[Aldolase activity and the lactate level in the cord blood as objective indicators of the status of newborn infants by the Apgar score].

In 63 newborns with general clinical status evaluated by means of the Apgar score the lactate level and aldolase activity were determined in cord blood. A greatly raised mean level of both these biochemical indicators of tissue hypoxia was demonstrated in newborns with low Apgar score. A considerable scatter of the determined biochemical parameters was noted in newborns born in serious condition and having identical low Apgar score.

[Effect of marked obesity on the emotional status of children].

Fifty children with overweight exceeding 50% of the expected weight were studied for establishing the occurrence of emotional disturbances caused by lack of acceptance by peers. Obese children were found to suffer often from discrimination by their schoolmates which was, in some degree, connected with their low fitness. This may lead to various emotional disturbances requiring even psychotherapeutic measures.

Intranasal LH-RH analogue treatment of precocious puberty.

The agonistic analogues of luteinizing hormone releasing hormone were used in the treatment of precocious puberty (pp) in children. 12 children with pp were treated for 6 months with an intranasal LH-RH analogue (C6-C3-C2-naphtyl/D-alanine)-LHRH. The majority of the children had central precocious puberty. Only one child had peripheral pp. Laboratory assessment of estradiol, testosterone, LH and FSH levels revealed that 1,000 micrograms/day of this analogue diminished the hormone levels only in the group of children with central and combined pp. Bone age, pelvic ultrasound examination, sexual development and growth rate were determined pre-, during and after the treatment. The growth velocity decreased and gonadal and sexual characteristic ceased advancing after the LH-RH therapy. Biochemical and hematological studies performed in our children did not show any significant changes during the treatment periods. No adverse effects were noted from the intranasal therapy. We conclude, that intranasal therapy is safe, convenient and effective in pp, but not of peripheral origin.

Clonal growth of mouse epidermal cells in medium with reduced calcium concentration.

Mouse keratinocytes can be grown at clonal densities in dermal fibroblast conditioned medium with a calcium concentration of 0.02 MM. Colony forming efficiencies of approximately 1-3% can be achieved with primary and secondary cultures and up to 6% with selected subclones. A substrate of frozen-thawed dermal fibroblasts enhances colony formation over plastic. Colony size increases more rapidly in the presence of epidermal growth factor in conditioned medium. Conditioning of medium by fibroblasts is optimum after day 6 of culture and conditioned medium can be sotred at -20 degrees C for at least 4 mo.

Benign paroxysmal vertigo of childhood: a migraine equivalent.

Eight patients with the syndrome of benign paroxysmal vertigo of childhood are presented. This symptom complex is characterized by attacks of vertigo in young children combined with nystagmus, ataxia and transiently decreased vestibular function, but without impaired consciousness. Diagnostic and follow-up studies revealed a close relationship to other forms of autonomic nervous system instability, particularly to the migraine symptom complex.

Quantitative assay for carcinogen altered differentiation in mouse epidermal cells.

Basal epidermal cells can be selectively maintained as a monolayer in culture medium containing a low ionic calcium concentration of 0.01-0.10 mM. Cessation of proliferation, maturation and shedding of squamous sheets can be induced in this population by increasing the calcium concentration above 0.1 mM. Since alterations in the regulation of proliferation and differentiation are associated with epidermal carcinogenesis in vivo, it appeared reasonable that changes in the phenotypic response to calcium might follow exposure to carcinogens in vitro. Support for this hypothesis was provided by the observation that malignant epidermal cells continued to proliferate when switched from low to high calcium medium, and could thus be selected from a mixture of such cells and a large excess of normal cells which did not survive after induced differentiation. Normal primary epidermal cells were plated in low calcium medium, treated on day 3 with a chemical carcinogen, maintained for 3-9 weeks in low calcium (0.02 mM) and then switched to high calcium medium (1.4 mM). After an additional 4 weeks, surviving epithelial colonies were fixed, stained with rhodamine and counted. Treatment of cultures with 7,12-dimethylbenz[a]anthracene or N-methyl-N'-nitro-N-nitrosoguanidine yielded 4-10 fold more colonies than solvent controls. Colony number was proportional to carcinogen dose for both agents, and increased with time in low calcium prior to selection by calcium increase. Cells obtained from colonies in treated cultures demonstrated characteristic epidermal morphology and keratinization, and could be subcultured, but did not grow in agar or produce tumors in syngeneic hosts. This model system represents a quantitative assay for carcinogen altered epithelial cell differentiation and may select for an early property of preneoplastic epidermal cells.
