Brain-derived neurotrophic factor induces long-lasting potentiation of synaptic transmission in visual cortex in vivo in young rats, but not in the adult.

Brain-derived neurotrophic factor (BDNF) rapidly enhances excitatory synaptic transmission in cortical slices. To date, however, a question of how long such an action persists remains unanswered as it is hard to record synaptic responses longer than several hours in slice preparations. To address this question and to investigate possible age-dependency of the action, we analysed effects of a brief application of BDNF and nerve growth factor (NGF) on field potentials of visual cortex in rats of postnatal days 13-17 and 19-24 and in the adulthood for 10-24 h. Evoked potentials to stimulation of the lateral geniculate nucleus were recorded simultaneously from two cortical sites into which the neurotrophin and control solution were injected. An application of BDNF induced a slowly developing increase in the field potential amplitude in young rats. The amplitude attained a plateau level 3-4 h after the infusion; 139 +/- 26% (mean +/- SD) and 132 +/- 21% of the baseline in the rats at P13-17 and P19-24, respectively. This potentiation remained stable from 4 to 8 h, then gradually decreased to the baseline 15-16 h after the infusion. NGF applied in the same way did not induce potentiation. An inhibitor of BDNF receptors blocked the potentiation when it was applied immediately after the BDNF application, but was not effective about 2 h later. In the adults, BDNF did not potentiate field potentials. These results indicate that BDNF induces synaptic potentiation lasting for several hours only in the developing cortex through processes downstream of receptor activation.

Detection of group C rotavirus in children with acute gastroenteritis in Blantyre, Malawi.

Among 606 children who were treated for acute gastroenteritis at the Queen Elizabeth Central Hospital in Blantyre, Malawi, Group C rotavirus (Gp C RV) was detected by enzyme-linked immunosorbent assay in fecal specimens from 16 (3.9%) of 408 inpatients and in 4 (2.0%) of 198 outpatients. Thirteen (65%) children excreting Gp C RV were coinfected with Group A rotavirus.

Level of AIDS and HIV knowledge and sexual practices among sexually transmitted disease patients in China.

BACKGROUND: Little is known about sex practices that increase the risk of contracting HIV infection or the level of HIV and AIDS knowledge among sexually transmitted disease (STD) patients in China. OBJECTIVE: To describe AIDS and HIV knowledge, sexual practices, and factors associated with never using condoms among patients at an STD clinic in Jinan, China. STUDY DESIGN: Clinic patients (n = 498) were randomly sampled to answer AIDS and HIV knowledge questions and to report sexual practices, including condom use. RESULTS: Patients had low levels of AIDS and HIV knowledge and engaged in high-risk sex behaviors. The majority of patients reported having had multiple sex partners. When having sex, few men and no women reported always using condoms. Gender, age, residence, AIDS and HIV knowledge, and having multiple sex partners were significantly associated with never using condoms. CONCLUSION: STD clinic patients report having engaged in high-risk sex behaviors. More research is needed to better understand the factors relevant to developing risk-reduction interventions for these patients in China.

Two-phase unfolding pathway of ribonuclease A during denaturation induced by dithiothreitol.

The dynamics of the unfolding process of bovine pancreatic ribonuclease A (RNase A) unfolded by dithiothreitol (DTT) at a low concentration of 1:30 were investigated in alkaline phosphate-buffered saline solutions at 303K and 313K by using proton nuclear magnetic resonance ((1)H NMR) spectra. Three NMR spectral parameters including Shannon entropy, mutual information, and correlation coefficient were introduced into the analysis. The results show that the unfolding process of RNase A was slowed to the order of many hours, and the kinetics of the unfolding pathway described by the three parameters is best fit by a model of two consecutive first-order reactions. Temperature greatly influences the rate constants of the unfolding kinetics with different temperature effects observed for the fast and the slow processes. The consistencies and the differences between the three sets of parameters show that they reflect the same relative denaturation pathway but different spectra windows of the unfolding process of RNase A. The results suggest that the unfolding process of RNase A induced by low concentrations of DTT is a two-phase pathway containing fast and slow first-order reactions.

Persistent activation of nuclear factor-kappaB by interleukin-1beta and subsequent inducible NO synthase expression requires extracellular signal-regulated kinase.

