Gingival fibromatosis, short stature, border-line IQ, facial dysmorphism and hepatomegaly.

Gingival fibromatosis, short stature, border-line IQ, facial dysmorphism and hepatomegaly: Gingival fibromatosis is a rare and benign disorder. The enlarged gingivae are firm and may interfere with speech, closure of the lips, and mastication. We report a thirteen years old girl, with gingival fibromatosis referred to the periodontics clinics. Full mouth gingivectomy and gingivoplasty were performed. Medical investigation showed short stature, low-borderline IQ, facial dysmorphism, and hepatomegaly. Histological analysis revealed hyperplasia in the epithelial area and fibrotic appearance of gingival connective tissue with dense collagen fibre clusters. Pedigree analysis confirmed that mode of inheritance is autosomal recessive. According to the combination of clinical findings, this case report may represent a previously unreported syndrome.

Increased cell proliferation and R.Msp1 fragmentation induced by 5-aza-2'-deoxycytidine in rat testes related to the gene imprinting mechanism.

DNA methylation is one of the crucial mechanisms for cellular and tissue differentiation during developmental stages in mammals. 5-aza-2'-deoxycytidine, a specific cytosine DNA Methyltransferase inhibitor, is known to cause DNA hypomethylation in CpG, CpNpG and CCGG sequences. Therefore the present study was designed to determine the effects of 5-aza-2'-deoxycytidine on the germinal cells of the adult rat testicular tissue. Rat testicular tissues from the 5-aza-2'-deoxycytidine treated experimental and non-treated control groups were processed for light microscopy and also for genomic DNA isolation assays. The isolated genomic DNAs were digested with R.Msp1 in order to determine the methyl pattern differences in the enzyme cognate CCGG sequence. Testicular tissues from treated rats showed increased cell proliferation when investigated at the light microscopical level. On the other hand, genomic DNA of these proliferative tissue showed high fragmentation sizes of R.Msp1 digestion when compared to controls. While the R.Msp1 digested control group DNA fragmentation condensed at approximately 4700-5100 bps size, the experimental group DNA fragmentation was condensed at 700-900 bps size. In addition, 5-aza-2'-deoxycytidine has effects on increased cell proliferation via the loss of somatic de novo gene imprinting. These results imply that abnormally imprinted normal somatic cells in mammals are susceptible to epigenetic modification. These results also suggest that the genomic DNA of testicular tissues from control rats is resistant to R.Msp1 while DNA from the experimental group testicular cells demonstrating high proliferation rate could not resist to R.Msp1 digestion due to DNA hypomethylation in CCGG sequence. In conclusion, it could be suggested that the reversal of gene imprinting in germinal cells may cause an increased cellular proliferation and R.Msp1 fragmentation when induced by 5-aza-2'-deoxycytidine.

Comparison of the effects of vasectomy and experimental cryptorchidism in rats: I. Testicular histology.

The effects of cryptorchidism, one of the most important congenital malformations resulting with the defects in testicular structure thus infertility, is the best known subject in male reproduction. Those best known effects make the usage of cryptorchidism as a second control criteria when the effects of other male infertility reasons are attempted to be determined. Therefore the persistency or reversibility of the effects of vasectomy on testicular structure have been compared to the testicular features following experimental cryptorchidism and to the control group findings. To do this, rats were divided into 3 groups. While the control group animals were sham operated, the experimental group animals were undergone either vasectomy or experimental cryptorchidism procedures. Testicular histology of the control and experimental groups was determined using convenient staining methods at the light microscopical level. The comparison of the findings from the control and experimental groups showed that the testicular features of the vasectomized group were similar to those seen in the control group in contrast to the experimental cryptorchidism group which had irreversible testicular alterations. Results of the present study indicated the convenience of the usage of vasectomy in male contraception.

Comparison of the effects of vasectomy and experimental cryptorchidism in rats: II. Epididymal histology.

Vasectomy is a male contraception method that its side effects and reversibility rate are still controversial. Effects of vasectomy on epididymal morphology and function, which plays a crucial role in sperm maturation and storage, have not yet fully described. Therefore the present study was planned to find out the effects of vasectomy on epididymal structure, and comparisons of these effects were also made with the effects of a persistent infertility model, experimental cryptorchidism. To do this, 15 male Swiss albino rats were divided into 3 groups. While 5 animals were sham-operated, the rest were undergone either vasectomy (5 rats) or experimental cryptorchidism (5 rats) procedures. Epididymal tissue samples were collected prepared for light microscopy. Tissue sections were stained using appropriate staining techniques and evaluated under the light microscope. While there were distinct alterations in the epididymal morphology following experimental cryptorchidism, vasectomized rat epididymal features seemed similar to those seen in the sham-operated group. In conclusion, it could be suggested that the vasectomy might be a convenient method for male contraception.

Does vancomycine increase aminoglycoside nephrotoxicity?

Vancomycine and amikacin combined is an effective antibiotic combination used in the treatment of serious Gram-positive bacteremia in childhood. However, both drugs may have a potential nephrotoxic effect when used individually. The present study investigates whether the nephrotoxicity of these drugs that display a nephrotoxic effect when used separately increases when used in a combined manner. Ten 2-month-old rats were subjected to intraperitoneal injections of vancomycine (100 mg/kg) and amikacin (80 mg/kg) and ten other 2-month-old rats were administered amikacin (80 mg/kg) for a period of 15 days. The control group of animals consisting of five 2-month old rats were untreated. The renal tissues obtained by laparotomy were processed for both light and electron microscopy. Paraffin sections and ultrathin sections were evaluated in order to determine the renal structure of the control and the experimental groups. In addition, serum creatinine and blood, urea and nitrogen (BUN) levels were measured in blood samples obtained from the rats. In the amikacin and combined amikacin-vancomycine treatment groups, renal morphological changes were observed at the glomerular and tubular levels when compared to the control group, and the serum creatinine and BUN levels were also higher in these groups. Although both experimental groups were different from the control group, they had similar morphological and biochemical features. It was concluded that vancomycine did not influence the nephrotoxic effect of amikacin and both drugs could be used in a combined manner.
