Circadian Control of the Accumulation of mRNAs for Light- and Heat-Inducible Chloroplast Proteins in Pea (Pisum sativum L.).

The levels of the mRNAs for light-inducible, nuclear-coded chloroplast proteins vary rhythmically in pea (Pisum sativum L.) plants either grown in a dark-light cycle or under constant light conditions. This has been observed for the early light-inducible protein, the light-harvesting chlorophyll a/b protein, and the small subunit of the ribulose-1,5-bisphosphate carboxylase. The mRNA levels are high in the morning, exhibit a minimum in the first half of the night, and increase again during the second half of the night. The amplitude of fluctuation is between 5- and 10-fold. A similar change in the mRNA abundance was found for four nuclear encoded heat-shock proteins of 18, 24, 26, and 30 kilodaltons. The ability of plants to transcribe heat-shock genes upon heat-shock for 2 hours varies through the day. The maxima for induction are found in the second half of the night and the morning. The minima are reached during the afternoon. The degree of fluctuation is between 3- and 5-fold. The levels of mRNAs for cytosolic as well as for plastid heat-shock proteins oscillate in parallel.

The early light-inducible proteins of barley. Characterization of two families of 2-h-specific nuclear-coded chloroplast proteins.

Within 1-2 h of illumination of etiolated barley plants the mRNAs of seven nuclear-coded proteins are transiently induced. It is proposed that at least some of these proteins are precursors to chloroplast membrane proteins since after posttranslational transport 2-h-specific bands of 18.5 kDa, 18 kDa and 13.5 kDa have been found bound to thylakoid membranes. cDNA clones for these early light-inducible proteins (ELIPs) have been isolated. Hybrid-release translation shows that part of their information must be homologous since the complete set of early light-inducible translation products is obtained with all investigated clones although the proportions of the translated bands vary for individual clones. From hybridization data it is concluded that two ELIP families of high (24-27 kDa) and of low (16-18 kDa) molecular mass exist which are induced in parallel. Induction of ELIPs occurs even at very low light intensities and is saturated at about 1000 lx. Therefore, ELIPs are not considered to represent light stress proteins but to play a regulatory role during development.

The prognostic value of EEG patterns in epilepsies with infantile spasms.

By scoring EEG patterns (hypsarrhythmia = 10, absence of sleeping patterns = 10, focal epileptic discharge = 5, general-treatment or in whom infantile spasms never disappeared even during ACTH. A low voltage EEG did not have any ending ACTH therapy free of seizures showed lower scores compared to those infants relapsing after the end of ACTH treatment or in whom infantile spasms never disappeared even during ACTH. A low voltage EED did not have any prognostic significance. Using EEG scores it might be possible to separate non-responders and responders after 3 weeks of ACTH therapy, thus shortening ACTH treatment in non-responding infants.

[Serum endotoxin level in the course of open peritonitis treatment].

In 27 patients with severe diffuse purulent or fecal peritonitis planned relaparotomies with peritoneal lavage or continuous dorsoventral lavage with open abdomen were performed after surgical treatment of the primary infection. During the course of the lavage treatment serum endotoxin was measured daily. The endotoxin-induced liberation of lysosomal proteases was studied by determining the elastase from polymorphonuclear leucocytes. 16 surviving patients showed decreasing endotoxin levels and decreasing elastase concentrations during the course of abdominal lavages. Planned peritoneal lavage and continuous dorso-ventral lavage seem to have the same potency in eliminating endotoxin from the infected peritoneal cavity. In letal courses endotoxinemia either persisted at high levels or even progressed inspite of lavage treatment.

Optimizing a hydroxyapatite/tricalcium-phosphate ceramic as a bone graft extender for impaction grafting.

The mechanical properties of morsellized bone allografts and synthetic hydroxapatite/tricalcium-phosphate (HA/TCP) ceramic extender materials for the use in impaction grafting revision hip surgery were investigated using two test methods: a basic compression test and an endurance test in an in-vitro model of an impaction grafted femur. Formalin fixed ovine bone graft was identified as mechanically similar to fresh human bone and thus suitable as an experimental material for in-vitro testing. For 1 : 1 volumetric mixes of bone allograft and synthetic extender, the granular ceramic's properties were varied in porosity, chemical composition, sintering temperature and particle size. Initial mechanical stability, a crucial prerequisite for clinical success in impaction grafting, was increased for all bone/extender mixes. A high porosity, tricalcium-phosphate rich ceramic of medium particle size and sintered at high temperatures was recognized as an optimized extender material for impaction grafting balancing the mechanical and biological demands. Using the extender without bone graft as a pure replacement is not recommended.

Regional variation in corneal topography and wound healing following photorefractive keratectomy.

BACKGROUND: After excimer laser photorefractive keratectomy, corneal epithelial and anterior stromal wound healing may produce corneal haze and variability in refractive results among patients and eyes. METHODS: We report a retrospective study of 17 selected eyes that received excimer laser photorefractive keratectomy with one of three brands of laser and that exhibited regional variations in corneal wound healing as observed with slit-lamp microscopy and videokeratography (Tomey TMS) within the ablation zone. RESULTS: All 17 eyes were selected to show that the clear areas in the ablation zone corresponded with flat zones on the videokeratographs and the areas of subepithelial haze corresponded with steeper zones. Spectacle-corrected visual acuity was measured in 12 or the 17 eyes, and was 20/20 or better in 58% and 20/30 or better in 100%, indicating that the regional variation did not severely degrade Snellen visual acuity. Further studies are needed to determine how often regional variability occurs and to better define the relationship of corneal haze and corneal curvature. CONCLUSION: Regional variations in corneal topography and haze can occur within the ablation zone in an individual cornea after photorefractive keratectomy.

[Collection of Arabidopsis thaliana mutants with altered sensitivity to oxidative stress inductors].

Selective systems for screening Arabidopsis thaliana (L.) Heynh. mutants with altered sensitivity to the oxidative stress (OS) inductors norflurazon (NF), acifluorfen (AF), and plumbagin (PB) were developed and a collection of 28 mutants was obtained. Dwarf and necrotic forms predominated among the NF-tolerant mutants, while pigment mutants and those with changed root morphology prevailed among the AF-tolerant and PB-sensitive mutants, respectively. Genetic and biochemical analysis of certain mutants was performed; quantitative and qualitative changes in the content of superoxide dismutase and peroxidase isoforms have been revealed. These data, complemented by the data on the cross-tolerance (sensitivity) of the mutants to paraquat, indicate a correlation between tolerance to the OS inductors and the functions of antioxidant systems.