The role of extracellular signal-regulated kinase (ERK) was studied in the signaling pathway by which interleukin-1beta (IL-1beta) increases the expression of inducible NO synthase (iNOS) in rat vascular smooth muscle cells. IL-1beta induced a rapid and transient activation of nuclear factor-kappaB (NF-kappaB), followed by a prolonged activation of NF-kappaB that was required to induce iNOS expression. Either PD98059 or U0126, selective inhibitors of ERK activation, did not influence IL-1beta-induced early activation but effectively reduced the prolonged activation of NF-kappaB and significantly reduced IL-1beta induction of iNOS. Transfection with antisense, but not sense, phosphorothioate-modified oligodeoxynucleotides directed toward ERK also reduced IL-1beta-induced prolonged NF-kappaB activation and iNOS expression. IkappaBbeta, but not IkappaBalpha degradation, induced by IL-1beta was markedly attenuated when ERK activation was inhibited and could be partially responsible for the persistent NF-kappaB activation. These data suggest that ERK activity is required for persistent NF-kappaB activation by IL-1beta that is necessary for iNOS gene expression.

Phosphopeptides derived from hen egg yolk phosvitin: effect of molecular size on the calcium-binding properties.

Two different phosphopeptide (PPP) fragments derived from partially dephosphorylated hen egg yolk phosvitin were prepared by tryptic digestion, and their Ca2+ binding property compared with that of commercial casein phosphopeptides (CPP). The smaller fragment of less than 1 kDa and O-phospho-1-serine did not bind Ca2+ to any significant extend, while PPP of 1-3 kDa showed a higher ability than CPP to render soluble calcium. The results show that not only the phosphoserine residues are critical for Ca2+ binding, but also the molecular size of the phosphopeptides.

Thermophilic anaerobic digestion of methanol in UASB reactor.

A 5.1 L laboratory scale upflow anaerobic sludge bed (UASB) reactor was operated at 55 degrees C over 130 days in order to investigate the feasibility of treating methanol-containing wastewater under thermophilic conditions, focussing on start-up and process stability. Batch assays were conducted to elucidate the most probable pathway for methanol conversion. The results demonstrated a good performance, with a chemical oxygen demand (COD) removal averaging 82% throughout the experiment. No significant VFA accumulation was detected in the effluent, even with bicarbonate concentration exceeding 20 mM. Acetate was the main component of the VFA at relatively low organic loading rates (OLR). At high OLR, the main components were propionate and butyrate. Reactor performance was hardly affected when the system was exposed to non-optimal conditions, i.e., temperature drop, overloading and no feeding. Good thermophilic granular sludge was retained in the reactor. Washout of biomass was not severe during the experiment. From the pathway analysis it could be concluded that indirect pathways play an important role in the methanol degradation by the cultivated consortia.

Naloxone increases vascular responsiveness in chronic morphine treated rats and facilitates intracellular signaling in cultured vascular cells.

AIM: To probe the changes of vascular responsiveness and intracellular signaling during opiate withdrawal syndrome. METHODS: Morphine withdrawal syndrome in rats was precipitated by iv naloxone following daily injection of increasing dose of morphine for 2 weeks, the changes in mean arterial blood pressure (MBP) caused by acetylcholine (ACh) were recorded. Isolated mesenteric vascular beds were perfused with Kreb's solution containing different concentration of drugs. The cytoplasmic Ca2+ ([Ca2+]i) in Fura 2-loaded cultured bovine aortic endothelial cells (aec) and smooth muscle cells (smc) were assayed. The positive immunoreaction to the phosphorylated cyclic AMP responsive element-binding protein (phospho-CREB) in cultured smc was calculated. RESULTS: Naloxone 2 mg/kg iv following chronic morphine treatment precipitated severe opiate withdrawal signs in company with blunted hypotensive effect of Ach that was the same as chronic morphine treated rats before withdrawal. In the mesenteric vascular beds from chronic morphine treated rats, the EC50 of pressor effect of norepinephrine (NE) was decreased from (2.06 +/- 0.38) to (1.14 +/- 0.21) micromol/L (n = 8, P < 0.01) after the perfusion solution containing morphine 20 micromol/L was replaced by Kreb's containing naloxone 25 micromol/L. Furthermore, NE-induced perfusion pressure increases were completely prevented by Kreb's contained morphine 40 micromol/L. Morphine acutely applied to control smc produced some variable and naloxone-reversible [Ca2+]i changes, but naloxone did not. However, naloxone increased [Ca2+]i in two thirds of smc preincubated with morphine 0.1 or 0.5 mmol/L for 48 h from (97 +/- 20) to (167 +/- 29) nmol/L (n = 9, P < 0.01) and from (106 +/- 19) to (225 +/- 48) nmol/L (n = 10, P < 0.01), respectively, and it also increased the ratio of positive immunoreaction to phospho-CREB from (7.7 +/- 3.2) % to (19.6 +/- 4.7) % (n = 6, P < 0.01) in smc preincubated with morphine 0.5 mmol/L. In addition, naloxone decreased [Ca2+]i from (146 +/- 34) to (78 +/- 24) nmol/L in one third morphine-preincubated aec (n = 10, P < 0.01). CONCLUSION: That naloxone enhances vascular responsiveness to NE in chronic morphine treatment rats may be relevant to [Ca2+]i transient facilitation in company with cAMP-dependent phosphorylation enhancement.

