[Fusion of protoplasts in Streptomyces antibioticus].

The analysis of Streptomyces antibioticus recombinants isolated under non-selective conditions for stability of inheritance of parental properties demonstrated the existence of non-stable diploids and stable haploid recombinants. Also, genetic analysis of heteroclones isolated on selective medium after plating spores of regenerants was conducted. The results of analysis of haploid recombinant and heteroclones pointed to linkage of genetic loci and the possibility to use the technique of protoplast fusion for establishment of genetic map of S. antibioticus.

[The determination of the site specificity of type-II restriction endonuclease in Streptomyces sp. 48].

The activity of restriction endonuclease which splits DNA of lambda and T7 phages into 8 fragments has been detected in the mycelium lysates of the 2-days culture of Streptomyces sp. 48. The fragments dimensions according to the data of agarose gel electrophoresis, coincide with those of AsuII--fragments of DNA of lambda and T7 phages. This allows the studied Ssp48 restrictase to be considered the isoschizomer of AsuII restriction endonuclease.

[The isolation and characteristics of mutant Streptomyces globisporus 1912 defective for landomycin E biosynthesis].

Mutants defective in the synthesis of antitumor angucycline antibiotic landomycin E were obtained from the asporogenic and initial sporulating strains of Streptomyces globisporus 1912 by means of nitrosoguanidine treatment and UV-irradiation of the mycelium fragments and protoplasts. The frequency of induction of LanE mutants in the protoplasts of strain 3-1 was 3 times higher (0.55%) as compared to mycelium fragments (0.17%). After mutagenic treatment of spores the yield of such mutants was lower (0.044-0.071%). It was shown, that izoflavon daidzein was an effective and necessary regulator of the landomycin E synthesis. The collection of 53 landomycin defective mutants was divided into three groups on the basis of their ability to produce antibiotic and regulator: 1) mutants LanE-Dai+ (45 strains), which did not synthesize landomycin E, but produced daidzein; 2) mutants LanE+Dai- (4 strains), which could restore landomycin E synthesis after adding exogenic daidzein to the medium and 3) mutants LanE- Dai- which did not synthesize landomycin E in the presence of daidzein.

[Restriction analysis of plasmid of the strain Streptomyces sp.27].

Plasmid pSS27 was found in streptomycete strain isolated from the soil sample in the Kyiv region. The paper deals with the data of restriction analysis of streptomycete plasmid pSS27 and its molecular weight. Plasmid pSS27 has unique restriction sites for endonucleases BglII and EcoRI, there are also the restriction sites for SalGI (3 sites), PstI (4 sites), BglI (5 sites), Bsp120I (7 sites) and SacII (11 sites). The molecular weight of plasmid DNA pSS27, being equal to 13.7 +/- 0.2 kb has been determined as a result of its restriction analysis.

[Restriction analysis of streptomyces plasmid PSS14].

The plasmid pSS14 having a unique restriction site for endonuclease BamH1 and several identification sites for different restrictases: 5 sites of BglI, 2 sites of BglII, 8 sites of Bsp120I and 3 sites of SacII has been found in the strain Streptomyces sp. 14. Basing on the results of plasmid DNA of pSS14, its molecule weight 13.1 +/- 0.3 tcn has been established.

[Spontaneous and induced variability of Streptomyces aureofaciens--chlortetracycline producer].

Spontaneous and induced variability of Streptomyces aureofaciens strain T[symbol: see text] 633 [symbol: see text] and its variants has been investigated. Correlative relation between the antibiotic formation and colonies morphology was observed. Combined effect of nitrosoguanidine and UV-beams resulted in the obtaining of 27 variants which exceeded activity of strain T[symbol: see text] 633 [symbol: see text] by 30-35% and were stable as to antibiotic biosynthesis during 7-10 generations.

[The stability of plasmid pSG 1912 inheritance by the cells of Streptomyces globisporus 1912 and of heterologous streptomycete strains].

Streptomyces globisporus 1912 strain contains plasmid pSG1912 (11, 2kb) determining some phenotypic properties. The plasmid is able to exist and to provide for exhibition of these properties both in autonomous and integrated state. It is established that plasmid pSG1912 and constructed derivatives are stably inherited by cells of S. globisporus 1912 and heterologous hosts, and besides, molecular size and determined properties being retained.

[The cloning of fragments of the streptomycete plasmid pSG1912 as a part of the vector pUC19].

DNA fragments of plasmid pSG1912 isolated from Streptomyces globisporus 1912 have been cloned into vector pUC19 in the E. coli cells. Stability of inheritance of hybrid derivatives in enterobacteria has been studied. The clones where recombinant plasmids are stably inherited have been chosen. The detailed restriction maps of cloned DNA fragments of pSG1912 have been constructed.
