4-Ethoxyamphetamine: effects on intracranial self-stimulation and in vitro uptake and release of 3H-dopamine and 3H-serotonin in the brains of rats.

Experiments were conducted to compare the effects of 4-ethoxyamphetamine, a novel "designer" amphetamine, with (+)-amphetamine and an earlier "designer" amphetamine, 4-methoxyamphetamine, on rats. (+)-Amphetamine significantly decreased frequency threshold measures in an intracranial self-stimulation (ICSS) procedure using medial forebrain bundle electrodes, while 4-methoxyamphetamine and 4-ethoxyamphetamine increased these ICSS frequency thresholds. 4-Methoxyamphetamine and 4-ethoxyamphetamine had more potent effects on inhibition of uptake and stimulation of spontaneous release of 5-hydroxytryptamine (serotonin) than of dopamine. It is concluded that the neuropsychopharmacological profile of 4-ethoxyamphetamine is unlike that of (+)-amphetamine, but similar to that of 4-methoxyamphetamine, a potent hallucinogen in humans.

Analysis of estrogen sulphate mixtures in pharmaceutical formulations by reversed-phase chromatography.

The relative retention times of estrogen sulphates for reversed-phase, ion-pair and argentous chromatography have been determined. Detector wavelengths of 200 nm, 200 nm and 270 nm were used, respectively. Samples are prepared by dissolution of pharmaceutical formulations in the high-performance liquid chromatography (HPLC) eluent. With reversed-phase chromatography, the estrone, equilin and 17 alpha-dihydroequilin estrogen sulphates are resolved from other estrogen sulphates in 20 min. Any traces of free estrogens which might be present are eluted too slowly for the formation of chromatographic peaks. With ion-pair chromatography, any hydrolyzed estrogens would be eluted before the sulphates with the formation of resolved chromatographic peaks. With argentous chromatography the retention times of the estrogen sulphates varied by nearly a factor of four. The utility of HPLC for the analysis of estrogen sulphates in pharmaceuticals is discussed.

Language in dementia of recent referral.

Twenty-four recently referred patients with dementia were assessed on a range of language tests and a mental status test. The tasks which appeared to present the most difficulties for the patients were written spelling, pragmatic processing tasks like sentence disambiguation and proverb interpretation. 'Straight' linguistic processing tasks sensitive to aphasia, like oral reading, serial identification of objects, naming and correction of semantically incorrect sentences, appeared to present fewer problems. It is concluded that certain language tasks may be useful and sensitive detectors of developing dementia, and that in early dementia those aspects of language which depend on straight linguistic processing can be relatively preserved.

Geriatric medicine in a community hospital.

A study is reported of the care that was being received by the patients over the age of 60 years in general practitioner community hospital beds at Abingdon Hospital in Berkshire. The appropriateness of this care is discussed and measures are suggested for improving it. The practical working relationship between general practitioners attending the community hospital and the local consultant physicians in geriatric medicine is also discussed and again measures for improving their combined services to the community hospital are suggested.
