Replication-defective adenovirus infection reduces Helicobacter felis colonization in the mouse in a gamma interferon- and interleukin-12-dependent manner.

Helicobacter infection leads to chronic inflammation of the stomach. Although the infection persists in spite of an immune response, animal studies have shown that adjuvant-based oral vaccines can protect against infection and even eliminate established infection. These vaccines are thought to induce a Th2 immune response, counterbalancing the Th1 response seen with natural infections. As a prelude to using adenovirus vectors carrying cytokine genes to modulate the immune response to established Helicobacter felis infection, we first examined the effect of the replication-defective adenovirus (RDA) vector itself. C57BL/6 mice chronically infected with H. felis (8 to 10 weeks) received intramuscular injections of RDA. The effect of RDA on the severity of H. felis colonization and the degree of gastric inflammation was assessed 2 weeks later. RDA caused a significant decrease in H. felis colonization without significantly altering the associated inflammation. RDA did not alter the H. felis-specific immunoglobulin G1 (IgG1), IgG2a, and IgA responses in the serum but was associated with an increase in gamma interferon (IFN-gamma)-producing CD8(+) spleen cells. To determine if IFN-gamma or Th1 cytokines were involved in the response to RDA, we examined RDA treatment of H. felis infection in mice lacking either IFN-gamma or interleukin-12 (IL-12). RDA failed to alter H. felis colonization in either of these two mouse strains. Thus, viral infection of mice chronically infected with H. felis led to a significant decrease in H. felis colonization in an IFN-gamma- and IL-12-dependent manner. These results demonstrate that Th1 responses associated with systemic viral infection can influence an established H. felis infection.

N-acetyl-L-cysteine enhances interleukin-1beta-induced nitric oxide synthase expression.

The effect of N-acetyl-L-cysteine on interleukin-1beta-induced nitric oxide synthase expression was studied in rat vascular smooth muscle cells to determine if the reduction/oxidation state would modulate cytokine-induced changes. Interleukin-1beta induced the production of nitrite, a stable metabolite of nitric oxide in a time- and dose-dependent manner. Cytokine-induced nitrite production was enhanced by the addition of N-acetyl-L-cysteine in a dose-dependent manner, with a >50% increase produced by the addition of 1 mmol/L N-acetyl-L-cysteine. There was no influence on nitrite production when the cells were treated with N-acetyl-L-cysteine alone. Northern and Western blot analyses revealed that the upregulation of interleukin-1beta-induced nitric oxide production by N-acetyl-L-cysteine resulted from an enhanced expression of inducible nitric oxide synthase. Interferon-gamma or tumor necrosis factor-alpha when used alone had no influence on nitrite production in the absence or presence of N-acetyl-L-cysteine. Nitrite accumulation was higher by the cells treated with interleukin-1beta combined with either interferon-gamma or tumor necrosis factor-alpha compared with those treated with interleukin-1beta alone. N-Acetyl-L-cysteine upregulated nitrite production and inducible nitric oxide synthase expression induced by combination treatment with interleukin-1beta and either interferon-gamma or tumor necrosis factor-alpha. However, N-acetyl-L-cysteine had no significant influence in cytokine-induced activation of nuclear factor-kappaB or signal transducer and activator of transciption-1, as assessed by electrophoretic mobility shift assays. These results demonstrate that N-acetyl-L-cysteine possibly acted as a thiol-containing reducing agent and facilitated the expression of inducible nitric oxide synthase in rat vascular smooth muscle cells by cytokines through a mechanism that is independent of nuclear factor-kappaB or signal transducer and activator of transciption-1.

Lack of effect of McN-A-343 on membrane current and contraction in guinea pig ventricular myocytes.

