[Gonococcal infections in Gabon (Haut-Ogooué)].

The isolation of N. gonorrhoeae in the region of Franceville was attempted on the one hand with 261 men and 261 women presenting urethral or cervical discharge, and on the other hand with 530 women attending a prenatal clinic of whom 42 had cervical discharge while 488 did not. Amongst the men, 69.7% of the urethritis cases were due to N. gonorrhoeae while this bacteria could be isolated from 15.3% of the women with cervical exudates. N. gonorrhoeae could be also identified in 5.5% of the pregnant women of whom 79% were without exudate. The frequency of N. gonorrhoeae isolates from STD consultations was 440 cases per year per 100,000 while the male: female ratio was 4.6. The isolation rate from pregnant women would indicate however a frequency in women in excess of 1 500 cases per 100,000. The isolation of N. gonorrhoeae was tabulated according to the age of the patients. The 15-20 year group was clearly the most frequently infected. Trichomonas was observed in 2% of the men and 23% of the women with discharges. Yeasts (chiefly Candida albicans) were found in 23.5% of the women but in none of the men.

[The placenta of premature and hypotrophic Gabonese infants].

494 placentae coming from two different geographical parts of Gabon were studied. They were grouped into control, premature, small-for-dates and stillborn. The urban population gives rise to most mothers of about 26 years of age, who deliver normally at 38 weeks of babies weighing 3,300 g and 50 cm long, with placentae weighing 550 g, as well as premature babies and those in which there is white cell infiltration of the membranes, signs of toxaemia and normal placentae. On the eastern plateaux of Gabon there are more mothers around the age of 23, who deliver normally at 37 weeks of pregnancy of babies weighing 3,100 g and 48 cm long, with placentae weighing 450 g, with small-for-dates babies and inflammation of the placenta. Malaria is the cause of many premature deliveries and of small-for-dates and stillborn, especially in the plateaux.

Antiinflammatory and immunostimulant activities of six sulphur compounds--four benzenesulphonates, levamisole, and pyritinol hydrochloride--assayed in mouse cell activation studies.

The effect of a phlogogenic agent and of six sulphur compounds on peritoneal macrophages, spleen B lymphocytes, and von Kupffer cells of the liver has been observed in BALB/C mice. The phlogogenic agent (0.5% carrageenin + 10% peptone) was a potent stimulant of the macrophages (increasing their number, protein content, and acid phosphatase activity), the lymphocytes (increase of haemolytic plaque counts), and the von Kupffer cells (increase of the colloidal carbon clearance rate which measures phagocytic capacity). Of four acid sulphur compounds (benzenesulphonates)-ethamsylate, diethylammonium persilate, calcium dobesilate, and piperazine sultosilate--not previously screened for antiinflammatory activity, all except piperazine sultosilate inhibited the macrophages, all except calcium dobesilate stimulated the lymphocytes, and only calcium dobesilate stimulated the von Kupffer cells. Two non-acid sulphur compounds--levamisole and pyritinol hydrochloride--had no effect on the macrophages but were potent stimulants of the lymphocytes and the von Kupffer cells. Because of these immunostimulant properties, levamisole and pyritinol hydrochloride may prove useful in the treatment of diseases such as rheumatoid arthritis in which the patient's immune potential has to be restored.

The placental lesions in congenital syphilis. A study of six cases.

Placentas from six mothers with serological tests suggestive of recent syphilitic infection and whose babies were suspected of being or proven to be infected by Treponema pallidum are described. One placenta from this series was large, bulky and pale, while the other 5 were without remarkable gross features. In all cases, the associated histological lesions were 1) hypercellular areas in the terminal and stem villi and 2) a focal peri- and/or intravillous polymorphonuclear concentration with or without necrosis. The former change which was the most frequent was characterized by an apparent increase of villous stromal cells, ultrastructurally identified as mesenchymal cells and Hofbauer cells. In addition, numerous fetal monocytes were found in the villous vascular lumina. The findings described here and in the literature suggest that congenital syphilis is associated with a spectrum of placental changes. We believe that these changes depend on the immunological reaction of the fetus. According to the sequence of events described in untreated patients, we distinguish two morphological phases: 1) an inductive phase without placental changes and 2) a reactive phase characterized by a predominantly lymphocytic inflammatory infiltration of the villi, followed by a reaction of mononuclear phagocytes.

Haemorrhagic fever in Gabon. I. Incidence of Lassa, Ebola and Marburg viruses in Haut-Ogooué.

A serological enquiry aimed at determining the incidence of infection with Lassa, Ebola and Marburg viruses was conducted on the human population of the region of Haut-Ogooué (Gabon) and on primates. The results, obtained by the indirect immunofluorescence technique, showed that more than 6% of the human population had had contact with Ebola virus but no antibodies against Marburg or Lassa viruses were found. Most sera reacted to an Ebola antigen from a Zairian strain, but showed little or no reaction to an antigen from a Sudanese strain.