Effects of methylprednisolone and aprotinin on phospholipase D activity of leukocytes in systemic inflammatory response induced by cardiopulmonary bypass.

AIM: To investigate the role of leukocyte phospholipase D (PLD) in systemic inflammatory response induced by cardiopulmonary bypass (CPB) and the effects of methylprednisolone and aprotinin on leukocyte PLD activity. METHODS: Forty-two patients who received CPB open heart surgery were divided into 3 groups: methylprednisolone group, aprotinin group, and control group. Arterial blood (10 mL) was collected for assay of leukocyte PLD activity, myeloperoxidase (MPO) activity, and CD11b expression at 8 different time points in perioperative period. Plasma IL-6, IL-8, and C-reactive protein levels were also determined. RESULTS: At the time point of ascending aorta declamped, leukocyte PLD activity for control group was (18 +/- 8) nmol choline . h-1 . mg-1, which was higher than that of pre-CPB (P < 0.01); the PLD activity for methylprednisolone group was (10 +/- 6) nmol choline . h-1 . mg-1 that was lower than control (P < 0.05), while it had no statistical difference compared with that of pre-CPB. In methylprednisolone group, PLD activity elevation was postponed to the time point of CPB stopped. There was no statistical difference in PLD activity between aprotinin group and control (P > 0.05). After administration of methylprednisolone or aprotinin, leukocyte CD11b expression, plasma IL-6, IL-8, C-reactive protein levels, and MPO activity decreased by different extent. CONCLUSION: Leukocyte PLD activity was elevated significantly in systemic inflammatory response induced by CPB and methylprednisolone partially blunted the CPB-induced inflammatory response by inhibiting PLD activity.

[Amphotericin B for treatment of fungal infections in 40 patients with malignant hematologic diseases].

OBJECTIVE: To observe the therapeutic effect and side effects of amphotericin B for fungal infections in patients with malignant hematologic diseases. METHODS: 40 patients (male 27, female 13; average age 35.5 years) with malignant hematologic diseases were given amphotericin B, 5-50 mg/d per day for 5-85 days (average time 21 days). RESULTS: The clinical efficacy rate of amphotericin B was 52.5%, and the fungal elimination rate was 56.2%. Among the side effects, rigor and fever were present in 2.5% of the patients. Hypokalaemia was found in 12.5%, hepatotoxicity in 15.0% and nephrotoxicity in 15.0%. CONCLUSION: As amphotericin B has a broad anti-fungal spectrum and relatively good efficacy, it is still a high-efficiency drug in treatment of systematical fungal infections. However, the use of drug is limited because of its many side effects. Our study indicates that if it is used properly and hepatic and renal function tests are carried out regularly, amphotericin B is a relatively safe and effective drug.

[A model of calibrated optic nerve crush injury in rats].

OBJECTIVE: To establish a calibrated optic nerve crush injury model in rats. METHODS: Twenty-eight healthy Sprague-Dawley (SD) rats were randomly divided into two groups. Seven rats were in normal control group, i.e. group A. In group B, model of partial optic nerve crush injury group, partial optic nerve crush injury model was induced in the right eyes of another 21 rats by a special designed optic nerve clip with 40-gram holding force at optic nerve 2 mm behind the eyeball clipped for four seconds to partially block the optic nerve axoplasmic transport. The left eyes served as a control. According to the sacrificing date of the rats, three subgroups in group B were further divided. Therefore, seven rats were in each subgroup. One, eleven and eighteen days later, 3% fast blue was injected into both superior colliculi to each subgroup and group A. The eyes were enucleated another three days later, flat mounts of the retinal from both eyes were prepared on a slide and observed under a fluorescence microscope. Four photos with 400 x magnification were taken from quadrants of the retina 1 mm away from the optic disc. The labeled retinal ganglion cells (RGCs) photos were counted by a computerized image analyzer. The labeled RGCs rate, a comparison of the labeled RGCs from both eyes in a rat were used for statistical analyzing. (The labeled RGCs rate = RGCs from injured right eye/RGCs from uninjured left eye x 100%). RESULTS: In group A, the group of normal control group, the labeling rate from the ratio of RGCs counting of right eye to left eye was 99.79% +/- 13.05% and from the ratio of the RGCs of left eye to right eye was 101.86% +/- 13.91%. In group B, model of partial optic nerve crush injury group, the labeled RGCs rate were 77.79% +/- 7.11%, 63.76% +/- 3.79% and 54.66% +/- 4.75% in day 4, day 14 and 21 respectively. CONCLUSION: Using a special designed clip with 40-gram holding force constantly pressed for four seconds to create an optic nerve crush injury model in rats. As time past along, it is revealed that the labeled RGCs rate gradually reduced.

