Anticipatory contrast effect in rats: a new view with lick response analysis and the effect of dopamine blocking.

A negative anticipatory contrast effect (NACE) is manifested as a suppression of the effect of a reward stimulus to elicit consummatory or instrumental responses when a second more potent reward is presented right afterward. No well-supported theory explains the effect. We analyzed NACE with rats' licking responses to 3 solutions with varying reward value: 8% sucrose mixed with 0.008% quinine (A), 8% sucrose with 0.002% quinine (B), and 32% sucrose with 0.008% quinine (C). 3 groups (n = 4) of rats were given 2 solutions in succession, each 5 min with 15 s in between, for a total of 11 days: Group AA (control), A then A; Group AB, A then B; Group AC, A then C. On days 13 to 16, and 12 and 17, pimozide (0.4 mg/kg, i.p.), and the vehicle solution were injected, respectively. Results show that the licking of A was a function of what solution to follow: In AC, the total number of licks, the short interlick interval (SILI), and the number of licks per s during the initial contact were less than AA. In AB, only SILI was less. Licking of A was initially low but increased over days, except for AC. The failure of AC to increase the licking was attributed to a "retroactive overshadowing" effect of a more potent stimulus C blocking stimulus A to form a reward learning, thus preventing its rewarding property to be fully manifested. It may also be due to stimulus A becoming a "signal" for incoming reward, thus degrading its own rewarding function. The shorter SILI in AB and AC suggested a reward devaluation; however, devaluation alone did not predict the consummatory response. Pimozide reduced licking number overall without altering the relative reward property or SILI suggesting that NACE was not a dopamine-mediated motivated choosing behavior.

Complex response competition and dopamine blocking: choosing of high cost sucrose solution versus low cost water in rats.

The effect of DA blocking with pimozide on complex response competition involving coupling of cost and benefit was studied with a double-bottle test in lickometer: The competition occurred between two responses; licking of 2% sucrose solution requiring a high effort of standing up and licking of water requiring a low effort of crouching posture. Rats normally chose sucrose over water (76% with vehicle); however, pimozide (0, 0.25, 0.375 and 0.5 mg/kg) diminished licking of the sucrose solution dose-dependently (57% at 0.5 mg/kg) without affecting licking of water. Analysis of lick pattern suggested that this shift in choice was attributable not much to a potential effect of pimozide to hamper motor capability or to reduce rewarding impact of sucrose because (a) the difference in licking profile for sucrose and water remained intact, (b) rats remained fully capable of assuming the standing posture for sustained and efficient licking of sucrose, and (c) the integrity of licking pattern largely remained. Pimozide appeared to selectively affect the behavior that cost more but with higher reward; rats settled with behavior that cost less with lower reward. Rats became less motivated to invest effort for a better reward, a behavior pattern described as "indolent". A more complex view of incentive motivation including coupling of behavioral cost and benefit was presented as a theory to explain the DA mediation of reward-maintained behaviors.

Molecular characterization of glucose-6-phosphate dehydrogenase deficiency in Chinese infants with or without severe neonatal hyperbilirubinaemia.

To characterize mutations in the glucose-6-phosphate dehydrogenase (G6PD) gene in Chinese infants, we studied 213 G6PD-deficient infants without blood exchange transfusion (BET) therapy, and 34 patients who required BET therapy for their severe hyperbilirubinaemia after birth. Nine different point mutations were characterized in all infants. Of these mutations, the G to T substitution at cDNA nucleotide (nt) 1376, which accounts for the mutations in 131 (53.0%) neonates, followed by G to A substitution at nt 1388 in 18 (10.5%) infants, A to G substitution at nt 493 in 17 (6.9%) infants, A to G substitution at nt 95 in 10 (4.1%) infants, C to T substitution at nt 1024 in six (2.4%) infants, and G to T substitution at nt 392 in three (1.2%) infants, G to A substitution at nt 487 in two (0.8%) infants, C to T substitution at nt 1360 in two (0.8%) infants and C to T substitution at nt 592 in two (0.8%) infants. Mutations in 48 (19.5%) G6PD-deficient infants were not characterized. Most (64.7%) mutations in the G6PD-deficient infants who required BET therapy after birth result from a G to T substitution at nt 1376. The enzyme activity of G6PD deficient infants who required BET therapy is significantly lower than for those who did not, even in a group with the same variant (as in 1376 mutation). Severe neonatal jaundice requiring BET therapy can take place with the majority of variants encountered in this area.