Study of the mode of action of ribosomal vaccines from Klebsiella and Streptococcus pneumoniae and their ribonucleic and protein fractions using passive immunization.

The vaccinating potency of ribosomal fractions of Klebsiella pneumoniae and Streptococcus pneumoniae as well as their ribosomal RNA and protein fractions has been studied with respect to their ability to induce cellular or humoral immunity. Experiments with transfer of serum or spleen cells from vaccinated animals have shown that anti-Klebsiella immunity is essentially cellular, while streptococcal immunity is exclusively humoral. Results have been discussed as a function of differential results for the various fractions under study.

On the immunogenicity of ribosomes and ribosomal proteins isolated from Klebsiella pneumoniae and Streptococcus pneumoniae.

The two pathogenic species Streptococcus pneumoniae and Klebsiella pneumoniae were used to analyze the immunogenic role of proteins in ribosomal preparations. The protective activity of ribosomes prepared from either strain and further purified by washing with high-salt concentrations, followed or not by sucrose gradient separation of the particles, was identical to that of crude unwashed ribosomes. Similarly, no substantial alteration of the level of protection was observed after treatment with the antibiotic puromycin. Therefore, the immunizing efficacy of ribosomes does not appear to be due either to the nonribosomal proteins adsorbed at the surface of organelles or to the growing polypeptide chain. It seems rather to be attributable to the structural ribosomal proteins themselves, which were indeed shown to induce alone a significant level of protection.

[Importance of respiratory route in experimental salmonellosis in Balb/c mice].

Infection of Mice with S. typhimurium, was conducted by respiratory, oral and peritoneal routes. The results obtained showed that infection occurred more quickly by i. p. than by other routes. However i. n. inoculation is the more efficient concerning Bacteria concentration per Mousse to obtain the DL 100. A study of Salmonella growth in vivo was conducted in lungs, spleen, liver and blood. The results are discussed in relation to the calf respiratory Salmonellosis actually observed.

[Enzymatic activity of peritoneal macrophages of Balb C mice after enteral or parenteral stimulation with bacterial extracts].

A study of peritoneal macrophage activation has been conducted by simultaneous investigation in eleven enzymes in Balb/c Mice stimulated with bacterial extracts, by oral and parenteral route. The results showed important changes in Macrophage number and activity. On the other hand this activation depended upon the route of administration, antigen nature, and time interval between stimulation and peritoneal harvesting.

Role of interferon in mice in protection against influenza A virus by bacterial ribosomes together with membranal glycoproteins of Klebsiella pneumoniae as adjuvant.

Nonspecific protection against infectious aerosols of influenza A virus was obtained in Swiss mice after vaccination by aerosols of bacterial ribosomes together with membranal glycoproteins extracted from Klebsiella pneumoniae as the adjuvant. It was shown that repeated stimulant aerosols were necessary to obtain this protection. Routine estimation of serum interferon levels after administration of the association of ribosomes plus membranal glycoproteins to the animals by aerosol or intravenous route showed that there was no correlation between protection and the presence of serum interferon. It was shown that the serum interferon-inducer activity was due to ribosomes. No induction of serum interferon was obtained with membranal glycoproteins used separately. Local liberation of interferon in the mucous membrane of the upper respiratory tract was not investigated.

[Effect of anti-inflammatory agent administration on Balb/c mouse peritoneal macrophage activity].

A study of the mouse Balb/c peritoneal macrophages has been made after oral antiinflammatory drugs administration on animals which previously had an inflammation with carrageenan. The results show an enhancement of the number of the macrophages, of their enzymatic activities and of their proteins' content. These three parameters fall down more or less rapidly according to the considered antiinflammatory drug.

[Studies in the BALB/c mouse of the efficiency of spraying as compared to other inoculation ways for immunostimulant (author's transl)].

The of efficiency of spraying as compared to intranasal and digestive ways was tested on Balb/c mouse by using on the one hand an immunostimulant product and on the other various types of test : Myxovirus influenzae by aerosol, Streptococcus pyogenes intranasally, Klebsiella pneumoniae intraperitoneally. Results showed that in the case of influenza virus the immunostimulant inhaled as liquid aerosol or powder aerosol, or deposited on the nasal mucosa was very active. When the test germ was Streptococcus and the contamination intra-nasal, only the homologous way was efficient. When the test germ was Klebsiella pneumonia all inoculation forms except powder aerosol were effective. The comparative study of aerosols granulometry helps grasping this phenomenon.

[Experimental cholera in the monkey].

Experimental studies on cholera in Baboons (Papio, P. anubis) and Rhesus Monkeys, have led to unfavourable results. However we have obtained a typical cholera disease in cotton-top Marmoset Saguines oedipeus.
