[Design of a nutrient medium for bacteria of the genus Haemophilus--producers of restriction endonucleases].

A culture medium for the cultivation of hemophilic bacteria, containing acidic casein hydrolysate, aminopeptide and fodder yeast extract, has been proposed. The growth-stimulating properties of this medium have been studied on 5 strains producing restrictases differing in their specificity. In growing these producer strains in a model ANKUM-2 fermenter with the supply of carbohydrate substrates (glucose, sucrose, glycerin) the yield of biomass, considered to be high for hemophilic bacteria (10-14 g of humid substance from 1 liter of the medium), has been achieved. As shown on H. influenzae Rc B-2297 used as an example, an increase in the yield of microbial biomass leads to a decrease in restrictase specific activity.

[Effect of the composition of the nutrient media and culture conditions on the growth of strains of Campylobacter jejuni and C. coli].

The influence of the composition of culture media and cultivation conditions on the growth of Campylobacter strains has been studied. Blood-containing media have been found to ensure the most favorable growth conditions. No differences between the use of RCT supplement (rifampicin, cephalothin and trimethoprim in amounts of 10, 15 and 5 mg/l respectively) and DC supplement (0.1% of sodium deoxycholate and 15 mg/l of cephalothin), as well as between different methods used for the creation of microaerophilic conditions, have been established.

[Cultivation of the gonococcus in nutrient broths].

The dynamics of the multiplication of gonococci and the parameters of their growth have been studied in the process of batch cultivation in liquid culture media with different content of bovine blood serum. The protective and stimulating action of the serum on the growth of gonococci has been shown. The optimization of the process of cultivation has been carried out; as a result, the growth of test strains in a culture medium containing no serum has been achieved. Phasic changes in the ultrastructure of gonococci in the process of their cultivation in liquid media have been followed. 9- to 12-hour cultures of gonococci grown in liquid culture media have been found the most valuable and physiologically active.

[Effect of the peptide and amino acid composition of nutrient bases on the parameters of microorganism growth].

The parameters of the growth of three microbial strains cultivated in growth media prepared with the use of different nutrient bases, both locally produced and imported, have been studied. The characteristics of the processes of bacterial cultivation have been found to correlate with the peptide composition of hydrolysates used in the experiment. Nutrient bases containing a wide spectrum of peptides with different molecular weight have ensured the most favorable conditions for the growth of microorganisms belonging to the taxonomic groups under study.

[Development and experimental study of a dried nutrient medium for the microbiological diagnosis of streptococcal infections].

The comparative study of different protein bases has shown that the combined base containing animal blood hydrolysate (amino peptide) and acidic casein hydrolysate, moderately cleaved, in the proportion 1:1 is a good source of nitrogen and ensures the intensive growth of streptococci. As determined by the study of the physiological parameters and growth of streptococci, the presence of fodder yeast extract, glutamine, glucose and phosphates in media containing blood hydrolysate and casein hydrolysate has been found to render a stimulating effect on the growth and multiplication of these organisms. The data thus obtained have been used as the basis for developing the formula of a dried culture medium, capable of ensuring the growth of streptococci without blood or serum added and not inferior in its quality to Todd-Hewitt Broth manufactured by Oxoid Ltd. (Great Britain) and Difco Laboratories (USA). The physico-chemical and physiological characteristics of the proposed medium have been determined. The use of the new dried culture medium in medical practice will make it possible to improve the microbiological diagnosis of streptococcal infections.

[Peptide and amino acid composition of different protein bases for culture media].

The enzymatic hydrolysates under study, obtained from different raw materials, have been shown to contain a great variety of peptides with different molecular weight. The highest content of fractions with a molecular weight of 2000 D has been observed in enzymatic meat and casein hydrolysates manufactures in the GDR. Low-molecular fractions (100-200 D) prevail in amino peptide. A great variety of peptides with different molecular weight is observed in Hottinger's meat hydrolysate and in blood clot hydrolysate obtained from the blood of laboratory animals. All peptide fractions have been shown to contain a wide spectrum of free amino acids. These data on the peptide and amino acid composition of different protein bases facilitate their rational use of microbiological culture media.

[Electron microscopic study of microorganisms grown on nutrient media with a nonnutrient raw material base].

A comparative electron-microscopic study of Sh. flexneri 8512, Str. Dick Ia and Cor. pseudodiphthericum test strains, grown in experimental culture media prepared from inedible raw materials and in currently used culture media based on fish and meat, has been carried out. The ultrastructure of the test strains grown in culture media prepared from inedible raw materials has been shown to remain unchanged.

[Comparative study of different substrates as the nutrient base of a culture medium for Bacteroides and Fusobacterium].

The results of the comparative study of different substrates prepared from foodstuffs and inedible raw materials and intended for use as the nutrient base of a medium for cultivating the organisms of the genera Bacteroides and Fusobacterium have demonstrated that commonly used nutrient bases prepared from meat, fish, casein and inedible animal blood (blood substitutes) are suitable for this purpose. The dried nutrient base prepared from the mixture of outdated blood substitutes (hydrolysin and casein hydrolysate) (see text for Russian letters) has been found to ensure the optimum growth of Bacteroides and Fusobacterium.

[Development and experimental study of a dry selective medium for staphylococci].

A new dried selective medium for the cultivation of staphylococci has been developed. The medium is based on the mixture of hydrolysin past its expiration date and aminopeptide. To inhibit extraneous microflora, sodium chloride in a high concentration is introduced into the medium. The possibility of using this selective medium in the form of milk-yolk-salt agar for the determination of the pigment formation and lecithinase activity of staphylococci is shown.
