2)O(3)in vitro on ovarian cancer cell lines sensitive (3AO) and resistant (3AO/CDDP) to cisplatin. The 3-(4,5-dimethy-thiazoyl-2-yl)-2,5-diphenyl-tetrazolium bromide assay was used to evaluate cytotoxicity. Flow cytometric analysis was used to determine the apoptosis, cell cycle distribution. We clearly demonstrated that As(2)O(3) induced cell apoptosis and inhibition of cell growth in both the cell lines. Furthermore, we identified that As(2)O(3)-induced apoptosis involved Fas pathway. As(2)O(3) is an active agent against ovarian cancer cells and could be effective in the clinical treatment of ovarian cancer.

Discovery of novel inducers of cellular differentiation using HL-60 promyelocytic cells.

Non-physiological inducers of terminal differentiation have been used as novel therapies for the prevention and therapy of cancer. We have used cultured HL-60 promyelocytic cells to monitor differentiation, proliferation and cell death events as induced by a large set of extracts derived from plants. Screening of more than 1400 extracts led to the discovery of 34 with potent activity (ED50 <8 mg/ml). Bioassay-guided fractionation led to the isolation of zapotin and 2',5,6-trimethoxyflavone as active principles from Casimiroa edulis, dibenzyltrisulfide and 2-[(phenylmethyl)dithio]ethanol as active principles from Petiveria alliacea, and desmethylrocaglamide from Aglaia ponapensis. Zapotin demonstrated the most favorable biological profile in that induction of differentiation correlated with proliferation arrest, and a lack of cytotoxicity. We conclude that the HL-60 cell model is a useful system for the discovery of novel pharmacophores with potential to suppress the process of carcinogenesis, and that flavonoids may be especially useful in this capacity.