We asked whether agonist occupancy of M(1) muscarinic receptor (mAChR) causes increased L-type Ca(2+) [I(Ca(L))] and contractions in ventricular myocytes. Voltage-clamp pulses evoked I(Ca(L)) in guinea pig ventricular myocytes superfused with Tyrode's solution (22-24 degrees C). The mAChR agonists carbachol (Cch, nonselective), McN-A-343 (McN, M(1)-selective), and oxotremorine (Oxo, M(2)-selective) were tested at 0.1 mM. None of these agonists affected basal I(Ca(L)). McN did not change isoproterenol-stimulated I(Ca(L)) in 13 of 15 cells. The slight decrease in two cells was not muscarinic because atropine (1 microM) did not antagonize it. Carbachol or Oxo decreased isoproterenol-stimulated I(Ca(L)) by 87 +/- 6.7 (n = 8 cells) and 49 +/- 9.0% (n = 4 cells), respectively. Atropine blocked inhibition by Cch or Oxo. External stimulation evoked contractions of single myocytes (35 degrees C). McN increased contraction in 1 of 22 cells stimulated at 0.2 Hz and in 0 of 16 cells stimulated at 1.0 Hz. Carbachol significantly increased contraction in 10 of 15 cells at 0.2 Hz and in 8 of 10 cells at 1.0 Hz stimulus frequency. Summarily, the M(1)-selective agonist McN had a negligible role to regulate I(Ca(L)). The antiadrenergic effect of mAChR agonists is attributable to M(2) receptor occupancy. That Cch, but not McN, increased cell shortening excludes participation of M(1) mAChR in the stimulant effect of Cch on guinea pig ventricular myocyte contractions.

An outbreak of group C rotavirus gastroenteritis among children attending a day-care centre in Belém, Brazil.

In August 1993, an outbreak of group C rotavirus-associated gastroenteritis occurred among children attending a day-care centre in Belém, Brazil. Of the 64 children, 21 (33%) became ill. Group C rotavirus was identified in faecal specimens from 8 (38%) children with diarrhoea by electron microscopy (EM) and an enzyme immunoassay (EIA), using antibodies specific to the Cowden strain of porcine group C rotavirus. By polyacrylamide gel electrophoresis (PAGE), a pattern similar to that of group C rotavirus was observed in 5 (62.5%) of the 8 EM- and EIA-positive samples. These 5 faecal samples were confirmed to be positive for group C rotavirus by reverse transcriptase-polymerase chain reaction, using specific VP6 and VP7 primers. This is the first report of an outbreak of diarrhoea in North Brazil associated with group C rotavirus. These findings suggest that group C rotavirus may be an important aetiological agent of diarrhoea in this region, which requires further study.

[Colorectal cancer: lymphatic metastasis and choice of operation].

OBJECTIVE: To study lymphatic metastasis of colorectal cancer for deceding the of dissection extent of dissection. METHODS: Six hundred and twenty-three lymph nodes from 74 cases of colorectal cancer were analyzed after D(3) resection. They were classified into peritumor, longitudinal spread, and upward spread groups. The metastasis rate of lymph node and its incidence were calculated. RESULTS: The metastasis rate of lymph node and its incidence in the peritumor, longitudinal spread, and upward spread (N(2) and N(3)) groups were 47.2% and 38.6%, 22.8% and 18.6%, and 29.7% (20.3% and 9.4%) and 10.2% (19.0% and 5.3%), respectively. The distribution rate of metastatic lymph node was 61.2%, 17.1%, and 21.6% in the peritumor, longitudinal spread, and upward spread groups, respectively. Within 2.0 cm on the anal side of rectal cancer, the metastasis rate was 3.7%, whereas in 2.0 - 4.0 cm no metastasis was observed cancer at the lower part of the rectum demonstrated a higher lateral metastasis rate. CONCLUSIONS: Colorectal cancer tends to metastasize to longitudinal and the risk of metastasis increased in the stage of pT(3) and pT(4) or low differentiation stage. Jump metastasis is also a feature. D(3) radical resection is necessary for colorectal cancer, but extended lateral lymph node needs enectomy or total mesorectal excision. No residual tumor tissue exists at the anastomosis site after low anterior resection when the excision distance is beyond 2.0 cm from the anal margin of rectal cancer.

[PCR-based DNA analysis of vWA31A, TH01, F13A01, FESFPS, TPOX and CSF1PO].