Syntheses of 2,5- and 2,6-difluoronorepinephrine, 2,5-difluoroepinephrine, and 2,6-difluorophenylephrine: effect of disubstitution with fluorine on adrenergic activity.

Synthetic routes to difluorinated analogs of the adrenergic agonists, norepinephrine (NE), epinephrine (E), and phenylephrine (PE) have been developed. The syntheses were based on elaboration of the ethanolamine side chains from the appropriately polyfunctionalized benzaldehydes. The benzaldehydes were prepared from precursor difluorinated benzenes by sequential regioselective lithiations and reaction with electrophiles to introduce hydroxyl and carboxaldehyde functionalities. Binding and functional assay data demonstrate that the 2,6-difluorinated analogs are relatively inactive at both alpha- and beta-adrenergic receptors. These results are consistent with earlier observations that 2-fluoro substitution of adrenergic agonists decreases alpha-adrenergic activity whereas 6-fluoro substitution decreases beta-adrenergic activity.

[Multiple noncontiguous spinal fracture].

Twenty-seven cases of multiple noncontiguous spinal fracture were reported. The injury located mostly at thoracolumbar levels, and primary injuries occurred in thorax (n = 13, 50%), second injuries occurred in lumbus (n = 14, 53.8%). The cord function of 12 patients were Frankel A, 5 were C and 4 were D; five patients were normal (Frankel E). The incidence of neurologic injury was significantly different between multiple noncontiguous and contiguous spinal fractures (P < 0.01). There were 17 followed-up cases. Eight were recovered with Frankel 1 degree, 2 with 2 degree and 4 without change. The importance of correct diagnosis is emphasized and it is recommended that total spine radiography be performed as soon as possible.

Sensitivity to dopamine blocking in rat licking behavior: function of taste stimuli, response difficulty, and response measures.

The behavioral circumstances determining the efficacy of DA blocking was studied by observing the effects of pimozide, tastes and effort required to ingest liquid on consummatory licking responses. The microstructure of ingestive behavior was obtained to delineate differential behavioral adjustments. Twelve water-deprived rats were trained to lick water, 2.00% sucrose, and 0.01% quinine from a spout located either at a down position of 7 cm above (low effort) or an up position of 24 cm above the floor (high effort). They were injected with pimozide (0.25 mg/kg, ip) or its vehicle and licking responses were observed. Results indicated that the pimozide effect was dependent on tastes and positions as well as the behavioral aspects observed. Pimozide affected ingestion of quinine in many ways, but did not affect that of sucrose solution. The position affected ingestion of quinine in more ways than that of sucrose solution. The taste effect of quinine was more complex than that of sucrose. The complex behavioral adjustment to the reduced incentive support with quinine probably rendered the behavior vulnerable to interference by DA blocking and effort requirement.

A new, general synthetic route to multidentate N,S ligands for use in technetium-99m radiopharmaceuticals. Preparation of diamido disulfur, diamino dithiol, and tripodal N3S3 prototypes. Comparative biodistributions of [99mTcvO-DADS]- analogues which contain 5,5,5- and 5,7,5-membered chelate ring systems.

A new, versatile synthetic route to a variety of tetradentate N2S2 and hexadentate N3S3 ligands for use in technetium-99m radiopharmaceuticals has been developed. The key reaction employs 1-(ethoxycarbonyl)-2-ethoxy-1,2-dihydroquinoline (EEDQ) for coupling an appropriate di- or triamine with S-protected thioglycolic acid. Selected DADS (diamido disulfur) and DADT (diamino dithiol) and analogues derived from ethanediamine-1,2 and butanediamine-1,4, as well as a tripodal N3S3 analogue, were synthesized in high yield. The labeling of DADS analogue ligands derived from butanediamine-1,4 (DADS-bn) with 99mTc results in a single radiochemical product, or the expected number of stereoisomeric products. FAB MS analysis, using 99Tc, indicates that DADS-bn ligands form a tetradentate 5,7,5-membered ring chelate system with the monooxo Tc(V) core: [TcO-DAD-bn]-. In rat biodistribution studies the DADS-en and DADS-bn complexes show very similar biological activity. Phenyl substituents on the 7-membered ring give rise to a 99mTc complex of increased lipophilicity, increased liver uptake, and minimal hepatobillary clearance. Thus, new classes of DADS ligand analogues which contain a 5,7,5-membered chelate ring system are readily synthesized and labeled with 99mTc to form stable complexes. The presence of the 7-membered chelate ring allows for facile introduction of pendant groups into the ligand system, and thus for a ready route to control the biodistribution of the resulting 99mTc radiopharmaceutical. The 99mTc labeling of the DADT analogues derived from butanediamine-1,4 provided to be erratic, despite the use of a variety of labeling techniques; the larger ring system of DADT-bn apparently does not favor the monooxo Tc(V) core and appears to generate a mixture of mono- and dioxo Tc(V) cores.