[CT and MRI and clinical study of chordoma in the skull base].

OBJECTIVE: To investigate the diagnostic features of CT and MRI in chordoma of the skull base (CSB) and to estimate their value of clinical application. METHODS: Sixteen patients with CSB were performed CT and MRI examinations of the head, and 13 patients with nasopharyngeal carcinoma (NPC) and 11 patients with pituitary adenoma (PA) which invaded the skull base were selected as control. In addition, 7 cases of chordoma were performed multiple planar reconstruction (MPR) of MRI. RESULTS: MRI was superior to CT in depicting the location and the extent of CSB. The bone destruction at mid-line skull base was demonstrated equally well on both CT and MRI. Based on the location of the tumor displayed on MRI, the CSB could be classified into selloclival, nasopharyngeal, and occipito-temperal types. The T2 weight imaging(T2WI) signal intensity of chordoma was significantly higher than those of NPC and PA (P < 0.01). On MPR images of MRI, oppressed and displaced optic nerves caused by chordoma were noted in 3 cases and encased optic nerves in 4 cases, encased abducent nerves in 7 cases with abducent paralysis and mildly oppressed oculomotor nerves in 3 cases. CONCLUSION: The extremely high T2WI signal intensity is the characteristic of MRI finding in CSB and has great differential value for the tumors of the skull base. The MPR of MRI plays an important role in the surgical treatment for these tumors.

Study on TEMPO-mediated selective oxidation of hyaluronan and the effects of salt on the reaction kinetics.

2,2,6,6-Tetramethyl-1-piperidinyloxy radical (TEMPO)-mediated oxidation of hyaluronan was studied at pH 10.2 and temperature of 0 degrees C with NaOCl as the primary oxidant. As with other polysaccharides, a high selectivity of oxidation was observed. The degradation of the polymer was essentially caused by the oxidation process. The primary oxidant and the pH of the reaction mixture did not alter the molecular weight of hyaluronan during oxidation. The kinetics of the oxidation process was investigated at different concentrations of reactants and the inorganic salts, NaBr, NaCl, and Na2SO4. An increase in the salt concentration in the mixture causes a major decrease in the rate of the oxidation, and this decrease is independent of the nature of the salt.

Purification and partial characterization of fructosyltransferase and invertase from Aspergillus niger AS0023.

Fructosyltransferase (EC.2.4.1.9) and invertase (EC.3.2.1.26) have been purified from the crude extract of Aspergillus niger AS0023 by successive chromatographies on DEAE-sephadex A-25, sepharose 6B, sephacryl S-200, and concanavalin A-Sepharose 4B columns. On acrylamide electrophoresis the two enzymes, in native and denatured forms, gave diffused glycoprotein bands with different electrophoretic mobility. On native-PAGE and SDS-PAGE, both enzymes migrated as polydisperse aggregates yielding broad and diffused bands. This result is typical of heterogeneous glycoproteins and the two enzymes have proved their glycoprotein nature by their adsorption on concanavalin A lectin. Fructosyltransferase (FTS) on native PAGE migrated as two enzymatically active bands with different electrophoretic mobility, one around 600 kDa and the other from 193 to 425 kDa. On SDS-PAGE, these two fractions yielded one band corresponding to a molecular weight range from 81 to 168 kDa. FTS seems to undergo association-dissociation of its glycoprotein subunits to form oligomers with different degrees of polymerization. Invertase (INV) showed higher mobility corresponding to a molecular range from 82 to 251 kDa, on native PAGE, and from 71 to 111 kDa on SDS-PAGE. The two enzymes exhibited distinctly different pH and temperature profiles. The optimum pH and temperature for FTS were found to be 5.8 and 50 degrees C, respectively, while INV showed optimum activity at pH 4.4 and 55 degrees C. Metal ions and other inhibitors had different effects on the two enzyme activities. FTS was completely abolished with 1 mM Hg(2+) and Ag(2+), while INV maintained 72 and 66% of its original activity, respectively. Furthermore, the two enzymes exhibited distinctly different kinetic constants confirming their different nature. The K(m) and V(m) values for each enzyme were calculated to be 44.38 mM and 1030 micromol ml(-1)min(-1) for FTS and 35.67 mM and 398 micromol ml(-1) min(-1) for INV, respectively. FTS and INV catalytic activity was dependent on sucrose concentration. FTS activity increased with increasing sucrose concentrations, while INV activity decreased markedly with increasing sucrose concentration. Furthermore, INV exhibited only hydrolytic activity producing exclusively fructose and glucose from sucrose, while FTS catalyzed exclusively fructosyltransfer reaction producing glucose, 1-kestose, nystose and fructofuranosyl nystose. In addition, at 50% sucrose concentration FTS produced fructooligosaccharides at the yield of 62% against 54% with the crude extract.