The allele and genotype frequencies of 6 tetranucleotide STR loci were investigated in a sample of 132 unrelated individuals from Chinese Han population. The PCR products were analyzed on 6% denaturing PAGE and detected using fluorescently labeled primers in an automated 377 sequencer(PE). All loci meet Hardy-Weinberg equilibrium. There was no random association of alleles among the 6 loci. The allele frequencies were compared with other population databases. Except locus vWA31A, the observed heterozygosity at other 5 loci was significantly lower than that reported in Caucasian and Black population studies. The calculated DP = 0.99999, PE = 0.9708, pM = 1.059 x 10(-5). The allelic frequency data can be used in forensic identification and paternity testing.

[Clinical analysis of non-Hodgkin's lymphoma in head-facial].

OBJECTIVE: To explore the clinical features, diagnosis and treatment of non-Hodgkin's lymphoma in head facial. METHOD: The clinical data of 32 cases non-Hodgkin's lymphoma in head facial were analysed retrospetively. RESULT: The rate of diagnosis was low in early. It frequently misdiagnosed as chronic inflammation, malignant granulomatosis, epithelial cancer, and undifferentiated cancer, etc. CONCLUSION: The biopsy shoud be performed routinely and it could do repeatly. It is necessary to do immunohistochemical examination. Only by earlier diagnosis, prompt and reasonable treatment, the curative effect would be better.

[Noso-orbital related disease: an analysis of 39 cases].

OBJECTIVE: To enhance the Diagnostic and therapeutic levels of naso-orbital diseases. METHOD: Thirty-nine cases of naso-orbital diseases were analyzed retrospectively. RESULT: Among the 39 cases, 7 had no nasal symptoms. 9 had their symptoms mainly confined to eyes and were initially consulted with ophtalmologists. 1 case was consulted with neurologist at first. The main pathologies were as following: bony absorption of orbit wall in inflammatory disorders; compression, displacement and bony absorption of orbit wall in benign tumors and mucoceles; bony destruction in malignant tumors. Tranmatic fractures might involve multiple bony structures. CONCLUSION: Nosal and orbital organs are closely related anatomically. Imaging investigations are important in diagnosis of naso-orbital related diseases. CT is helpful for early diagnosis. Treatment for benign tumors is mainly by operation, while malignant tumors should be treated by operation and radiotherapy together.

[Combined window genyantrum in middle and inferior meatus under sinus endoscopy].

OBJECTIVE: For clearing away process of genyantrum and preventing its recurence. METHOD: 36 patients (40 sinuses) were treated with combined window in middle and inferior nasal meatus under sinus endoscopy. The process was cleared away completely. RESULT: There is no complication after one year following up. The cure rate is 100% and the recurrence rate is 0. CONCLUSION: It shows that the method has the advantage of clearing away the process of genyantrum and preventing recurence.

Synthesis of rotavirus-like particles in insect cells: comparative and quantitative analysis.

When the three major structural proteins, VP2, VP6, and VP7, of rotavirus are co-expressed in insect cells infected with recombinant baculoviruses, they self-assemble into triple-layered virus-like particles (VLPs) that are similar in morphology to native rotavirus. In order to establish the most favorable conditions for the synthesis of rotavirus VLPs, we have compared the kinetics of 2/6/7-VLP synthesis in two different insect cell lines: High Five cells propagated in Excell 405 medium and Spodoptera frugiperda 9 cells in Excell 400 medium. The majority of VLPs produced in both cell lines were released into the culture medium, and these released VLPs were predominantly triple-layered and were found to be stable for the period of six or seven days examined. The optimal synthesis of VLPs depended upon the cell line and the culture medium used as well as the time of harvesting infected cell cultures. The highest yield of VLPs was obtained from High Five cultures in the late phase of infection when the yield was at least 5-fold higher than that from S. frugiperda 9 cultures on a per cell basis. Our results demonstrate the usefulness of High Five cells for the production of VLPs as potential rotavirus subunit vaccines.