Dystrophin immunostaining of muscle from Chinese patients with various neuromuscular diseases.

The localization of dystrophin was studied using the immunohistochemical method of diagnostic muscle specimens from 68 patients, aged 9 days to 65 years, with various neuromuscular disorders. Additionally muscle specimens from 2 normal humans and 2 normal mice were used as positive controls, and those from 2 mice with x-linked muscular dystrophy as negative controls. The specimens from all 14 Duchenne muscular dystrophy (DMD) patients, including one with preclinical DMD, showed negative dystrophin staining except for two which had 0.2% to 0.8% positive fibers. The mdx mice also showed negative dystrophin staining. In Becker muscular dystrophy (BMD), muscle fibers stained in a patchy or discontinuous fashion. Two symptomatic DMD carriers exhibited a distinct mosaic pattern of dystrophin positive and negative fibers. In contrast, dystrophin was present in all 7 biopsies from patients with 4 other types of muscular dystrophy (limb-girdle, congenital, myotonic and facioscapulohumeral). Other specimens, those from normal humans and control mice, revealed homogeneous immunostaining along the surface membranes of all muscle fibers. We thus conclude that immunohistochemical dystrophin staining can aid in differentiating DMD from preclinical DMD or BMD, as well as in the detection of DMD carriers.

Localization of functional projections from corpus callosum to cerebral cortex.

In 30 patients with intractable generalized epilepsy treated with cerebral commissurotomy, the corpus callosum was stimulated intraoperatively at a 1 cm interval with electric current, and evoked potentials (EPs) were recorded from different areas of the brain for determining the distribution patterns of functional projections from the corpus callosum to the cerebral cortex. The surface of the corpus callosum in man was 12 cm long and it was divided into 12 segments (1 cm each). Stimulation of the first segment resulted in EPs only in the frontal lobe. Stimulation of segments 2-4 produced EPs mainly in the anterior, middle and posterior frontal lobe, anterior and central temporal lobe, rarely in parietal and occipital lobes. Stimulation of segments 5-8 induced EPs mainly in the frontal and temporal lobes, but rarely in the parietal and occipital regions; on stimulation of segments 9-11, EPs occurred only in the parietal and occipital regions. These results were confirmed electrophysiologically in 5 cats and anatomically in 8 cats. These data provide an anatomical basis for selective cerebral commissurotomy in the treatment of intractable epilepsy. Hence in patients with a concentration of epileptic discharges in the frontal lobe, the 4 anterior segments of the corpus callosum should be incised. Epileptic discharges in the frontotemporal region indicates that the middle segments should be incised and epileptic activities originating from the parieto-occipital regions can be treated effectively by selective section of the posterior corpus callosum (segments 9-12).

[The prevalence of amphetamine use in adolescent students: self-reported and urine analysis].

The prevalence of amphetamine use was studied using an anonymous questionnaire given to 3548 adolescent students aged 12--15, with a response rate of 98.6%. This questionnaire was developed and evaluated by the authors. A complete set of data was collected on 3200 subjects, making up 90.2% of all students surveyed. Eighty-five (2.7%) students admitted that they had used amphetamines; that is, 64 of the 1,584 male (4.0%), and 21 of the 1,616 female (1.3%) respondents. The life-time prevalence odds ratio estimation was elevated in male (POR = 3.2, 95% CI 1.9--5.3), as compared to female students. The life-time prevalence rates increased with age only among male students (P trend = 0.01). Seven hundred and twenty four male students were randomly tested for the presence of amphetamines in their urine using thin layer chromatography and fluorescence polarization immunoassay. Of six students who tested positive, five asserted that they had not used amphetamines. The point prevalence rate of amphetamine use based on the urine tests was 0.9% among the 652 students who had completed questionnaires. This rate was higher than on self-reported use of amphetamines (rate difference 0.9%, 95% CI 0.2--1.6%). In this group, 27 (4.1%) admitted that they had used amphetamines. The estimated overall life-time prevalence rate was at least 4.9% (32/652), with 95% CI between 3.2% and 6.6%.

Serum immunosuppressive acidic protein levels in blackfoot disease patients and cancer patients.

