Fifty patients underwent myelography with the newly developed CSF-isotonic, dimeric, non-ionic contrast medium Iotrolan. Repeat spinal and cranial computerised tomographic studies with measurement of the attenuation values were conducted to demonstrate the administered contrast medium in the spinal canal and intracranial subarachnoid space. The patients were examined neurologically, observed clinically and asked about concomitant symptoms. The period of retention and the spread of the contrast medium in the subarachnoid space was not found to have any influence on the side effects rate. In particular, no association was confirmed between intracranially demonstrable contrast medium and the occurrence of side effects.

FAS1, the structural gene of the pentafunctional fatty acid synthetase subunit beta in Saccharomyces cerevisiae has been sequenced. Its reading frame represents an intron-free nucleotide sequence of 5,535 base pairs, corresponding to a protein of 1,845 amino acids with a molecular weight of 205,130 daltons. In addition to the coding sequence, 1,468 base pairs of its 5'-flanking region were determined. S1 nuclease mapping revealed two transcriptional initiation sites; 5 and 36 base pairs upstream of the translational start codon. Within the flanking sequences two TATATAAA boxes, several A-rich and T-rich blocks and a TAG...TATGTT...TATGTT...TTT sequence were found and are discussed as transcriptional initiation and termination signals, respectively. The order of catalytic domains in the cluster gene was established by complementation of defined fas1 mutants with overlapping FAS1 subclones. Acetyl transferase (amino acids 1-468) is located proximal to the N-terminus of subunit beta, followed by the enoyl reductase (amino acids 480-858), the dehydratase (amino acids 1,134-1,615) and the malonyl/palmityl transferase (amino acids 1,616-1,845) domains. One major inter-domain region of about 276 amino acids with so far unknown function was found between the enoyl reductase and dehydratase domains. The substrate-binding serine residues of acetyl, malonyl and palmityl transferases were identified within the corresponding domains. Significant sequence homologies exist between the acyl transferase active sites of yeast and animal fatty acid synthetases. Similarly, a putative sequence of the enoyl reductase active site was identified.
Four groups of subjects of different ages and sex (group I: 20-29 years, women; group II: 20-29 years, men; group III: 30-39 years, women; group IV: 30-39 years, men) undertook dynamic one-hand work (load range 40%-80% of maximum voluntary contraction, at 60 working cycles/min) to allow a study of cardiovascular responses as shown by the resultant changes in blood pressure and heart rate. During fatiguing dynamic one-hand work, there was a large increase in systolic and diastolic blood pressures in both sexes after a few minutes. For all load levels, the systolic blood pressure was found to be higher by about 4 kPa in men (groups II and IV) than in women (groups I and III). Other age-related differences became evident in the diastolic blood pressure changes. The values obtained for the older groups were higher than those in the two younger groups. These differences in blood pressure response are possibly due to sex-related differences in the release of catecholamines, or to age-related organic changes in the vessels.
The tonoplast of isolated vacuoles from photoautotrophic suspension cells of Chenopodium rubrum L. was studied by means of the patch-clamp technique. In a symmetrical K(+) concentration of 46 mM, similar to in vivo conditions, the tonoplast displayed a membrane potential near zero and a linear current-voltage relationship with a mean slope of 1.0 S/m(2). ATP at 2 mM hyperpolarized the tonoplast (vacuole positive) by 15-20 mV and, in a parallel experiment, acidified the vacuole (outside pH 7.0) to pH 5.0, as monitored by accumulation of acridine orange. Analysis of the voltage-clamp current indicates a 2-fold, ATP-dependent increase of the membrane capacitance, from 4 to 8 mF/m(2), and an ATP-independent, unidentified ion channel having a mean opening time of about 5 msec and a conductivity of 0.5-1.0 pS.

Latero-basal fractures of the skull base due to a blunt head injury may cause vascular and nervous lesions. The most important lesion is rupture of the middle meningeal artery in the foramen spinosum leading to an acute epidural haematoma. Its early diagnosis is of vital importance. Lesions of the cranial nerves are rarely surgically remediable. Diagnostic steps in the management of these lesions are outlined. Close cooperation between ENT specialists and neurosurgeons is of the highest importance, particularly when dealing with petrous bone lesions producing C.S.F. otorrhea or the prolapse appearance of contused brain into the auditory canal.

There are load-dependent increases in the bioelectrical activity (iEMG) of muscles of the shoulder and the right upper extremity and selected parameters of the cardiopulmonary system. At comparable loads the amplitudes of the EMG were higher in the group of men than in the group of women. The iEMG values were found to be higher in the younger group of women than in the older one. The higher values of iEMG of the younger women corresponded to a higher oxygen uptake and higher heart rate as well as to a lower physical endurance. The physical endurance limit (derived from behaviour of the EMG) was estimated at 5 watts (women) and 10 watts (men).