Formation of plasmid DNA strand breaks induced by low-energy ion beam: indication of nuclear stopping effects.

Plasmid pGEM 3zf(+) was irradiated by nitrogen ion beam with energies between 20 and 100 keV and the fluence kept as 1x10(12)ions/cm2. The irradiated plasmid was assayed by neutral electrophoresis and quantified by densitometry. The yields of DNA with single-strand and double-strand breaks first increased then decreased with increasing ion energy. There was a maximal yield value in the range of 20-100 keV. The relationship between DNA double-strand breaks (DSB) cross-section and linear energy transfer (LET) also showed a peak-shaped distribution. To understand the physical process during DNA strand breaks, a Monte Carlo calculation code known as TRIM (Transport of Ions in Matter) was used to simulate energy losses due to nuclear stopping and to electronic stopping. It can be assumed that nuclear stopping plays a more important role in DNA strand breaks than electronic stopping in this energy range. The physical mechanisms of DNA strand breaks induced by a low-energy ion beam are also discussed.

Expression of parathyroid hormone/parathyroid hormone-related protein receptor in vascular endothelial cells.

The parathyroid hormone (PTH)/PTH-related protein (PTHrP) receptor has been reported to be expressed in many tissues, including vascular smooth-muscle cells (VSMCs), but it has not been identified in vascular endothelial cells. To determine whether vascular endothelial cells can express the PTH/PTHrP receptor, its gene expression was examined in simian virus 40-transformed rat lung vascular endothelial cells (TRLECs) by the reverse-transcription polymerase chain reaction (RT-PCR). Results in TRLEC, with rat VSMCs and kidney as controls, showed identical 741-bp products. Furthermore, incubation with PTHrP[1-34] reduced the thrombin-stimulated endothelin-1 (ET-1) expression in TRLECs. Our results demonstrate that vascular endothelial cells can express the PTH/PTHrP receptor and therefore are also a target tissue for PTHrP.

The effect of bu shen sheng xue fang on beta-thalassemia at gene level.

Isoelectrofocusing electrophoresis, PCR-SSCP analysis with DNA direct sequencing and RT-PCR were used to analyze the constituents of hemoglobin, determine the pattern of globin gene mutation, and detect its mRNA transcription extent. The results indicated that Bu Shen Sheng Xue Fang (BSSXF) significantly increased the contents of hemoglobin (Hb) and fetal hemoglobin (HbF), and the chain ratio in patients with beta-thalassemia. The drug is markedly effective in heterozygous, but not effective in homozygous or genetic compound patients. It was suggested that the beta-globin gene defects were compensated by elevating the gamma/beta + gamma ratio, promoting the transcription and expression of gamma-globin gene in synthesizing HbF.

[Study on electroneuromyography of 175 workers exposed to carbon disulfide].

A study on electroneuromyography of 175 workers exposed to carbon disulfide demonstrated that electroneuromyographic abnormalities of these workers were in accordance with characteristic alteration of axonal polyneuropathy. Electroneuromyography was an objective and sensitive method to detect the early damage of peripheral nerve caused by low-level exposure to carbon disulfide. The dose-response relationship between the exposure to carbon disulfide and the damage of peripheral nerve was demonstrated.

[Speech reconstruction after laryngectomy by using modified Pearson's operation].

Since 1990, 16 patients received speech reconstruction after laryngectomy by using modified Pearson's operation. Among them, T3, T4 laryngeal cancer 12 cases, T4 hypopharyngeal cancer 4 case, 14 were male and 2 were female, the eldest was 73 years old and the youngest 51 years old, and the average age was 58 years. In the operation, the tracheo-pharyngeal speech tube were sewn up with mucosa remained at laryngopharynx. During 1-5 years follow up, the speech function and swallow function were satisfactory. The 3 and 5-year survival rates were 77.78% and 60.00% respectively. They obtained a good long-term speech function except one. It showed that speech reconstruction with the modified Pearson's operation has important value to advanced pharyngeal and laryngeal carcinoma.