Immunosuppressive acidic protein (IAP) was first found in the ascitic fluids of cancer patients. Its biochemical properties are significantly different from those of acidic protein in the serum of normal persons. Previous studies have indicated that the serum IAP concentration increases in most cancer patients and decreases to a normal level as such patients are cured. Therefore, it has been suggested as a useful marker for follow-up in operated cancer patients. In this study, analyses and comparisons of serum IAP concentrations have been made among 53 normal persons in Blackfoot disease endemic areas, 25 patients with diabetes, cataracts, hypertension and cardiovascular disease in Blackfoot disease endemic areas, 50 breast cancer patients, 13 colorectal cancer patients, and 18 Blackfoot disease patients. Serum IAP concentrations were found as follows: 454 +/- 138 micrograms/ml for normal subjects and 499 +/- 132 micrograms/ml for disease patients in Blackfoot disease endemic areas; 520 +/- 149 micrograms/ml for breast cancer patients; 864 +/- 341 micrograms/ml for colorectal cancer patients and 950 +/- 368 micrograms/ml for Blackfoot disease patients. Serum IAP concentrations were much higher in Blackfoot disease patients, than in normal persons in Blackfoot disease endemic areas (p less than 0.001), and as high as in colorectal cancer patients. In Blackfoot disease patients, the mean serum IAP concentration of 6 patients coming from the Blackfoot disease endemic areas was as high as 1,238 +/- 404 micrograms/ml, showing a positive rate of 100% to IAP (i.e. IAP concentration exceeds 500 micrograms/ml). We conclude that serum IAP assay of Blackfoot disease patients may be useful for prognosis and therapeutic monitoring.

[Effect of penicillamine and tetramethyl pyrazine on the hypoxic pulmonary hypertension in rats].

Penicillamine (P) and tetramethyl-pyrazine (T) were administered to treat experimentally hypoxic pulmonary hypertension in rats exposed to hypoxia of 380 torr for up to 21-days. The results of this study showed: Hypoxia increased RVP, the RV/LV + S ratio, the MT% and the MA%. P and T all prevented the hypoxia-induced increases in RVP, the RV/LV + S ratio, but the effect of T on the pulmonary artery remodeling induced by hypoxia was weaker than that of P. The MT% and the MA% were significantly lower in H/P group than in H group (P less than 0.01, P less than 0.05; respectively). Although the MT% was lower in H/T group than in hypoxia group (P less than 0.05), there was no statistical difference in the MA% between H/T group and H group (P greater than 0.05). The MT% and the MA% were significantly lower in H/PT group than in H group (P less than 0.01, P less than 0.01, respectively), and there was no statistical difference in MA% between H/PT group and N group (P greater than 0.05). It was concluded that P prevented primarily pulmonary artery remodeling induced by hypoxia and T inhibited mainly hypoxic pulmonary vasoconstriction response, so prevented partially hypoxic pulmonary hypertension and right ventricular hypertrophy.

Indoor air pollution in developing countries.

Of the four principal categories of indoor pollution (combustion products, chemicals, radon and biologicals), research in developing countries has focused on combustion-generated pollutants, and principally those from solid-fuel-fired cooking and heating stoves. Such stoves are used in more than half the world's households and have been shown in many locations to produce high indoor concentrations of particulates, carbon monoxide and other combustion-related pollutants. Although the proportion of all such household stoves that are used in poorly ventilated situations is uncertain, the total population exposed to excessive concentrations is potentially high, probably several hundred million. A number of studies were carried out in the 1980s to discover the health effects of such stove exposures. The majority of such studies were done in South Asia in homes burning biomass fuels or in China with coal-burning homes, although a sprinkling of studies examining biomass-burning have been done in Oceania, Latin America and Africa. Of the health effects that might be expected from such exposures, little, if any, work seems to have been done on low birthweight and eye problems, although there are anecdotal accounts making the connection. Decreased lung function has been noted in Nepali women reporting more time spent near the stove as it has for Chinese women using coal stoves as compared to those using gas stoves. Respiratory distress symptoms have been associated with use of smoky fuels in West India, Ladakh and in several Chinese studies among different age groups, some with large population samples. Acute respiratory infection in children, one of the chief causes of infant and childhood mortality, has been associated with Nepali household-smoke exposures. Studies of chronic disease endpoints are difficult because of the need to construct exposure histories over long periods. Nevertheless, chronic obstructive lung disease has been associated with the daily time spent near the stove for Nepali women and found to be elevated among coal-stove users compared to gas-stove users in Shanghai. In contrast to early reports, there seems to be little or no risk of nasopharyngeal cancer from cookstove smoke. Several studies in China, however, have found smoke to be a strong risk factor for lung cancer among non-smoking women. In addition, severe fluorosis has been observed in several parts of China where coal fluoride levels are high.(ABSTRACT TRUNCATED AT 250 WORDS)

[Experimental study of removal of sublaminar wires after spinal fixation].

The result of experimental study of removal of sublaminar wires was reported. The experimental shows that sublaminar wires when with drawn at 45-degrees is safer than that at 90-degrees. Because of bony fusion, clinically could be drawn out only vertically either by rolling it up ward or down ward confirmed that there is no difference between this two methods. The cut end of wire should be short and straight. All the fixing wires became useless because of their loosening. Impressions were found on the inner face of lamina, fibrous sheaths. There is no different reactions between twisted and parallel double wires, but we recommend double parallel wires.

[Synthesis of indolinyl-N,N-dimethylcarbamates as reversible anticholinesterase agents].

A series of new indolinyl derivatives (I1-5, II1-3 and III1-8) with different substituents at 1-, 3- or 5-position were synthesized in order to study the relationship of structure and anticholinesterase activities of CUI XING NING and its derivatives. The method of phase transfer catalysis was successfully applied in the C3-alkylation of intermediate A as well as in the N-alkylation and the carbamylation. Three by-products were obtained during the C3-alkylation and their structures and all of the structures of the 24 new compounds (including the intermediate) were identified by spectral and elementary analysis. Preliminary pharmacologic tests showed that most of the compounds have potent anticholinesterase activities. All of the substituents of 1-, 3- or 5-position of the compounds may affect their activities (see Table 4).

Comparison of disposition values obtained by two assay methods for quinidine gluconate in patients with ventricular tachycardia.

Eight patients with previously untreated ventricular tachycardia, age 48.54 +/- 28.02 years (mean +/- SD), were enrolled in a protocol evaluating the disposition of quinidine gluconate as determined by two assay methods. Patients received two infusions of 5 mg/kg over 30 minutes separated by 20-30 (24.9 +/- 4.0) minutes of electrophysiologic testing. Blood samples were obtained at 0.17 hours and just prior to the second infusion, and then at 0.17, 0.25, 0.33, 1.0, 6.0, 12.0, and 24.0 hours after the second infusion. Paired serum samples were assayed for quinidine concentrations by fluorescence polarization immunoassay and high-performance liquid chromatography. The two assays compared well, with a linear regression equation of Y = 0.927X + 0.247 with a correlation coefficient of 0.985. With the exception of the beta elimination rate constant and beta distribution volume, t test comparison of disposition values demonstrated no significant difference. Differences in the estimates of the beta elimination rate constant reflected differences in the two methods and indicated that even though both assays were comparable, subtle differences in specificity could be reflected in significant differences in this variable.

Disposition of 3-hydroxyquinidine in patients receiving initial intravenous quinidine gluconate for electrophysiology testing of ventricular tachycardia.

The formation rate constant and elimination rate constant for 3-hydroxyquinidine were determined in eight patients with ventricular tachycardia. These two parameters (mean +/- SD) were found to be 0.784 +/- 0.202 and 0.042 +/- 0.058 h-1, respectively. Coefficients of determination for the computer-generated line of best fit for serum concentration-time data were 0.986 +/- 0.008. Patients received two infusions of quinidine gluconate 5 mg/kg over 30 minutes separated by a 20-30 minute electrophysiologic testing period. Unbound and total 3-hydroxyquinidine concentrations were also determined. Among the eight patients, 3-hydroxyquinidine was 61.9 percent bound. Studies in healthy volunteers had shown 50 percent binding. Linear regression of unbound and total 3-hydroxyquinidine was described by the equation Y = 0.3814X-1.448, r = 0.813. Although half-lives of 3.5-12.4 hours had been reported in healthy volunteers, prolonged half-lives were observed in all but two of our arrhythmia patients.

[The immunotoxicological basis for the revision of suspended particulates health standard].

The paper presented the effects of suspended particulates and coal combustion on body's immune function, including the effects of suspended particulates (SP) injected intratracheally, and of inhalation of coal combustion emissions on animal immune function, as well as the effects of indoor coal combustion products and atmospheric pollution on the immune function of children. On the basis of these studies, the maximum daily average allowable concentration of 0.15 mg/m3 of atmospheric SP is justifiable from the immunotoxicological point of view.
